Abstract

Turmeric (Curcuma longa) as a medicinal plant has widely consumed by pregnant women. The absence of dose standardization and the regulation of turmeric consumption in pregnancy raise concerns, especially if taken in early pregnancy. Angiogenesis is the process of new blood vessel formation from previously existing blood vessels, and plays an important role in embryogenesis and placentation during pregnancy. Angiogenesis is regulated by angiogenic molecules, such as VEGF and VE-cadherin. Turmeric has antiangiogenic effects in which the crude extract is pharmacologically more potent compared to the pure curcumin form. This study aimed to determine whether a turmeric decoctum affects the expression of VEGF and VE-cadherin in chick embryos. Turmeric was extracted by the decoction and freeze dried methods to obtain turmeric decoctum powder. This was tested on embryonated chicken eggs, which were divided into four groups; control group (2% DMSO) and treatment groups receiving various doses of the turmeric decoctum (200 ppm, 300 ppm and 400 ppm). The eggs used were less than 7 days after oviposition and incubated for 16 hours prior to injection of the treatment solution in ovo to the center of the yolk, followed by reincubation for up to 48 hours. Intraembryonic VEGF and VE-cadherin expression were assessed by whole-mount immunohistochemistry and quantified using image analysis techniques. This study showed administration of turmeric decoctum up to 400 ppm had no effect on the expression of VEGF and VE-cadherin in chick embryos.

Abstract

Tanah marginal masih menjadi masalah terhadap pertumbuhan tanaman tebu. Tanah yang kering dengan kandungan air yang terbatas mengakibatkan kebutuhan air tanaman tebu tidak terpenuhi. Cara untuk mengatasi masalah tersebut ialah dengan meningkatkan kadar lengas tanah melalui penambahan polimer superabsorben. Percobaan ini bertujuan untuk mendapatkan dosis polimer superabsorben dan kadar lengas optimum  pada budidaya tanaman tebu. Bahan yang digunakan adalah tebu varietas PS 862, polimer superabsorben, dan media tanam berupa tanah 3 : 1 pasir. Percobaan ini menggunakan Rancangan Petak Terbagi yang terdiri dari dua faktor dan diulang sebanyak tiga kali. Faktor pertama adalah kadar lengas tanah (K) yang terdiri dari empat taraf yaitu 100% kapasitas lapang (K1), 75% kapasitas lapang (K2), 50% kapasitas lapang (K3) dan 25% kapasitas lapang (K4). Faktor kedua ialah dosis polimer superabsorben (P) yang terdiri dari lima taraf yaitu tanpa polimer superabsorben (P0), 0,5 g polibag-1 (P1), 1 g polibag-1 (P2), 1,5 g polibag-1 (P3) dan 2 g polibag-1 (P4). Analisis data yang digunakan adalah uji F. Percobaan ini dilaksanakan di Pusat Penelitian Perkebunan Gula Indonesia (P3GI) Pasuruan pada bulan Januari 2015 hingga April 2015. Hasil percobaan menunjukkan bahwa tidak tedapat interaksi pada persentase perkecambahan, panjang tanaman, jumlah daun, jumlah anakan, tinggi batang dan jumlah batang. Faktor tunggal kadar lengas tanah menunjukkan beda nyata  parameter panjang tanaman, jumlah daun, jumlah anakan, tinggi batang dan jumlah batang, sedangkan faktor tunggal polimer superabsorben berbeda nyata pada parameter panjang tanaman dan jumlah batang.

Abstract

Abstract.Currently ethanol produced from molasses, cassava and maize, thereby competing with the need for food, animals feed and other industrial raw materials, so that the supply of raw materials is not continuity. If this continues, it will certainly have an impact on food prices spike, it will create new problems on the economy of the community, and one of an alternative raw material for bioethanol production is cellulose biomass.To convert biomass into bioethanol through several stages, including pretreatment, hydrolysis and fermentation. The content of lignin in biomass will disturb the enzymatic hydrolysis process, so it is necessary to encourage the delignification in the pretreatment stage. Microwave - alkali pretreatment can reduce more lignin and hemicellulose in rice straw, so it needs to be an effort to change the composition of lignocellulose and improve the hydrolysis of corn cob is fermented to become ethanol. Pretreatment of corn cobs research was done using randomized block design arranged in factorial and repeated 2 times. The first factor is the concentration of NaOH for 0,5 N, 1 N and 1,5N. The second factor is the time of exposure to microwave heat radiation for 10, 20 and 30 minutes. The results showed that cellulose content increases with prolonged exposure concentrations of NaOH and microwave, as well as to the content of hemicellulose and lignin decreased along with the increase of exposure duration of Microwave and the increase of NaOH concentrations.

Abstract

An incision wound is the destruction of several cells in the skin area that has experienced an incision. Several factors influencing wound healing are neutrophils as causative agents of inflammation, angiogenesis, and wound closure. These three factors should undergo physiologically to obtain the optimal healing process. Currently, herbal products are preferred because they have lower side effects than synthetic ones. Papaya leaves contain flavonoids with various functions, including wound closure and anti-inflammatory and antioxidant effects. Topical intervention, especially gel, is good for healing because it can keep the wound moist. This research aimed to determine the effect of the administration of papaya leaf gel on wound closure, neutrophil, and angiogenesis. Thirty white rats were used as the model animals in a randomized experimental study divided into five groups comprising negative control (0,9% NaCl), positive control (10% Povidone-iodine), and three intervention groups treated with gel of Carica papaya leaves at a dosage 10%, 20%, and 30%. All groups were topically administered treatment once a day on the dorsal surface for seven days. Carica papaya leaves contain flavonoids, saponin, tannin, and alkaloids qualitatively and 7,734 mg/kg of the flavonoid quantitatively. There were no significant differences in wound size in all three groups on three days, but Carica papaya leave gel had a superior effect compared to 10% PI and 0,9% NaCl in the wound closure process. There were significant differences in all groups in neutrophil and angiogenesis. Nevertheless, 20% papaya leave gel showed the lowest number of neutrophils and had a superior effect on improving the new blood vessels significantly compared to negative and positive groups.

Abstract

Sipermetrin adalah pestisida golongan insektisida, yang dapat mempengaruhi organ reproduksi hingga menyebabkan infertil. Tujuan penelitian adalah mengetahui pengaruh teh hijau terhadap jumlah sel epitel tuba fallopi dan endometrium tikus yang dipapar sipermetrin.  Ekstrak teh hijau merupakan hasil maserasi larutan etanol 96%. Penelitian ini dilakukan secara eksperimental menggunakan 25 ekor tikus betina dewasa. Hasil penelitian menunjukkan kelompok kontrol positif memiliki jumlah sel epitel yang paling sedikit, sedangkan kontrol negatif memiliki jumlah sel epitel yang paling banyak. Kelompok perlakuan menunjukkan bahwa P3 dengan dosis teh hijau paling tinggi menunjukan jumlah sel epitel yang paling banyak namun tidak berbeda signifikan dengan kontrol negatif.

Abstract

Acute myeloid leukemia (AML) is a malignancy that is often found in children. Many studies into the failure of apoptosis function, or programmed cell death, is one of the most important regulatory mechanisms of cellular hemostasis which is closely linked to the development of cancer, are important. Also, regulation of the apoptotic (p53) and anti-apoptotic (surviving) proteins influence treatment outcome. One role of p53 is to monitor cellular stress necessary to induce apoptosis. Surviving (BIRC5) is a group of proteins in the apoptosis inhibitor which works by inhibiting caspase-3. The role of surviving is considered very important in oncogenesis proliferation and cell growth regulation. Chemotherapy in childhood AML can inhibit cell growth and induce slowing as well as stopping the cell cycle. Thus, the aim of this study was to compare p53 and surviving before and after receiving induction chemotherapy in children with AML and also to determine the p53/surviving ratio. Peripheral blood mononuclear cells were collected from AML children before treatment and three months after starting their induction therapy. p53 and surviving were measured by flowcytometry using monoclonal antibodies. Data were analyzed by t-test for comparison between groups and Spearman’s test to find out the correlation between variables with a significant value of p < 0.05. A total of 8 children were evaluated. The intensity of p53 expression was not significantly increased after induction phase chemotherapy (p = 0.224), but surviving expression and the ratio of p53/surviving were significantly increased in the treatment group compared with the levels prior to chemotherapy (p = 0.002, p = 0.034), and there was a strong negative correlation between p53 and surviving after chemotherapy (r = −0.63, p = 0.049).

Abstract

Streptococcal pharyngitis sequel such as Rheumatic Fever (RF) or Rheumatic Heart Disease (RHD) is an autoimmune response mediated by T cells and IgG. Since it is an autoimmune process, the result of bacterial culture as the gold standard of diagnosis often shows negative results. IgG against the 33 kDa OMP is considered as an important mediator in the process of these autoimmune diseases, so its presence in blood serum can be used as a diagnostic tool. The purpose of this study is to prove that 33 kDa OMP is one of the immunogenic parts of the Streptococcus Group A β-hemolytic, so it is expected that IgG anti-33 kDa OMP can recognize and respond the bacteria and to support the probability of the Streptococcus Group A β-hemolytic infection. This study was a laboratory experimental study with a control group design. Animal used was RattusNovergicus immunized with whole cell bacteria or 33 kDa OMP mixed with Complete Freund’s Adjuvant or Incomplete Freund’s Adjuvant. Polyclonal IgG was obtained by drawing blood serum from the animals after immunization with Streptococcus Group A β-hemolytic for 4 weeks (A; n = 5) and 8 weeks (B; n = 5) or immunization with OMP 33 kDa for 4 weeks (C; n = 5) and 8 weeks (D; n = 5) and also negative control group (E; n = 5). Immunological tests were done using Dot Blot assay, ELISA, and immunocytochemical examination. The data obtained was then evaluated with statistical tests Kruskal-Wallis, Mann-Whitney and Repeated ANOVA (p < 0.05). The result showed that there was a difference in humoral immune response (IgG) between the groups albeit the difference was not significant (p > 0.05). Dot Blot and immunocytochemical tests indicated that IgG anti-33 kDa OMP were able to recognize and respond the Streptococcus Group A β-hemolytic antigen. This study concluded that 33 kDa OMP was the immunogenic part of the bacteria and that IgG anti-33 kDa OMP could recognize and respond the Streptococcus Group A β-hemolytic bacteria.

Abstract

Turmeric (Curcuma longa) is widely used as herbal medicine, not an exception by pregnant women. Turmeric consumption by expectant mothers requires standard dose, because of its antiangiogenic effect could be harmful on placentation process and embryonic development. This experiment was undertaken to determine the effect of different concentrations of turmeric decoctum to the expression of Vascular Endothelial Growth Factor Receptor-2 (VEGFR-2) and Angiopoietin 1 (Ang-1) on the 48-hours-old chicken embryo. In this study, turmeric was extracted using decoction method to mimic the common method as adopted by people. The turmeric decoctum were freeze dried into a powder form and was used in preparing the stock solution for 200 ppm (P1), 300 ppm (P2), and 400 ppm (P3) as experimental treatments. The control group (P0) received 2% DMSO without turmeric decoctum. These were administered on the yolk sack of 16 hours incubation of fertile chicken egg by number of 200 µL. After 48 hours incubation, the expression of VEGFR-2 and Ang-1 on the chicken embryo were counted by ImageJ software. The results revealed that there is no significant effect of turmeric decoctum to the expression of VEGFR-2 and Ang-1. This suggested that turmeric decoctum was safe up to 400 ppm on chicken embryo.

Abstract

Pellucide zone 3 (ZP3) involves in fertilization mechanism. Moreover, antibody of ZP3 can develop to inhibit egg and sperm interaction. This study aims to determine the effect of hZP3 (mab-hZP3) monoclonal antibody on the expression of Bcl-2 and Bax in follicle granulosa cells of the mice ovary. Female mice BALB/c were divided into 12 groups which consisted of control and experimental treatment group. Each group was added with 30% of total mice as error sample (1 mice). Each groups were treated differently: 50 µl adjuvant Al(OH)3 in 50 µl Tris HCl, 20 µg Mab-hZP3, 40 µg Mab- hZP3, and 60 µg Mab-hZP3. Each group was dissected at day 10, 15 and 20. Measurement of Bcl-2 and Bax was performed with immunohistochemistry. Data was analyzed by Two-Way ANOVA. There was no significant effect of Mab-hZP3 administration in various doses on Bcl-2 (p=0.0825), and Bax (p=0.836). There was no significant effect of administration of Mab-hZP3 in time (p=0.807), neither on Bcl-2 expression (p=0.088) and Bax (p=0.227). The lowest Bcl-2 level was found in dose of 60 µg in day 15. There was no significant effect of Mab-hZP3 in various doses and time (p=0.691), neither to Bcl-2 and Bax. Such results obtained due to the specificity of a monoclonal antibody that recognizes specific antigen. Mab-hZP3 is proposed as immunocontraception for women causing no disturbance of folliculogenesis.

Abstract

Streptomyces hygroscopicus (S.hygroscopicus) is a Gram-positive soil bacterium that can produce secondary metabolites from fermentation that have a therapeutic effect. The fermented S. hygrocospicus metabolites that are still in the form of crude extracts are difficult to develop as drug preparations because the active compounds are not yet known, so it will be challenging to determine the dosage of drugs that have a therapeutic effect. Therefore, it is necessary to carry out exploratory research to narrow down the secondary metabolite profile from the fermentation of S. hygroscopicus, using extraction and fractionation methods, which are then identified by Thin-Layer Chromatography (TLC) using a combination of solvents. This study used the extraction method with a separating funnel. The fractionation was carried out using the BUCHI (Sepacore®) Flash Chromatography and Reveleris® PREP Purification System column chromatography gradually using ethyl acetate and n-hexana.  47 and 60 of the fractionation results were taken as samples, that further were profiled using TLC and given the appearance of 10% KOH stains and p-Anisaldehyde - sulfuric acid, so that various classes of compounds with different Rf values were obtained, namely Monoterpenes, Triterpenes, Steroids, Saponins, Coumarin, Scopoletin, and Alkaloids.
