Abstract

ABSTRACTWhite spot syndrome virus (WSSV) is the most dangerous shrimp pathogen and give a very bad impact to the shrimp industry in many countries, including Indonesia. Furthermore, the vaccine usage is less effective since the local WSSV suspected to have some mutation and have differences in sequence from the one that used for recombinant vaccine production so far. Therefore, it is necessary to produce recombinant vaccine for immunization of local shrimp according to WSSV isolates from Indonesia. Viral protein 19 (VP19) is an important WSSV envelope proteins because of its involvement in the systemic infection of shrimp, so it can be developed as a recombinant vaccine. The objectives of this study are to obtain VP19-encoding gene of WSSV from Situbondo, Indonesia. This research conducted through several stages, DNA genome isolation of Penaeus monodon infected with WSSV, amplification of VP19-encoding gene, isolation of VP19-encoding gene, ligation of VP19-encoding gene into pGEM-T Easy vector, transformation to Escherichia coli, recombinant vector screening, vector isolation using mini preparation technique, vector verification, and sequencing. The result of cloning shows that the size of VP19-encoding gene from Situbondo isolates is similar to VP19-encoding gene from the abroad one, which is 366 bp. However, the sequence result shows that there are some differences on bases and amino acid. However, the result of sequencing shows there are difference of five base and three amino acid arranged VP19 sequence the Situbondo isolate with isolate from several countries. The similiarity of the Situbondo isolate sequence with the other isolate (from Indonesian and other countries) was only 98%. This difference indicates the importance of making a vaccine from a local WSSV genes especially Situbondo isolate.

Abstract

Indonesia boasts a rich and diverse traditional culture, shaped by its historical context, geographic conditions, religious influences, values, and the varied experiences of its people. The distinctiveness and variety of these traditional cultures offer valuable insights and serve as resources for learning mathematics and economics. Common perceptions often dissociate mathematics from everyday activities such as farming, trading, and other practical tasks. This research aims to investigate the mathematical principles embedded in the Bedeyé activities within the traditional economic and trade practices of the Sasak tribe. Employing a qualitative research approach with ethnographic methods, data were gathered through observation, literature review, and interviews with cultural practitioners, traditional traders, community leaders, and cultural experts. The research findings reveal that Bedeyé activities exemplify traditional barter-based trading practices, where goods are exchanged for other goods. The mathematical concepts identified in the Bedeyé tradition include comparative mathematics, set theory, and volume estimation. Moreover, the educational values inherent in Bedeyé activities encompass itiq, saling redaq, semaiq, and saling saduq. These mathematical principles observed in the traditional trading systems of the Sasak tribe, as demonstrated through Bedeyé activities, can be leveraged as educational tools and foundational elements for teaching mathematics.

Abstract

This article seeks to explain the emergence of an Indonesian post-Orientalist study of Islam from the 1990s onwards, which results from the increased influx of Indonesian-born scholars into the study of Indonesian Islam, a field previously dominated by Western-born scholars. In contrast to Edward Said’s adverse Orientalists, to the Arabic-based dirāsah islāmiyyah, and the previous generations of Indonesian Western-educated scholars, the post-1990s generation of pesantren-and-Western-educated Indonesian scholarship has taken selectively elements from Islamic texts and traditions, humanities, and social sciences in analyzing contemporary Islamic beliefs and practices. With an eclectic intellectualism combining faith and public mission, Indonesian Muslim scholars have reinterpreted Qur’anic and classical Islamic concepts while engaging different Western theories regarding religion, law, identity, and social movements. By analyzing local and national figures and movements, using diverse sources, and negotiating the tensions between the normative, practical, scriptural, and contextual, they aim to represent Islam and Muslims in their diversity and complexity in global, national, and local dynamics. With collaborative work at home and abroad, they contribute to pursuing different trajectories with scholarship and activism for Indonesian society and beyond.

Abstract

This study aims to evaluate the effect of fermentation with lactic acid bacteria Pediococcus pentosaceus on phosphorus content in rice bran. This bacteria is a probiotic isolated from animal intestinal microflora, with high colonization ability due to its suitability to the original environment. The study focused on the ability of the isolate to break down phytic acid bound to the bran, with the main indicator being an increase in phosphorus levels. The fermentation results showed the highest viability at an incubation temperature of 37°C, with a maximum number of 240 × 10⁸ CFU/g at 12 hours, but decreased thereafter. Although there was an increase in phosphorus levels, the results were not statistically significant (p>0.05) in both dry bran (P1) and wet bran (P2) treatments. The highest increase in phosphorus occurred at a temperature of 44°C (0.91±0.004%; +25%) and at 37°C (0.87±0.002%; +19%). This suggests that temperature affects the effectiveness of fermentation, but water content and incubation time may be limiting factors. Overall, fermentation of rice bran with P. pentosaceus did not show a significant increase in phosphorus levels.
