Abstract

Karakterisasi Biokimia Enzim Fitase Rekombinan (phyK) dari Klebsiella sp. ASR1 Yang Dienkapsulasi Dengan AlginatEnzim fitase melepas molekul fosfor pada atom C dari benzena Inositol fitat. Tetapi fitase memiliki kelemahan tidak mampu bertahan terhadap kondisi ekstrim dalam lambung nonruminansia. Solusi dalam penelitian ini yaitu fitase dienkapsulasi menggunakan alginat. Penelitian ini bertujuan mengkarakterisasi fitase setelah dienkapsulasi menggunakan alginate. Hasil penelitian ini yaitu fitase yang dienkapsulasi memiliki aktivitas tertinggi pada pH 6,0, sedangkan fitase tanpa enkapsulasi pada pH 5,0. Suhu optimum untuk aktivitas tertinggi fitase yang dienkapsulasi yaitu 70ºC, sedangkan fitase tanpa enkapsulasi 37ºC. Untuk perlakuan penambahan ion logam, aktivitas tertinggi fitase yang dienkapsulasi terjadi dengan penambahan 0,1 mM Fe2+ dan 1,0 mM Ca2+, sedangkan fitase tanpa enkapsulasi dengan penambahan 0,1 mM Fe2+. Berdasarkan hasil penelitian ini, fitase yang dienkapsulasi memiliki keunggulan lebih banyak dibandingkan dengan fitase tanpa enkapsulasi, karena mampu bertahan pada pH dan suhu tinggi, dan beberapa efek ion logam.Kata Kunci: alginat, asam fitat, enkapsulasi, fitase, fitase rekombinanABSTRACTPhytase enzymes release phosphorus molecules on the C atom from benzene inositol phytate. But phytase has the disadvantage of being unable to withstand extreme conditions in the non-ruminant stomach. The solution in this research was phytase encapsulated using alginate. This study aims to characterize phytase after being encapsulated using alginate. The results of this study were the encapsulated phytase had the highest activity at pH 6.0, while the unencapsulated phytase at pH 5.0. The optimum temperature for the highest activity of the encapsulated phytase was 70ºC, while the unencapsulated phytase 37ºC. For treatment of metal ion addition, the highest activity of the encapsulated phytase occurred with the addition of 0.1 mM Fe2+ and 1.0 mM Ca2+, while the unencapsulated phytase with the addition of 0.1 mM Fe2+. Based on the results of this study, the encapsulated phytase had more advantages compared to the unencapsulated phytase, as the former withstand high pH and temperature, and some metal ion effects.

Abstract

This research was aimed at producing a crude intracellular phytase characterized from recombinantbacteria. The recombinant bacteria (pEAS1AMP) was produced by way of transforming pET-22b(+)+pEAS1 into competent E. coli BL21 and E. coli BL21(DE3) cells. Crude intracellular phytaseproduction was induced using 1,5 mM Isopropyl-β-D-thiogalactopyranosid (IPTG). Recombinantbacteria product and enzyme activity test followed the Sajidan method. E. coli BL21(+)pEAS1 and E.coli BL21 (DE3)(+)pEAS1 recombinant bacteria showed growth after 20 hours and 10 hours oftransformation. Phytase activity of E. coli BL21 (DE3)(+)+pEAS1 showed higher than those of E. coliBL21(+)+pEAS1. Crude intracellular phytase of pEAS1AMP recombinant bacteria has an optimumactivity at pH 5, 40oC, incubation period of 60 minutes, substrate concentration of 2%, molecular weight(MW) of 47.3 kDa, Km = 15.91 υM and Vm = 2.41 υM/second. Mg2+ acts as a cofactor but Fe3+ (10-4M) acts as an inhibitor.

Abstract

Tujuan dari pengabdian adalah untuk melatih masyarakat non produktif untuk mengoptimalkan potensi lokal daerah melalui kegiatan socioentreprenuership. Pengabdian ini memanfaatkan potensi lokal yakni ikan lele untuk ditingkatkan nilai gunanya dan nilai jualnya. Selain untuk melatih pemanfaatan ikan lele, pengabdian ini juga bertujuan untuk meningkatkan kemampuan peserta dalam memasarkan produk dengan cara atau merode digital. Metode yang digunakan dalam layanan ini dimulai dengan Workshop, Praktek, dan Implementasi. Dalam melakukan pelatihan ini, para peserta diminta untuk mengerjakan tes terkait dengan pengetahuan dan cara meningkatkan nilai jual ikan lele serta cara memasarkannya. Analisis data yang digunakan adalah pre-test dan post-test. Hasil dari kegiatan ini adalah bahwa ada peningkatan dalam pengetahuan dan cara meningkatkan nilai jual ikan lele serta cara memasarkannya.

Abstract

The purpose of the research was to improve student’s biology learning activity in class X-2 SMA Muhammadiyah 1 Karanganyar in the academic year 2013/2014 through by implementation of Guided Discovery learning model. The research was a classroom action research which performed in two cycles. Each cycle consisted of 4 phases, that was planning, action, observation, and reflection. Subject of the research was students of class X-2 SMA Muhammadiyah 1 Karanganyar in the academic year 2013/2014. Data of the research was obtained by test and non test technique consisted observation, interview and documentation. The obtained data were analyzed using descriptive technique and were validated using triangulation of methods. Target of the research was to achieve 75,00% for each aspect of student’s biology learning activity. The results of the research showed the percentage student’s biology learning activity aspect for oral activities pre cycle (31,94%), cycle I (69,50%), and cycle II (75,00%), motor activities pre cycle (25,46%), cycle I (77,00%), and cycle II (79,33%), mental activities pre cycle (31,02%), cycle I (72,26%), and cycle II (79,81%), emotional activities pre cycle (30,56%), cycle I (81,00%), and cycle II (87,38%), writing activities pre cycle (46,30%), cycle I (77,00%), and cycle II (80,77%), listening activities pre cycle (36,11%), cycle I (76,00%), and cycle II (82,41%), drawing activities pre cycle (27,78%), cycle I (63,00%), and cycle II (78,70%), and visual activities pre cycle (30,56%), cycle I (72,00%), and cycle II (78,70%). The results showed that the average percentage student’s biology learning activity aspect such as pre cycle (32,47%), cycle I (73,47%), and cycle II (80,26 %). The conclusion of the research is the implementation of Guided Discovery learning model can improve student’s biology learning activity in class X-2 SMA Muhammadiyah 1 Karanganyar in the academic year 2013/2014. Keywords : Guided Discovery, Student’s Biology Learning Activity

Abstract

The aim of this research is improve quality of biology learning for teacher performance, conditioning class, student attitude in class, and student motivation of learning in student grade of X-E SMA Negeri 1 Kartasura academic year 2011/2012. This research refers to The Classroom Action Research with subject were 39 students in grade of X-E SMA Negeri I Kartasura. The sources of data consist of information from teacher and student, places and events that happened during teaching and learning process, and also some documents. The data of research is collected through observation sheet, questionnaire and interview. The research uses triangulation as validation data. The research uses descriptive as analytic data. The result of this research shows that there has been improvement percentage of the quality of biology learning in every aspect which includes teacher performance, conditioning of class, student attitude, and student motivation of learning. The percentage’s mean of the questioner result in  sequence I is  conditioning of class 72,56%; student attitude 65,14%; student motivation of learning 75,02%, and of observasi data are teacher performance 83,33%; conditioning of class 71,54%; student attitude 63,37%; student motivation 75%.  The reflection of the first sequence shows that students having low of argument explanation in the learning goal, so the researcher gave further treatment in sequence II.  In the second sequence the percentage’s mean of the questioner result is teacher performance 93,33%; conditioning of class 82,69%; student attitude 86,08%; student motivation 86,22%, in this cycle every quality of biology  learning aspects improved in line with the target. It can be concluded from the research that Contextual Teaching Learning (CTL) approach application accompained by Subject Specific Pedagogy (SSP) can improve quality of biology learning of students in grade X-E SMA Negeri 1 Kartasura academic year 2011/2012. Key Words: Quality of biology learning, Contextual Teaching Learning, Subject Specific Pedagogy

Abstract

Penelitian ditujukan untuk memberikan pelatihan dan pengembangan usaha masyarakat non produktif melalui UMKM berbasis bahan dasar bekatul. Kegiatan pengabdian yaitu pemberian materi tentang kandungan bekatul, cara pengolahan bekatul sebagai pakan ternak berkualitas dan makanan bergizi, pengemasan dan pemasaran produk. Jumlah peserta yang diikutsertakan dalam program ini yakni sebanyak 35 masyarakat dengan teknik pengumpulan data penelitian dengan cara: observasi potensi, wawancara, analisis data sekunder, dan dokumentasi. Metode kegiatan pengabdian dilakukan dengan cara pemaparan materi oleh ahli, fact finding (analisis kebutuhan), discussion (diskusi antara pemateri), asking and answering (tanya jawab dan diskusi), dan recitation (resitasi atau pembagian tugas), practice and experiment (praktik dan eksperimen). Hasil kegiatan ini berupa temuan bahwa saat ini masyarakat memiliki pemahaman pengolahan bekatul terbatas pada produk olahan untuk pakan ternak. Kegiatan diskusi memberikan paradigma baru terhadap masyarakat untuk mengolah berbagai macam produk makanan berbahan bekatul. Pelatihan ini memberikan bekal kepada masyarakat untuk menganalisis produk berbahan bekatul untuk dikembangkan menjadi produk olahan makanan sehingga memiliki nilai jual yang lebih tinggi. Kegiatan pengabdian membekali masyarakat untuk memiliki pengetahuan dasar pengolahan makanan hingga ke tahap pemasaran sehingga diharapkan dapat meningkatkan perekonomian masyarakat.

Abstract

Tujuan penelitian ini adalah untuk membuat produk, mengetahui kelayakan, dan mengetahui efektivitasmultimedia interaktif berbasis inkuiri berwawasan potensi lokal menggunakan macromedia flash pada materitumbuhan lumut dan paku. Penelitian ini menggunakan model pengembangan R&D Borg and Gall yangmeliputi tahap: research and information collection, planning, develop preliminary form of product, expertjudgement, revisi produk, preliminary field testing, dan uji coba lapangan. Uji coba lapangan dilakukan dalamseting eksperimen yang melibatkan 3 kelas. Sampel penelitian ditentukan dengan teknik cluster randomsampling terdiri dari satu kelas. Kelas kontrol (X.3) belajar menggunakan model konvensional, kelas Media(X.4) belajar menggunakan media yang telah dikembangkan, dan kelas Agregasi (X.5) belajar denganmenggunakan model, modul, dan media yang telah dikembangkan. Teknik pengumpulan data menggunakanmetode tes untuk data prestasi belajar kognitif, angket dan lembar observasi untuk prestasi belajar afektif danprestasi belajar psikomotor. Hasil penialaian media oleh ahli media, ahli materi, dan praktisi adalah 97.83,87.96, dan 93.6. Penggunaan multimedia interaktif berpengaruh signifikan terhadap hasil belajar siswa, sehinggaefektif digunakan dalam pembelajaran.

Abstract

The purpose of this research is to improve quantity and quality of student’s question through problem based learning in biology at grade X IIS 4 of SMA Negeri 5 Surakarta. The research was 4 cycles action research that conducted in academic year 2014/2015. Data were collected through observation, interview and video recording, and validated using triangulation method. Quantity of question collected by counting the number of questions during the learning process. Quality of question categorized based on revised Bloom’s Taxonomy. The result of this research shows: 1) PBL effectively increased the quantity and quality of questions; 2) the quality of student’s question in each cycle at factual, conceptual, procedural dimension. Metacognitiv dimension was identified in the last cycles; 3) the increasing of quantity student’s question from first cycles to the fourth cycles is 156 question at first cycles, 130 question at second cycles, 136 question at fthird cycles and 226 question at fourth cycles.Keywords: quantity and quality of student’s question, problem based learning

Abstract

The purpose of this research is to increase higher order of student’s question through problem based learning at conceptual dimension in biology learning. Population of this research was students at grade X IIS 4 of SMA Negeri 5 Surakarta. The research was four-cycles action research that conducted in academic year 2014/2015. Data were collected through observation, interview and video recording. Student’s question collected by counting the number of questions during the learning process and categorized based on revised Bloom’s Taxonomy. Higher order question include C4, C5 and C6 question. The result of this research shows: 1) C4 and C6 of conceptual question was identified in each cycles. C5 of conceptual question was identified in the first and  third cycles; 2) the number of C4 conceptual question is 0% at pra cyles, 2.52% at first cycles, 6.06% at second cycles, 3.53% at third cycles, 6.57% at fourth cycles. The number of C5 conceptual question is 0% at pra cycles and 0.5% at first and third cycles. The number of C6 conceptual question is 0% at first cycles, 17.68% at first cycles, 8.08% at second cycles, 19.19% at third cycles and 35.35% at fourth cycles; 3) The higherest number of HOT’s question that identified was C6 question. The conclusion of this research is PBL effectively increased the higher order of student’s questions at conceptual dimension. Keywords: higher order of student’s question, problem based learning

Abstract

Penelitian bertujuan mengetahui pengaruh pembelajaran problem based learning terhadap penguasan fakta, konsep, prosedur dan metakognisi pada pembelajaran problem based learning pada pelajaran biologi. Penelitian merupakan quasi eksperiment dengan populasi penelitian adalah peserta didik kelas 10 SMAN 4 Surakarta, dengan 2 kelas sebagai sampel penelitian kontrol dan perlakuan. Penentuan sampel menggunakan purposive random sampling untuk dua kelas yang mempunyai kesamaan kemampuan dengan uji F(32;0,05)=3,998. Analisis pertanyaan peserta didik sebelum dan sesudah menggunakan pembelajaran problem based learning menggunakan kualifikasi pertanyaan berdasarkan Taksonomi Bloom. Data dianalisis dengan anacova. Hasil penelitian menunjukan bahwa pembelajaran problem based learning mempengaruhi penguasaan pertanyaan ditinjou dari jumlah pertanyaan pada pengetahuan fakta, konsep, prosedur  namun tidak untuk metakognisi.Keywords: problem based learning, pertanyaaan, fakta, konsep, prosedur, metakognisi
