Abstract

Islam and Javanese culture cannot be separated, both of them have a role and influence in navigating the times, Islam has brought many changes, starting from the kingdoms that began to stand on the island of Java. And the Islamic religion in Java has a unique diversity because the spread of Islam in Java is more dominant, resulting in the formation of diverse cultural acculturation. This acculturation occurs in many ways, one of which is visible, namely a result or work of carving, wayang, and slametan art. In the relationship between Islam and Javanese culture, each has a role and influence in spreading the religion of Islam, from the beginning of its development to enriching new cultures, and of course there are also some cultures that do deviate from the teachings of Islam, but Islam and Javanese culture can walk side by side.

Abstract

Abstract

Abstract

This study aimed to assess the presence of single nucleotide polymorphisms (SNPs) in the exon 1 region of the tyrosinase (TYR) gene in Bali cattle with abnormal coat color. This study analyzed 43 Bali cattle, 26 individuals with typical or standard coat color, and 17 individuals with albinism. The genetic diversity of the TYR gene was determined via direct sequencing and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) techniques. The sequence data of the TYR gene were scrutinized using BioEdit and MEGA10 software to detect single nucleotide polymorphisms (SNPs) related to different types of coat color features in Bali cattle. The genetic diversity information was derived from computations performed using PopGen 1.32 software. The results showed that exon 1 of the TYR gene was affected by the new SNP g.939A>G, which is polymorphic in Bali cattle. In conclusion, the SNP c.939A>G can be further analyzed for use as a candidate-assisted selection (MAS) for abnormal coat color in Bali cattle.

Abstract

Bali Cattle are known as cattle with a unique coat color pattern. At Taro village was found different coat colors of Bali cattle, namely Taro cattle perform white coat color. The TYR gene has a vital role in producing coat color. This research was aimed to determine mutations that caused of the white color in Taro cattle. Two methods of PCR and PCR-RFLP were applied in this study. PCR method followed by sequencing used 29 DNA samples consisted of 17 Taro cattle, 12 DNA samples of 3 breeds cattle as comparison while RFLP method applied 56 DNA samples (17 Taro; 31 wild Bali; 4 Simental, 4 PO/Ongole descendant). All DNA were collected from fresh blood, and extracted by using DNA Extraction Kit. Amplification of TYR (exon 5) gene was conducted using forward and reverse primers to get polymerase chain reaction products with lengths of 320 bp and sequenced with Sanger method (services). PCR-RFLP was conducted with enzyme restriction of AciI. Determination of TYR gene diversity was analyzed through sequencing data. TYR gene sequencing was analyzed by using Bioedit and Mega11 software to identify single nucleotide polymorphisms (SNPs). SNP genotyping was analyzed from PCR-RFLP. The results showed that the TYR gene (exon 5) of Taro cattle was found with 320 bp length. SNP was obtained at one position c.1467 T>C. This SNP has never been reported yet on the ensemble website. This finding suggests that a specific genetic marker for albinism in Bali cattle needs to be more explored

Abstract

Cattle coat color is governed by numerous genes, notably the tyrosinase gene (TYR). This study analyzed coat color anomalies like albinism and white spotting in Bali cattle. It aims to discern the TYR gene's diversity, expression patterns, and correlation with coat color abnormalities. The research encompassed 189 cattle, including those with standard coat color (n=53), white-spotted (n=11), and albino (n=17) Bali cattle, as well as Simmental (n=37), Limousin (n=14), Madura (n=21), and Peran-akan Ongole (PO) cattle (n=36). Total DNA was extracted and the TYR gene in exon 1 was amplified using forward and reverse primers with a target amplicon length of 994 bp. Direct sequencing unveiled TYR gene diversity, analyzed using BioEdit and MEGA6 software to identify SNPs. PCR-RFLP was used for SNP genotyping, while qPCR analyzed TYR gene expression. Two mutations (SNP g. 939A>G and SNP g. 887C>T) were discovered in Bali cattle TYR exon 1. SNP g. 939A>G exhibited polymorphism, with the highest GG genotype frequency in standard Bali cattle, indicating a high G allele frequency. Conversely, Madura, Simmental, Limousin, and PO cattle had the lowest allele fre-quency. Chi-square (χ² test) results showed non-significance across all cattle types. TYR gene expres-sion differed significantly between standard Bali cattle and albinos (p<0.05).
