Abstract

Kanker kolorektal mencapai sepuluh persen dari total tipe tumor di seluruh dunia dan merupakan kanker dengan mortalitas tertinggi di seluruh dunia. Kejadian kanker kolon terus meningkat seiring dengan pertambahan penduduk baik di negara berkembang maupun negara maju. Kemoterapi belum memberikan hasil yang optimal dan sering menimbulkan efek samping yang serius karena pada umumnya tidak bekerja spesifik pada sel kanker tetapi juga pada sel normal. Banyak bahan alam yang berpotensial untuk dikembangkan sebagai agen kombinasi seperti propolis, jinten, tapak dara, mengkudu, dan daun dewa. Propolis merupakan suplemen nutrisi yang dihasilkan oleh lebah dan telah digunakan sebagai pengobatan tradisional di dunia. Propolis sudah banyak dijadikan bahan penelitian karena aktivitas antibakteri, antijamur, antivirus dan hepatoprotektifnya. Propolis dan senyawa lainnya telah digunakan untuk mengobati inflamasi, untuk meningkatkan kekebalan tubuh, dan agen anti kanker. Penelitian bertujuan untuk melihat pengaruh pemberian propolis dari daerah Kerjo, Karanganyar, Indonesia terhadap ekspresi caspase 3, proliferasi dan induksi apoptosis pada kultur sel kanker kolon (cell line WiDr).  Penelitian menggunakan desain eksperimental laboratorik dengan post test with control group design. Penelitian menggunakan kultur sel WiDr (sel kanker kolon) dengan pemberian ekstrak ethanol propolis (EEP). Pengamatan ekspresi caspase 3 dengan metode imunositokimia, sedangkan pengamatan proliferasi dengan metode doubling time dan induksi apoptosis dengan metode double staining. Hasil Penelitian didapatkan EEP dengan IC50 sebesar 140 µg/mL meningkatkan ekspresi caspase 3 sebesar 42,1% dibandingkan kelompok kontrol 6,89%. EEP meningkatkan ekspresi caspase 3 sebanding dengan peningkatan konsentrasi EEP. EEP dengan dosis konsentrasi 70, 140, 280 µg/mL mampu menekan proliferasi baik pada proliferasi jam ke-24, 48 , maupun 72. EEP dengan IC50 sebesar 140 µg/mL meningkatkan induksi apoptosis sebesar 53,16% dibandingkan kelompokkontrol 0,64%. Pemberian EEP meningkatkan induksi apoptosis sebanding dengan peningkatan konsentrasi EEP. Penelitian ini menunjukkan bahwa EEP mempunyai sifat antiproliferasi dan mampu menginduksi apoptosis pada sel WiDr.  Kata kunci: EEP, caspase 3, proliferasi, apoptosis, WiDr

Abstract

Kanker kolorektal merupakan kanker ketiga terbanyak dengan lebih dari 1 juta kasus setiap tahunnya dan salah satu kanker dengan mortalitas tertinggi di seluruh dunia. Radioterapi dan kemoterapi pada kanker relatif terbatas karena toksisitasnya yang tinggi dan efek samping yang bersifat merusak. Pengembangan propolis merupakan strategi baru untuk terapi adjuvan yang diharapkan meminimalkan efek samping terapi standar yang ada. Peran propolis pada keganasan terkait kemampuannya dalam menginduksi apoptosis dan aktivitas antiproliferasi. Penelitian in vitro, menunjukkan propolis memiliki aktivitas proapoptosis pada berbagai jenis sel kanker, meliputi : kanker laring, kanker paru, kanker pankreas, kanker tiroid, kanker payudara, kanker prostat dan glioma. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian propolis yang berasal dari Kerjo, Karanganyar, Indonesia terhadap aktivitas antiproliferasi, terkait peningkatan ekspresi p21 dan induksi apoptosis terkait peningkatan ekspresi protein bax pada kultur sel kanker kolon (cell line WiDr). Jenis penelitian ini, eksperimental laboratorik dengan post test with control group design. Penelitian menggunakan kultur sel WiDr (sel kanker kolon) dengan pemberian ekstrak etanol propolis (EEP). Pengamatan ekspresi p21 dan protein Bax dengan metode imunositokimia, sedangkan pengamatan induksi apoptosis dengan flowcytometry. Analisis statistik menggunakan uji Kruskall Wallis dilanjutkan Mann WhutneyU test. EEP cenderung menekan viabilitas sel WiDr dengan IC50 sebesar 140 μg/mL. EEP konsentrasi 70,140, 280 μg/mL mampu meningkatkan ekspresi p21 yang sebanding dengan peningkatan konsentrasi yang diberikan. EEP konsentrasi 70 μg/mL (1/2 IC50) paling efektif dalam menginduksi apoptosis dan meningkatkan ekspresi Bax pada sel WiDr. Peningkatan konsentrasi EEP mengakibatkan kematian sel WiDr ke arah nekrosis. Penelitian ini menunjukkan EEP mampu menekan viabilitas sel WiDr. Aktivitas ini kemungkinan terkait dengan kemampuannya dalam meningkatkan ekspresi p21 sebanding dengan peningkatan konsentrasi yang diberikan. EEP pada konsentrasi 70μg/mL mampu menginduksi apoptosis pada sel WiDr terkait dengan peningkatan ekspresi Bax. Peningkatan konsentrasi EEP konsentrasi 140 dan 280 μg/Ml mengakibatkan nekrosis sel WiDr. Kata kunci: EEP, p21, protein bax, cell line WiDr

Abstract

Objective: to prove the effect of Tai chi on the increase of dynamic balance in the elderly. Methods: One group pretest posttest design with 23 subjects of elderly people aged 60-75 years old and given Tai chi twice a week for 30 minutes within one month (8 times). Measurement: Functional Reach Test (FRT) and Time Up and Go Test (TUG). Analysis: Normality test by Saphiro Wilk as well as pretest-posttest different test with paired T-test.  Results: Based on the analysis of normality test by Saphiro Wilk on the pretest and posttest, the obtained result is p>0.05 which means the data is normally distributed. In the meantime, the analysis of pretest-posttest difference test with paired T-test results in dynamic balance value before and after Tai chi as much as p<0.05 (p=0.000); and, it also generates the dynamic balance value before and after Tai chi with TUG as much as p<0.05 (p=0.000). This means there is a significant difference between before and after the treatment when measured by Functional Reach Test (FRT) and Time Up and Go Test (TUG).  Conclusion: Tai chi improves the dynamic balance in the elderly.

Abstract

Teacher quality is the most important factor in enhancing student achievement. In order to achieve the goal, lesson study comprises three main phases of plan, do, and see encourages teachers to do collaborative learning in which they create learning communities to discover, create, and negotiate new meanings to improve their teaching. Teachers at all stages of their careers could learn from one another. They could work together, discuss common teaching issues, share successful strategies, and develop new approaches to share problems. Organizing such networks of teacher in collaborative learning promotes an effective way to share pedagogical and content knowledge. In order to think productively about ways to enhance student learning, teachers need to consider both the material they are teaching and the ways that they are presenting it. Considering the importance of both of them, this study aims to know the impact of collaborative learning to teacher performance particularly in SD/MI Al Hikmah Semarang. By involving three model teachers conducting teaching and learning process, and six observers (teachers and lecturers) observing the teacher performance and student activities in the classroom conducted in two cycles, this study shows that collaboration among teachers increased their performance of teaching in which teachers emphasized using active engagement, appropriate learning designs, and application of theories on learning. The majority of teachers agreed that collaborative learning through lesson study helped their performance of teaching. Teachers who participated in collaborative learning had better achievement in their teaching performance rather than those who did not participate in collaborative learning.

Abstract

Language is a means of communication in Literature. People can understand and analyze any story by using language. They can also find out the content of story from different country. Readers can also compare story from one country to another country. They can also translate story from different country with their own mother tongue. One example is The Red Badge of Courage. We can get many lessons and ideas from this American novel. It is a war novel, which focus on American Civil War. Henry Fleming is one of main characters in the novel. Regarding to the history of Civil War, we still have many questions from the content of it. How about the main character‘s dreaming in civil war? Can his bravery influence to his friends? What is the moral value from this war novel? People can get many imaginations and lessons from the main character, Henry Fleming. Descriptive qualitative is a method in discussing this novel. Reading, understanding, and analyzing are the necessity things to do. The idea and opinions from novel and reference books are sources in getting idea. We can take this novel is as a zero to hero novel regarding to the main character‘s activities. All soldiers in his troop surprise when they see a brave-heart person, Henry Flaming. Furthermore, Henry shows that he can beat his cowardice to be a brave man. His wound is a proof of being a hero in his troop. His questions of being a brave soldier show up after he beats all trials in civil war. Self-confidence in Henry Flaming is a way to show that a soldier must have bravery in leading a troop.

Abstract

Coffee drinking is a habit that still doubtful for it’s benefits. Many active ingredients in robusta coffee (Coffea canephora) are very useful for health, especially the antioxidant content that can ward off free radicals that absorbed in the body. Free radicals can be caused by psychological stress exposure. Excessive amounts of radicals in the body cause oxidative stress. This study aimed to see the robusta coffee effect on decreasing levels of Malondialdehyde (MDA) with “randomized post test only control group” design with 25 samples of white male rat strain wistar (Rattus norvegicus). Samples were divided into 5 groups: control groups and randomized treatment groups. Treatment given in psychological stress and giving robusta coffee drink with single dose. This study used “one way anova” test and followed by “pos hoc tukey HSD” test for statistics. Robusta coffee had an effect in MDA levels changing. There was a MDA levels decreasing in 1 hour after robusta coffee drinking in experimental rats with oxidative stress conditions that were exposed by psychological stress.&nbsp;&nbsp;&nbsp;

Abstract

Exercise such as Diabetes dance of Persadia 1 is the evolvement of prior diabetes dance series with aerobic principle with more eccentric movement. Diabetes dance of Persadia 1 has been inspected effective for lowering fasting blood glucose, but its still unknown about its effect on blood level of total cholesterol. The aim of this research is to analyze the effect of exercise on total cholesterol level. This research is experimental research, with 16 subject of healthy housewifes and divided on 2 groups, that is exercise and control group. Exercise group was given with diabetes dance of Persadia 1 while control group have the unstructured activity. Blood level of total cholesterol (TC) has been measured after 24 hours post single bout exercise (acute-AE) and repeated exercise (chronic exercise-CE). Paired t-test analysis on both acute (AC) (p=0,005) and chronic (CC) (p=0,006) shows TC decrease significantly on control group, as well as on AE (p=0,001) and CE (p=0,001). However, TC tend to increase on CC though not significantly increase (p=0,916), while on CE tend to decrease significantly (p=0,041). This study shows that both acute and cronic exercise decrease total cholesterol level in human blood.

Abstract

Maternal Mortality Rate (MMR) in Indonesia based on 2015’s Health Profile of Indonesia has reached 305 per 100,000 live births. Preeclampsia is known being one of the most related factors of MMR Studies suggest that the use of folic acid may lower the risk of hypertensive disorders in pregnancy. It has been suggested that folic acid and Folic acid containing multivitamins may reduce the risk of preeclampsia by lowering plasma homocysteine concentrations in pregnant women. This cross sectional study aimed to analyze the correlation between intake of Folic acid with preeclampsia. Data was taken using Food&nbsp; Frequency Questionaire (FFQ) with semi quantitative approach by interviewing subjects. Sampling is obtained by simple random sampling with total 80 subjects. Data was analyzed using Chi square test. There were 15% of subjects diagnosed with preeclampsia and 83.3% of women with preeclampsia were deficient of Folic acid. Bivariate analysis showed &nbsp;there is correlation between intake of Folic acid during pregnancy with preeclampsia in Sidotopo Wetan Health Care Centre (p= 0.028), while OR value is 0.158 (95% CI 0.032 to 0.776). It can be concluded that insufficient intake of Folic acid during pregnancy correlate to preeclampsia incidents.
