Abstract

Exercise such as Diabetes dance of Persadia 1 is the evolvement of prior diabetes dance series with aerobic principle with more eccentric movement. Diabetes dance of Persadia 1 has been inspected effective for lowering fasting blood glucose, but its still unknown about its effect on blood level of total cholesterol. The aim of this research is to analyze the effect of exercise on total cholesterol level. This research is experimental research, with 16 subject of healthy housewifes and divided on 2 groups, that is exercise and control group. Exercise group was given with diabetes dance of Persadia 1 while control group have the unstructured activity. Blood level of total cholesterol (TC) has been measured after 24 hours post single bout exercise (acute-AE) and repeated exercise (chronic exercise-CE). Paired t-test analysis on both acute (AC) (p=0,005) and chronic (CC) (p=0,006) shows TC decrease significantly on control group, as well as on AE (p=0,001) and CE (p=0,001). However, TC tend to increase on CC though not significantly increase (p=0,916), while on CE tend to decrease significantly (p=0,041). This study shows that both acute and cronic exercise decrease total cholesterol level in human blood.

Abstract

Maternal Mortality Rate (MMR) in Indonesia based on 2015’s Health Profile of Indonesia has reached 305 per 100,000 live births. Preeclampsia is known being one of the most related factors of MMR Studies suggest that the use of folic acid may lower the risk of hypertensive disorders in pregnancy. It has been suggested that folic acid and Folic acid containing multivitamins may reduce the risk of preeclampsia by lowering plasma homocysteine concentrations in pregnant women. This cross sectional study aimed to analyze the correlation between intake of Folic acid with preeclampsia. Data was taken using Food  Frequency Questionaire (FFQ) with semi quantitative approach by interviewing subjects. Sampling is obtained by simple random sampling with total 80 subjects. Data was analyzed using Chi square test. There were 15% of subjects diagnosed with preeclampsia and 83.3% of women with preeclampsia were deficient of Folic acid. Bivariate analysis showed  there is correlation between intake of Folic acid during pregnancy with preeclampsia in Sidotopo Wetan Health Care Centre (p= 0.028), while OR value is 0.158 (95% CI 0.032 to 0.776). It can be concluded that insufficient intake of Folic acid during pregnancy correlate to preeclampsia incidents.

Abstract

This historical study is focused on Indis style houses and buildings in Java during colonial period. The study discusses the reception and adaptation of Dutch and other cultures together with the local cultures in the transformation and hybridization of Indonesian architectural design production and reproduction. The study concludes that Indis style is a proof for the western inability to avoid the social and cultural influence of the locals, representations of new hybrid identities who want to show themselves as closer to the west and distance from the  indigenous, representation of new identity among those who found a new definite motherland since others consider themselves alien, symbol of modernity for those who were looking for a new social status and justify their existence, and a representation of the history of colonialism itself. Kajian sejarah ini difokuskan pada rumah dan bangunan bergaya Indis di Jawa pada masa kolonial. Tulisan ini mendiskusikan tentang resepsi dan adaptasi budaya Belanda dan budaya lainnya bersama-sama budaya tempatan dalam transformasi dan hibridisasi pada proses produksi dan reproduksi desain arsitektur Indonesia. Kajian ini menyimpulkan bahwa gaya Indis merupakan bukti dari ketidakmampuan Barat untuk menghindar dari pengaruh sosial dan kebudayaan tempatan, representasi dari identitas hibrid baru yang ingin menunjukkan dirinya lebih dekat kepada Barat namun berjarak dari penduduk asli, representas dari identitas baru bagi mereka yang menemukan ibu pertiwi yang baru ketika kelompok lain menganggap mereka sebagai orang asing, simbol modernitas bagi mereka yang sedang mencari status sosial baru serta untuk menjustifikasi keberadaannya, dan sekaligus representasi dari sejarah kolonialisme itu sendiri.

Abstract

Kanker serviks merupakan kanker ginekologi tertinggi di dunia dengan infeksi Human Papilloma Virus sebagai penyebab terseringnya. Di Indonesia, kanker serviks merupakan keganasan yang paling banyak ditemukan dan penyebab kematian utama pada perempuan dalam tiga dasawarsa terakhir. Di Indonesia, setiap tahun terdeteksi lebih dari 15.000 kasus kanker serviks dan sekitar 8.000 kasus di antaranya berakhir dengan kematian. Terdapat beberapa jenis pengobatan yang biasa diberikan pada penderita kanker serviks, namun hasilnya relatif belum optimal. Keadaan ini mendorong usaha penemuan dan pengembangan strategi terapi baru dalam melawan kanker. Pendekatan yang menarik untuk dikembangkan adalah penggunaan kombinasi kemoterapi atau ko-kemoterapi. Ko-kemoterapi merupakan strategi terapi kanker dengan mengombinasikan suatu senyawa kemopreventif yang bersifat tidak toksik dengan agen kemoterapi. Salah satu produk natural yang potensial untuk dikembangkan sebagai agen ko-kemoterapi adalah propolis. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian propolis yang berasal dari Kerjo, Karanganyar, Indonesia terhadap induksi proses apoptosis dan aktivitas antiproliferasi, terutama terkait dengan penekanan ekspresi protein Cyclin D1 dan peningkatan ekspresi protein Bax pada kultur sel HeLa (cell line kanker serviks). Penelitian ini merupakan penelitian eksperimental laboratorik dengan menggunakan post test with control group design. Penelitian dilakukan pada kultur sel HeLa (sel kanker serviks) dengan pemberian propolis. Pengamatan ekspresi protein Cyclin D1 dan ekspresi protein Bax dilakukan dengan metode imunositokimia. Hasil penelitian menemukan bahwa ekspresi rata-rata Cyclin D1 pada kelima kelompok yaitu kontrol 84,13± 1,34µg/ml, EEP 1/2 IC 60,76±4,21, EEP IC 36,56± 3,63µg/ml, EEP 2 IC  24,92± 5,14µg/ml, 5-FU 13,15±3,66µg/ml. Terdapat perbedaan bermakna ekspresi Cyclin D1 antara kelompok perlakuan dibandingkan kelompok kontrol (p < 0,001). Rata-rata ekspresi Bax pada kelima kelompok yaitu kontrol 1,89± 0,46µg/ml, EEP 1/2 IC 69,44± 1,39, EEP IC 80,07± 3,52µg/ml, EEP 2 IC 83,96± 3,26µg/ml, 5-FU 92,78± 4,68µg/ml. Terdapat perbedaan bermakna ekspresi Bax antara kelompok uji dibandingkan kelompok kontrol (p < 0,001). Penelitian ini menyimpulkan bahwa pemberian ekstrak etanol propolis mempunyai pengaruh terhadap penekanan ekspresi Cyclin D1 dan peningkatan ekspresi Bax pada kultur sel kanker serviks (HeLa Cell line). Kata kunci: EEP, Protein Cyclin D1, Protein Bax, HeLa cell line

Abstract

Kanker kolorektal merupakan kanker terbanyak ketiga pada pria, kedua pada wanita, dan menjadi penyebab kematian keempat, sekitar 8% dari penyebab kematian karena kanker. Sebagian besar pasien kanker kolorektal didiagnosis dalam stadium yang sudah tidak dapat dioperasi. Propolis diketahui memiliki aktivitas anti kanker. Mekanisme anti kanker propolis melalui induksi apoptosis dan juga melalui penghambatan proliferasi dalam siklus sel. Penelitian ini bertujuan untuk mengkaji efek anti kanker ekstrak ethanol propolis (EEP) yang berasal dari Kerjo, Karanganyar, Indonesia pada kultur sel adenokarsinoma kolorektal (cell line WiDr) melalui pengaruhnya terhadap ekspresi protein Caspase 8, apoptosis dan proliferasi sel. Penelitian ini merupakan penelitian experimental laboratories, post test with control group design. Penelitian dilakukan pada kultur sel WiDr dengan perlakuan pemberian dosis ekstrak ethanol propolis, kontrol positif (5-FU), kombinasi ekstrak ethanol propolis dengan 5-FU, dan kontrol negatif (tanpa obat). Pengamatan ekspresi protein Caspase 8 dilakukan dengan metode imunositokimia, pengamatan apoptosis dilakukan dengan double staining menggunakan pewarnaan akridin oranye–etidium bromide, sedangkan pengamatan proliferasi menggunakan MTT assay dengan doubling time. Hasil Penelitian menunjukkan bahwa EEP cenderung menekan viabilitas sel WiDr dengan IC sebesar 140 µg/mL. EEP konsentrasi 70,140, 280 µg/mL mampu meningkatkan ekspresi protein Caspase 8 dan menginduksi apoptosis yang sebanding dengan peningkatan konsentrasi yang diberikan. EEP konsentrasi 70 µg/mL dapat menghambat proliferasi sel sebanding dengan EEP konsentrasi 140 dan 280 µg/Ml. Penelitian ini menunjukkan EEP mampu menekan viabilitas sel WiDr. Aktivitas ini kemungkinan terkait dengan kemampuannya dalam meningkatkan ekspresi protein Caspase 8 dan apoptosis sebanding dengan peningkatan konsentrasi yang diberikan. EEP pada konsentrasi terkecil yang diuji (70 µg/mL) mampu menghambat proliferasi pada sel WiDr sebanding dengan dosis yang lebih tinggi. Kata kunci: EEP, Caspase 8, apoptosis, proliferasi, cell line WiDr

Abstract

ABSTRAKLupus nefritis (LN) terkait dengan penebalan membran basal glomerulus. Pengendapan kompleks imun  memicu kaskade respon inflamasi disertai aktivasi reactive oxygen species (ROS), akhirnya terjadi aktivasi caspase-1, dan caspase-1, mengaktifkan IL-1B dan IL-18 yang berlanjut terjadinya apoptosis dan nekrosis dan akhirnya terjadi kerusakan ginjal. Peningkatan ekspresi caspase-1 dapat dicegah oleh N-Asetil Sistein (NAS), yang merupakan suatu senyawa dengan efek antioksidan dan antiinflamasi. Penelitian ini bertujuan untuk mengetahui pengaruh NAS terhadap ekspresi caspase-1 dan terjadinya apoptosis dan nekrosis ginjal pada mencit model lupus nefritis. Penelitian ini merupakan penelitian eksperimental laboratoris, dengan sampel 24 ekor mencit Balb/C betina yang dibagi menjadi kelompok kontrol, LN, dan LN+NAS. Untuk membuat model LN, hewan coba diberikan injeksi 0,5 ml pristan intraperitoneal dosis tunggal. NAS diberikan secara peroral dengan dosis 4,7 mg/hari) selama delapan minggu. Mencit kontrol tidak diinokulasi selama penelitian. Ekspresi caspase-1 dihitung dari 100 sel makrofag yang immunoreaktif dengan teknik imunohistokimia dan kejadian apoptosis dan nekrosis dihitung dengan dengan teknik histopatologi. Hasil penelitian menunjukkan bahwa, pemberian NAS menurunkan ekspresi caspase-1 (22,8 ± 6,4 vs 31,4 ± 7,5 per 100 sel makrofag imunoreaktif; p = 0,000)  dan menurunkan kerusakan ginjal (6,75 ± 2,92 vs 9,88 ± 3,56; p =0,010) dibandingkan kelompok LN. NAS secara bermakna menurunkan ekspresi caspase-1 dan derajat kerusakan ginjal pada mencit model LN. The deposition  of immune complexes trigger a cascade of inflammatory response accompanied the activation of reactive oxygen species (ROS), finally happened activation caspase-1, and enable the IL-1B and IL-18 to be the active form, and to be continued into occurrence of apoptosis and necrosis and eventually the kidney injury. Increased of caspase-1 expression can be prevented by N-Acetyl Cysteine (NAS), which is a compound with antioxidant and anti-inflammatory effects.This study aimed to analyze the effects of NAS on the expression of Caspase-1 and degree of renal injury in mice models of lupus nephritis. This study is an experimental research laboratory, with a sample of 24 females Balb/C mice were divided into a control group, LN and LN+NAS. To create a model LN, experimental animals given intraperitoneal injection of 0.5 ml Pristan single dose. NAS administered orally at a dose of 4.7 mg/day for eight weeks. Control mice not inoculated during the study. Expression of Caspase-1 was calculated from 100 macrophage cells immunoreactive with immunohistochemical and degree of renal injury with histopathological techniques. One way analysis of variance (Anova) for caspase-1 expression and degree  of renal injury, and p<0.05 were used to determine the significant differences. The provision of NAS decreased the expression of caspase-1 (22,8 ± 6,4 vs 31,4 ± 7,5  per 100 macrophage immunoreactive cells; p = 0.042) and degree of renal injury (6,75 ± 2,92 vs 9,88 ± 3,56; p =0,010) compared to LN group respectively. Abstract

Nephrotoxic effects of Doxorubicin (DXR) is still a problem in clinical practice. On the other hand Pentoxyfilline (PTX) as an electron-donor material can be nephroprotective. Therefore, combination of DXR and PTX would be expected to reduce nephrotoxic effects of DXR. In this study we examined the effects of PTX on TGF-B1 expression and interstitial fibrosis in an experimental model of DXR nephropathy in mice. Mice were divided into three groups of eight each i.e. untreated Swiss mice (controls), DXR treatment alone to induce nephropathy, and DXR treatment followed by PTX. Following 4 week treatment, each group was sacrificed. Examination of TGF-B1 expression was carried out by immunohistochemistry employing monoclonal antibody. Interstitial fibrosis examination was performed by a histopathologist using Verheoff van Giesen staining and the one way Anova was used for statistical analysis. It was observed that DXR treatment followed by PTX treatment prevented the increase of TGF-B1 expression and interstitial fibrosis in mice with DXRnephropathy (p&lt;0.05). These findings suggested the beneficial nephroprotective effect of PTX.

Abstract

Hepatocellular carcinoma (KHS) is 90% of primary tumors in the liver. One of natural treatment that have anti-cancer activity is propolis. Its anti-cancer activity works through induction of apoptosis and inhibition of cell proliferation. Bcl2 belongs to the family group B-cell lymphoma 2 protein which acts as an intrinsic pathway apoptotic protein. p21 is a tumor suppressor protein that has a primary function in inhibiting cell cycle progression. This study was aimed to determine the anti-cancer effect of propolis ethanol extract (EEP) originating from Kerjo, Karanganyar on hepatocellular cancer cell culture (Hep G2) through its potential as an anticancer in increasing p21 protein expression, reducing bcl2 protein expression in suppressing apoptosis and reducing cell proliferation . This research is an experimental laboratories with post test with control group design. Performed on Hep G2 cells with the treatment of EEP concentrations (½IC50, IC50, 2IC50), sorafenib IC50, a combination of EEP IC50 + sorafenib IC50, and controls. Observation of Bcl2 and p21 protein expression by immunocytochemistry method, observing cell proliferation with doubling time, observing apoptosis with flowcytometry. We use ANOVA statistical test, followed by the Tuckey post hoc test, were stated to be significant if p< 0.05. The inhibition concentration of 50% Hep G2 cells by EEP was 65.2 μg / mL, and sorafenib (6.08 μg / mL). EEP concentration of 2IC50 (p = 0.001), sorafenib 1 IC50 (p = 0.001), 1IC50 EEP + 1IC50 sorafenib (p= 0.001) able to increase p21 protein expression. The 2 EEP concentration (p= 0.040), was able to reduce the expression of Bcl2 protein, with p= 0.001. The strength of EEP 1IC50 is equivalent to sorafenib IC50 in suppressing 24-hour proliferation (p> 0.05). The strength of EEP ½IC50 is equivalent to sorafenib IC50 in suppressing proliferation 48 hours (p> 0.05). The strength of EEP 1IC50 is better than sorafenib IC50 in suppressing proliferation of 72 hours (p> 0.05). All EEP concentrations increase p21 protein expression, with the EEP 2IC50 group providing the strongest effect. All EEP concentrations were able to reduce cell proliferation, with the EEP 2IC50 group, giving the strongest effect. It was concluded that concentration of 2IC50 EEP has an effect on decreasing the expression of bcl2 protein and hepatoma cell apoptosis.Keywords: Ethanol Extract Propolis, Bcl2, P21, Apoptosis, Proliferation, Hep G2 Cells

Increased pressure in the portal vein Portal Hypertension (HP) causes esopha­geal varices and the more likely the patient to bleed. Endothelial dysfunction in cirrhosis of the liver causes a decrease in nitric oxide (NO) levels resulting in an imbalance between relaxation factors and portal vein contractions and increasing hs-CRP levels. Simvastatin can increase NO levels and improve endothelial dysfunction in patients with SH and can reduce levels of hs-CRP. This study aimed to determine the effect of simvastatin administration on nitric oxide and hs-CRP level in patients with liver cirrhosis.Subjects and Method: This was a randomized controlled trial conducted at Dr. Moewardi hospital, Surakarta, Central Java. A sample of 29 patients was divided into treatment and control groups. 14 patients in the treatment group were given simvastatin dose of 20 mg. 15 patients in the control group received a placebo. The dependent variable was a nitric oxide and hs-CRP levels. The inde­pendent variable was Simvastatin administration. Nitric oxide serum level was measured by an enzyme-linked immunosorbent assay (ELISA). Hs-CRP level was measured by the immune­chemilu­minescent method. The data were analyzed by t-test.Results: NO level after Simvastatin 20 mg administration (Mean= 7.93 µg/mL; SD= 1.78 µg/ mL) was higher than before (Mean= 5.14 µg/mL; SD= 2.25 µg/mL) and it was statistically significant (p = 0.001). Hs-CRP level after Simvastatin 20 mg administration (Mean= 2.04 mg/L; SD= 0.80 mg/L) was lowered than before (Mean= 7.82 mg/L; SD= 1.15 mg/L) and it was statistically significant (p= 0.001).Conclusion: Simvastatin administration increases NO serum levels and reduces the hs-CRP level in patients with liver cirrhosis.Keywords: nitric oxide, hs-CRP, simvastatin, liver cirrhosisCorrespondence: Budhi Hariyanto. Abstract

The purpose of this writing is to express that friendship is more important than hostility. The reason of hostility is annoyance, emotion and lack of patience in facing problems. The effect of it is the destruction of friendship and the increase in annoyance among them. Feminist is a method that is used in this writing. The application of this method is to express that man and woman have the right to make their own decisions. Both of them will respect and understand. They will have responsibility for everything they have done. This method is also expressing that human have freedom in their life. In this novel, there are four women. They are Carmen, Bridget, Lena and Tibby. The four of them spend their summer holiday into different places. In their summer holiday, they have experienced incidents which can destroy friendship. Carmen hates her father, Lena hates Kostos, Bridget hates Eric and Tibby hates Bailey. Finally, they realize that annoyance between them can destroy their friendship. The writer used Feminist approach because the main characters wanted to destroy the patriarchy of man. They insist that man can respect woman’s opinion.   Key words: Friendship, Conflict, Respect
