Abstract

Abstract

This Community Service Program (PKM) aims to improve the understanding and capacity of parents, especially mothers, in implementing resilient parenting as a strategy to build disaster awareness in early childhood. The activity entitled "Smart Mom for Disaster Preparedness" was carried out in Kasemen District, Serang City in an area that is classified as disaster-prone through a qualitative participatory approach with the Focus Group Discussion (FGD) method. Participants in the activity consisted of mothers, PAUD teachers, and partner school principals. The material provided focused on the importance of effective communication based on love languages to strengthen the emotional relationship between mother and child as the basis for children's psychological resilience in facing emergency situations. The results of the FGD showed an increase in participants' understanding of the concept of resilient parenting and the emergence of new awareness regarding the strategic role of families in disaster mitigation education. Participants also began to apply a more empathetic and reflective communication approach in daily parenting. This activity proves that community-based education with a participatory approach can be an effective strategy in building disaster preparedness from an early age, starting from the family circle.

Abstract

BACKGROUND: Currently, identification of breast cancer stem cells (BCSCs) commonly relies on CD24-/CD44+ expression profiles. However, few studies have integrated genomic mutation data with experimental gene expression validation in CSC and non-CSC populations. Genotyping results of CD24-/CD44+ MDAMB-231 cells revealed 36 mutations in BCSCs compared to non-BCSCs, with upregulated FOXO1 and FYN that might represent promising candidate biomarkers for this subpopulation. Therefore, in this study, single nucleotide polymorphism (SNP) and quantitative polymerase chain reaction (qPCR) analysis were performed to assess the association between mutations and expression trends of FOXO1 and FYN in MDAMB-231 cell, as breast cancer cell model with stem-like traits and well-characterized profile.METHODS: Genomic DNA was isolated from BCSC and non-BCSC DNA from the MDAMB-231 cell line. Mutation analysis was conducted using PLINK, while gene expressions of FOXO1 and FYN were quantified by one-step SYBR Green-based qPCR, using 18s rRNA as a reference. Data was then analyzed with the Livak (2−∆∆Ct) method.RESULTS: Among 36 mutations found in BCSCs of the MDAMB-231 cell line, PTEN (rs786204914) and CHEK2 (rs587782401) were identified as pathogenic. While FOXO1 (2.989±2.817 vs. 1.072±0.388) and FYN (1.405±0.072 vs. 0.855±0.140) mRNA levels were found to be higher in CSCs compared to non-CSCs, though these differences was not statistically significant.CONCLUSION: Pathogenic mutations in CHEK2 and PTEN were detected within BCSC population, implying a potential influence on the expression of FOXO1 and FYN, though not statistically significant. These findings suggest a possible, but as yet unverified, association between gene mutations and expression patterns, emphasizing the importance of further functional studies to validate FOXO1 and FYN as biomarkers for BCSCs.KEYWORDS: breast cancer stem cells, FOXO1, FYN, PTEN, CHEK2, mutation, biomarker

Abstract

Recent insights reveal that liver diseases influence not only hepatic function but also disrupt gut microbial balance through the gut–liver axis. The gut–liver axis establishes a bidirectional relationship between the intestines and the liver, allowing microbial by-products such as short-chain fatty acids (SCFAs) to influence liver function and health. Short-chain fatty acids are known to maintain intestinal epithelial integrity, reduce inflammation, and support liver function. Probiotic bacteria including Lactobacillus, Bifidobacterium, and Clostridium, are natural SCFA producers and may offer therapeutic potential for liver disease by targeting the gut-liver axis. This systematic review was conducted using the PRISMA 2020 methodology to identify and evaluate preclinical studies examining the impact of SCFA-producing probiotics on liver disease. We searched PubMed, Scopus, and Google Scholar from August to October 2023, using predefined inclusion criteria based on the PICO framework. The SYRCLE risk of bias tool was employed to evaluate potential biases. A total of 14 animal studies fulfilled the inclusion criteria and were incorporated into the final analysis. The included studies demonstrated that SCFA-producing probiotics improved liver function by reducing serum liver enzymes (ALT, AST), increasing tight junction proteins (occluding, ZO-1), modulating pro-inflammatory cytokines (IL-1β, IL-6, TNF-α,), and improving lipid metabolism. These outcomes were mediated by increases in SCFA levels and improved gut barrier integrity in models of NAFLD, ALD, NASH, and autoimmune hepatitis. These findings support the promising potential of SCFA-producing probiotics as adjunctive therapies for liver disease through modulation of the microbiota-gut-liver axis. Yet, continued research is needed to determine strain-specific efficacy, optimal dosage, long-term safety, and clinical applicability. Future research should also explore personalized probiotic strategies and the integration of probiotic therapy into standard liver disease management.

Abstract

The emergence of Bitcoin and blockchain technology as a decentralized financial infrastructure has shaken the foundations of the conventional criminal justice system, particularly in addressing money laundering offenses. This study aims to evaluate the effectiveness of existing criminal policies and reformulate the criminal law approach toward anonymous and cross-jurisdictional digital assets. Using a legal-normative method and comparative analysis of international policies, it was found that policies overly reliant on formal financial institutions become irrelevant in detecting peer-to-peer blockchain transactions. This study proposes a model for reconstructing criminal policy based on the principles of responsive law, adaptive digital surveillance (RegTech and SupTech), and transnational jurisdictional cooperation. The findings of this study are expected to provide conceptual and practical contributions to the design of a relevant, progressive, and digitally grounded criminal justice system.

Abstract

Qur’anic education is essential and highly needed for all groups of people—children, adults, and the elderly alike. Therefore, improving the quality of Qur'anic recitation is crucial and should be instilled from an early age. Qur’anic education serves as an alternative solution to the various problems found within society. The Qur'anic Education Center (TPQ) is one of the facilities that helps children improve and enhance their Qur'an recitation skills, as well as a means to instill Qur'anic values in them from a young age. As a result, children will be able to read the Qur’an correctly and fluently through the application of various teaching methods used in each TPQ, including the talaqqi and participatory methods, which actively involve children in correcting mistakes in Qur'anic recitation. This community service activity uses a descriptive analysis method, where data is collected through direct observation, and the results show that the talaqqi and participatory methods are highly effective and beneficial for children.
