Abstract

Penelitian ini bertujuan untuk mengevaluasi hasil vaksinasi rabies dan membandingkan kenaikan titer antibodi yang terbentuk di Kecamatan Cisolok dan Jampang Tengah sebagai Kawasan endemik rabies Kabupaten Sukabumi dan mengasosiasikan titer antibodi yang terbentuk dengan umur, jenis kelamin, dan cara pemeliharaan anjing yang diperiksa . Sebanyak 211 anjing diamati pada studi ini. Sampel serum prevaksinasi dan pascavaksinasi diambil dan selanjutnya diuji dengan uji ELISA tidak langsung untuk mengetahui titer antibodinya. Data titer antibodi dari masing-masing sampel dianalisis secara deskriptif, sedangkan asosiasi umur, jenis kelamin, dan cara pemeliharaan dengan titer antibodi yang terbentuk pascavaksinasi dihitung menggunakan uji chi-square. Hasil pengujian menunjukkan seroprevalensi rabies di Kecamatan Cisolok dan Jampang Tengah sebelum vaksinasi adalah 17,3% dan 2,6% dengan titer 0,58 EU/mL dan 0,55 EU/mL, sedangkan seroprevalensi setelah vaksinasi menjadi 62,5% dan 93% dengan titer 1,29 EU/mL dan 1,33 EU/mL. Variabel umur, jenis kelamin, dan cara pemeliharaan tidak berasosiasi nyata dengan peningkatan titer antibodi rabies setelah vaksinasi. Hasil studi menyimpulkan bahwa seroprevalensi rabies di kawasan endemik rabies di Kabupaten Sukabumi meningkat setelah dilakukan vaksinasi dari sebelumnya 11% menjadi 75,8%, dengan nilai peningkatan titer (Δ titer) sebesar 1,09 EU/mL.

Abstract

The establishment and spread of highly pathogenic avian influenza (HPAI) viruses of the H5N1 subtype in birds and coincident infections in humans since 2003 have raised concerns that we may be facing an influenza pandemic caused by an H5N1 influenza virus [4]. Globally, from January 2003 to 2 March 2017, there were 860 cases of human infection with avian influenza A(H5N1) virus reported from 16 countries worldwide. Of these 860 cases, 454 were fatal (CFR of 53%). The last case was reported from Indonesia on 26 September 2017 [7]. Although the human cases of H5N1 in Indonesia has decreased significantly since 2010, according to WHO until 2017 there have been reported 200 cases H5N1 in  human with 168 cases of deaths. The last case was reported from Indonesia on 26 September 2017. This is indicate Indonesia as the highest fatal case of H5N1 globally. With continued incidence of avian influenza due to existing AI H5N1 viruses in poultry, the avian influenza H5N1 has been believed remain to threaten Indonesia [6].Passive immunization using specific antibody against AI H5N1 from bovine colostrum is one of an alternative to control H5N1 virus infection due to lack of H5N1 vaccine production for human. Bovine colostrums consider an ideal alternative antibody source, as the antibody in the bovine’s blood is transported to mammary gland easily and accumulates in the colostrum in large quantities. As a “biological factory”  and the source of natural antibody, bovine colostrum could be designed to produce the specific antibody against certain disease for animal and human by immunizing the dry cow (with the antigen of interest).Vaccine is an antigenic material used to produce active immunity against diseases. Vaccination is the administration of vaccine to the individual to generate immunity against a disease [2]. According to [1], the exposure of a dry cow against antigen (vaccine) will produce specific antibody in their blood circulation. Therefore, evaluation on the status of antibody against AI H5N1 following the vaccination of dry Holstein cows with killed AI H5N1 vaccine is needed.

Abstract

Immunomodulator adalah zat yang memiliki kemampuan untuk meningkatkan  atau menekan respon imun. Cox (1988) melaporkan bahwa pengaruhnya  selain terhadap  respon imun, imunomodulator juga dapat memodulasi haematopoiesis, termasuk peningkatan jumlah RBC dan WBC (leukosit), peningkatan PCV dan aktivasi makrofag.  Sapi friesian holstein (FH) merupakan sapi perah yang dapat digunakan sebagai hewan donor penghasil immunoglobulin G anti AI H5N1 melalui produk kolostrumnya (Esfandiari et al, 2007).  Guna meningkatkan titer immunoglobulin pada hewan donor hiperimun sera umumnya hewan diberi imunomdulator.  Berbagai jenis bahan seperti glucan, lectin, dan berbagai jenis polisakarida dari tanaman maupun hewan (Alamgir dan Uddin 2010) serta polipeptide ribonukleotida dapat digunakan sebagai imunomodulator.(Hess dan Greenberg 2012)  Penggunaan  polipeptida ribonukleotida sebagai imunomodulator ada sapi saat ini belum banyak diteliti.  Pemberian imunomodulator memberikan dampak perubahan gambaran leukosit secara langsung maupun tidak langsung.  Dampak pemberian imunomodulator jenis polipeptide ribonukleotida pada sapi FH bunting trimester terakhir untuk tujuan produksi hiperimunsera melalui produksi kolostrum belum pernah dilaporkan.  Penelitian ini bertujuan untuk mengetahui efek pemberian imunomodulator peptide ribonukleotida pada sapi FH yang disuntik antigen AI H5N1 inaktif.

Abstract

Outbreaks of zoonotic diseases such as avian influenza and rabies during the last decade have caused fatalities and fear among people in Indonesia. The Government of Indonesia, the World Health Organization (WHO), the Food and Agriculture Organization (FAO), as well as other international and domestic NGOs have been working together to control those diseases and prevent human pandemic. However, the zoonotic disease control programs will not succeed without active participation of local communities including young people.Indonesia Scout Movement (Gerakan Pramuka) is a non-formal educational organization having more than 20 million members that serve educational process outside the school and outside the family using basic principles and methods of scouting (GOI 2010). Members of Gerakan Pramuka consist young people of different age categories namely  Cub Scouts/Siaga (ages 7 to 10 y.o), Scouts/ Penggalang (ages 11 to 15 y.o), Rover Scouts/ Penegak  (ages 16 to 20 y.o), Pandega (ages 21 to 25 y.o), and Adult members/Pembina  (ages > 25 y.o, or married person).Nowadays, Gerakan Pramuka has more than 20 millions members distributed in all districts in Indonesia, and thus may become a potential media to enhance young people awareness on zoonotic diseases control programs. Dissemination of information on the diseases and its prevention can be done through scout regular events such as Pesta Siaga  (Cub Scout Gathering), Jamboree (Scout Gathering), Raimuna (Rover Scout Gathering), and Karang Pamitran (Adult Scout member Gathering).Faculty of Veterinary Medicine, IPB, in collaboration with IPB Student Scouting Activity Unit, has developed a model for the community participatory program for zoonotic diseases control in Indonesia through scouting activities. Cub Scout Leader Gathering for Avian Influenza Awareness is a scout event that designed as a method for dissemination of information regarding prevention of avian influenza transmission to school age children and their relatives.

Abstract

Avian Influenza (AI) is well known as Avian flu, Fowl pest, Fowl plaque, or Flu burung, caused by influenza virus type A.  This virus is belonged to Orthomyxoviridae and could infect many kind of species such as bird, pig, horse, cat, as well as human.  Vaccination is applied to control the disease using inactivated vaccine, which induced the specific antibody against H5 antigen.  Passive immunization using specific antisera against H5 antigen is thought to be usefull in controlling the disease especially in the treatment of infected host.  In this experiment the neutralization ability of specific antisera against H5 were studied using various field viral isolates subtype H5N1.  Antisera was developed in Cavia porcellus which vaccinated with AI subtype H5N1 in activated vaccine.  The titre of antisera obtained is 28 used HI test.  Four AI virus subtype H5N1 isolates from 2003 to 2006 agains viral were we as tested virus.  The neutralization test showed that the sera were able to neutralizing 10 4 EID50 AI virus H5N1 with neutralization index range of 1.1-1.3.  The result indicated that the specific antisera had the neutralization potency to the field virus.   Key words: avian influenza, neutralization test, neutralization index

Abstract

Avian Beta Defensin 2 (AvBD2) gene, which is located in chromosome 3, plays an important role in the immune system of the chicken by inhibiting the development of microorganisms such as bacteria that infect body tissues. Defensins are produced through epithelial cells immediately after tissue injury or infection, which then processes the maturation of dendritic cells to initiate an immune response in the lymph nodes. The purpose of this study was to discover the polymorphism of the AvBD2 gene in IPB-D1 chickens. PCR and direct-DNA sequencing methods were used to identify the diversity of intron 1, exon 2, and intron 2 AvDB2 genes in 47 chickens. Genotype and allele frequency, and heterozygosity calculations were carried out to obtain information of the AvBD2 gene polymorphism. A total of 10 single nucleotide polymorphisms were found in the AvBD2 gene located in intron 1 (g.4843T>A, g.4853G>A, and g.4859T>C), exon 2 (g.4881A>G, g.4889G>A, and g.5002C>T), and intron 2 (g.5075C>T, g.5111T>G, g.5116G>T, and g.5177G>T). All SNPs are polymorphic. The g.5002C>T mutation causes changes in the amino acid Ala to Val which has the potential to be a candidate for characterizing disease resistance in IPB-D1 chickens.

Abstract

Tea mistlestoe extract (Scurrula oortiana) has cytotoxic activity which is potential to be used in preventing viral induced-chicken tumor. The following study was designed to evaluate the effects of different inoculation routes, dosage levels, and strains of embryonated chicken eggs as media for testing the tea mistlestoe extract (Scurrula oortiana) antiviral activity. Proper inoculation route was examined by inoculation of the extract at dose level of 0,2 mg/egg into embryonated layer eggs via allantoic cavity, chorio-allantoic membrane, and yolk sac. Effect of dose level of tea mistlestoe extract on embryo development was examined in groups of embryonated broiler eggs inoculated with the extract at 0.02, 0.2, 2, 20, or 200 mg/egg. Inoculation of tea mistlestoe extract into allantoic cavity was the safest procedure as indicated by the absence of embryos mortality, and faster embryo growth compared to those of chorio-allantoic membrane and yolk sac-inoculated eggs. The extract induced different growth effects when inoculated into embryonated layer or broiler eggs. Administration of the extract at dose levels between 0,02–200 mg/egg reduced significantly the weight of broiler embryoes, but not the relative weights of liver, heart and spleen. Administration of similar dosage in layer embryoes did not cause any significant difference in the embryoes weight. This study suggests that the study of antiviral activity of tea mistlestoe extract in embryonated chicken eggs should be carried out on embryonated eggs of layer breeds and the extract should be inoculated via allantoic cavity. Abstract

ABSTRAKPenelitian ini bertujuan untuk menganalisis gambaran sel darah putih dan sel darah merah sebagai indikator ketahanan tubuh pada anak ayam sentul. Penelitian ini menggunakan 20 ekor ayam betina dan 20 ekor ayam jantan yang dikawinkan, masing masing terdiri dari 10 ekor ayam dengan konsentrasi IgY tinggi dan 10 ekor ayam dengan konsentrasi IgY rendah. Berdasarkan hasil perkawinan tersebut diperolah 34 ekor anak ayam yang terdiri dari 17 ekor anak ayam yang berasaldari induk dengan konsentrasi IgY rendah. Parameter yang diamati meliputi gambaran sel darah merah (eritrosit, hematocrit dan indeks eritrosit) dan gambaran sel darah putih (leukosit dan diferensiasinya). Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan 2 perlakuan, semua perlakuan diulang 10 kali. Data yang diperoleh dianalisis menggunakan t-test program SAS.  Hasil penelitian ini menunjukan bahwa perbedaan konsentrasi IgY pada induk tidak mempengaruhi sel darah merah dan sel darah putih. Namun terdapat kecendrungan nilai limfosit yang rendah dan H/L yang tinggi pada anak ayam yang berasal dari induk dengan konsentrasi IgY rendah. Hasil ini mengindikasi bahwa anak ayam yang berasal dari induk dengan konsentrasi IgY rendah lebih rentan terhadap cekaman panas. Kata Kunci: ayam sentul, IgY,  gambaran darah, ketahanan tubuhABSTRACTThis study was to aim the description of white blood cells and red blood cells as indicators of immunity in Sentul chicks. This study used 20 hen chickens and 20 cock-chicken mated, consisting of 10 chickens with high IgY concentration and 10 chickens with low IgY concentrations. Based on the results of mating, 34 chicks were obtained consisting of 17 chicks that were from the hen with low IgY concentration. The parameters observed included the description of red blood cells (erythrocytes, hematocrit and erythrocyte index) and the description of white blood cells (leukocytes and their differentiation). The experiment was conducted as a completely randomized design with 2 treatments, all treatments were repeated 10 times. The data obtained were analyzed using the SAS program t-test. The results of this study indicate that the difference in IgY concentration in the hen did not effect on red blood cells and white blood cells. However, there is a tendency for low lymphocyte values and high H/ L in chicks originating from hens with low IgY concentrations. These results indicate that chicks from hens with low IgY concentrations are more susceptible to heat stress.Keyword: blood picture, IgY, immunity, sentul chicken

Abstract

Rabies is a zoonotic disease. Rabies protection level detection was performed using antibody titration. Blood sampling activities in the field require special handling to avoid blood lysis, the sample delivery requires a cold chain with a stable temperature. Alternative method forwhole blood sample delivery using filter paper were carried out on 48 samples from susceptible animals (dogs and cats) transpored through the Soekarno Hatta Agricultural Quarantine Center. This  study was designed to investigate the feasibility of filter paper sampling of blood at temperature of 26 °C to detect of rabies antibodies using indirect method of ELISA. The results of statistical analysis of  Randomized Block Design and Tukey's test showed that the antibody titres of whole blood extracted from filter paper diluted in the PBS T 100 μl were equivalent to antibody titres of serum. Assesment of filter paper capability has sensitivity and specificity as much as 96% and 76%, positive predictive value of 67%, negative predictive value of  94% and reliability level of filter paper were 0.5 is moderate category. This indicates that the filter paper can be used as an alternative method of blood transport medium for rabies ELISA test.

Abstract

Equine Infectious Anemia (EIA) disease is caused by EIA Virus (EIAV) from genus Lentivirus, subfamily Orthoretrovirinae, family retroviridae. This virus causes a persistent infection and potentially fatal in Equidae. There is no scientific literature yet for this disease in Indonesia. In order to determine this disease in our country, it is necessary continuing laboratory detection in imported Equidae and conducted further surveillance in Equidae throughout Indonesia. This research is aimed (1) to detect of any antibodi titre of EIAV in imported horse, (2) To get information about the relationship between laboratory result with the secondary datas of origin country. The EIAV antibody detected by indirect enzyme-linked immunosorbent assay (indirect ELISA). The secondary datas were collected from Health Certificate of origin country, World Animal Health Information System (WAHIS) Interface OIE and literatures. Total of 133 blood samples were collected from imported horse, ex-imported horse and lokal breed horse. The result showed that all of imported horse, ex-imported horse and lokal breed horse were negative antibody against EIAV. The initial information on Equine Infectious Anemia indicated that health requirement in imported horse has fulfilled, so that imported horse was free from EIAV.
