Abstract

The aim of this study was to evaluate the effect of different IgY concentrations on the production, reproduction and immunity of Sentul chicken. This research used 20 month-old Sentul chickens which consisting of 10 chickens with high IgY concentration and 10 chickens with low IgY concentration Parameters observed included prelayer performance (feed consumption, weight gain, feed conversion, morbidity and mortality), layer performance (feed consumption, egg production, egg weight, feed conversion, hatchability, morbidity and mortality), and immunity at chicks (IgY concentration of eggs and chicks serum). The experiment was conducted as a completely randomized design. The treatments were high and low IgY concentration. All treatments were repeated 10 times. The data obtained were analyzed using the SAS program t-test. Morbidity and mortality were analyzed descriptively. The results of this research indicated that the prelayer period, chickens with high IgY concentration had a higher performance than chicken with low IgY concentration. This is caused by about 30% chickens with low IgY were in sick condition for approximately seven days. Further, the layer period showed that the chickens with high IgY concentration had lower production performance than the chicken with low IgY concentration. The study concluded that chickens at low IgY concentrations are susceptible to exposure to antigens that lead to decreased productivity. In normal conditions hens, Sentul with high IgY concentration resulted in low production. The observation of chicken immunity showed that concentration of IgY from chicks and eggs did not differ between hens with high IgY concentrations and hens with low IgY concentrations.

Abstract

ABSTRACT Cockroaches are insects that act as mechanical vectors of pathogenic agents to humans such as Salmonella sp. The aim of this study was to identify the presence of Salmonella sp. in cockroach bodies on cargo and passenger ships. This research was conducted at Baubau Port from October to December 2017. A total of 24 ships were examined consisting of 12 passenger ships and 12 cargo ships. Cockroaches were collected from rooms inside each ship namely galley, bridge, deck and bathroom. A total of 3 196 cockroaches caught consisting of Periplaneta americana (69.50%), Blattella germanica (29.60%), Periplaneta brunnea (0.66%), Pseudophorapsis sp. (0.03%), Pycnoscelus surinamensis (0.03%), Periplaneta australasiae (0.03%), and Neuphoeta cinerea (0.03%). The presence of Salmonella sp. were examined for cockroach species with a large number of population, namely Periplaneta americana and Blattella germanica. A total of 95.3% of the 42 cockroaches examined in the laboratory contained Salmonella sp.. Based on this research it can be concluded that the cockroaches collected from the ships leaning at Baubau Port can serve as a vector of salmonellosis for both ship crew and passengers. Keywords: Cockroaches, Ships, Salmonella sp, Salmonellosis

Abstract

Koi herpesvirus (KHV) is one of the major pathogen for koi and common carp which cause high mortality and economic losses for the farmer. The purpose of this study was to determine the efficacy of glycoprotein-11 (GP-11) KHV DNA vaccine and compared to GP-25 KHV DNA vaccine. The vaccine in the form of naked DNA plasmidwas delivered by intramuscularly injection to the 3-month-old koi. The fish were divided into six groups, i.e. unvaccinated group (negative control C- and  positive control C+), and vaccinated group (2.5 μg/100 μl of GP-11 (group 1), 7.5 μg/100 μl of GP-11 (group 2), 12.5 μg/100 μl of GP-11 (group 3), and 12.5 μg/100 μl of GP-25 (group 4)). At day 42 post vaccination, all fish of each groups were challenged by injecting KHV titre 10-3 FID50. Number of dead fish was counted everyday after the challenge until 30 days. The results showed that vaccinated fish were had survival rate of 83.33-93.33% (group 2, 3 and 4). It’s show that GP-11 KHV DNA vaccine has high efficacy. As conclusion, GP-11 DNA vaccine could be an alternative DNA vaccine for preventing KHV infection.

Abstract

Kemampuan promoter dalam mengatur ekspresi gen penyandi protein imunogenik sangat menentukan efikasi suatu vaksin DNA. Penelitian ini bertujuan untuk mengukur tingkat ekspresi protein dan mRNA RNA2 penyandi coat protein (CP) virus viral nervous necrosis (VNN) yang dikendalikan oleh dua promoter berbeda, yaitu promoter β-aktin ikan medaka (mBA), dan keratin ikan flounder Jepang (JfKer). Uji ekspresi CP dilakukan menggunakan embrio ikan lele dumbo (Clarias sp.) sebagai model, sedangkan analisis mRNA dilakukan menggunakan ikan kerapu tikus. Konstruksi vektor ekspresi pmBA-CP dan pJKer-CP dengan konsentrasi 50 ng/μL KCl 1 M disuntikkan ke embrio ikan lele dumbo fase 1-2 sel. Sebanyak 30 embrio ikan lele dumbo diambil pada jam ke-6, 8, 10, 12, 14, dan 16 pascainjeksi untuk analisis protein. Hasil SDS-PAGE menunjukkan adanya protein berukuran sekitar 42 kDa, dan analisis western blot menggunakan antibodi (Ab) poliklonal anti-VNN membuktikan bahwa protein tersebut adalah CP. Keberhasilan deteksi protein spesifik menggunakan Ab anti-VNN tersebut menunjukkan bahwa embrio ikan lele dapat digunakan untuk menguji potensi produksi protein imunogenik yang dikendalikan oleh promoter berbeda. Pengujian ini juga menunjukkan bahwa, aktivitas promoter mBA lebih tinggi daripada promoter JfKer, sehingga uji ekspresi mRNA dilakukan menggunakan konstruksi pmBA-CP. Benih ikan kerapu tikus (panjang badan sekitar 5 cm) diinjeksi dengan pmBA-CP secara intramuskular dengan dosis 12,5 μg/ekor. Total RNA diekstraksi dari daging pada waktu 6, 12, dan 24 jam pascainjeksi. Hasil RT-PCR menunjukkan adanya ekspresi mRNA CP pada 24 jam pascainjeksi. Hal tersebut menunjukkan bahwa promotor mBA aktif mengendalikan ekspresi CP pada ikan kerapu tikus, dan pmBA-CP berpotensi digunakan sebagai vaksin DNA untuk menginduksi kekebalan ikan kerapu terhadap infeksi VNN.

Abstract

Provinsi Jawa Barat merupakan satu dari 24 provinsi di Indonesia yang dilaporkan belum bebas Rabies. Usaha pembebasan wilayah Jawa Barat dari penyakit Rabies sangat strategis mengingat posisi Jawa Barat berada di antara daerah bebas Rabies. Jampang Tengah menjadi salah satu kecamatan di Sukabumi yang pada tahun 2016 dilaporkan telah terjadi 13 kasus gigitan Rabies. Penelitian ini menggunakan 81 sampel darah anjing lokal yang dikoleksi dari enam desa di kecamatan Jampang Tengah dan diperiksa dengan menggunakan alat Rayto® hematology analyzer. Parameter profil hematologi dikelompokkan berdasarkan perbedaan umur dan jenis kelamin anjing. Hasil pemeriksaan hematologi secara umum berdasarkan umur menunjukkan nilai rataan yang lebih besar pada anjing lokal umur ≥1 tahun dan konsentrasi leukosit menunjukkan nilai statistik yang signifikan berbeda (p<0.05). Berdasarkan jenis kelamin, nilai rataan parameter hematologi secara umum lebih tinggi pada anjing lokal betina dan adanya perbedaan signifikan (p<0.05) pada konsentrasi leukosit dan granulosit.

Abstract

Viral nervous necrosis (VNN) is a disease that often infects groupers. It has caused mass death in more than 34 species of marine fish. DNA vaccination might become a solution againts the infection. The construction of the pmBA-CP DNA vaccine consisting of the beta-actin promoter of medaka fish (Oryzias latipes) and capsid protein (CP) encoding VNN RNA2 has been made in previous studies. This study aimed to test the efficacy of the pmBA-CP DNA vaccine for VNN. The experiment consisted of two stages, namely (1) detection of anti-VNN antibody induction in vaccinated fish using ELISA, and (2) challenge test for fish vaccinated with the VNN virus. Grouper (body length 8 cm to 10 cm) were divided into two groups with a density of 5 fish 60 L–1. The fish in the first group were vaccinated with pmBA-CP intramuscularly at a dose of 12.5 µg per fish, while the second group of fish were not vaccinated. Antibody titer testing was carried out before treatment, and 1 d, 7 d, 14 d, 21 d, 28 d, and 35 d after vaccination. The challenge test was carried out on the 60th day after vaccination. The results showed that the S / P ratio in the vaccinated fish serum was higher than unvaccinated fish at 21 d to 35 d post-vaccination. DNA vaccination was able to induce anti-VNN antibodies of grouper. The results of the challenge test for vaccinated fish using VNN virus titer 103.5 FID50/0.2 mL showed 60% of survival rate. Thus, the pmBA-CP DNA vaccine could be useful for increasing grouper immunity, and support production of grouper.

Abstract

The Thy1 gene is a member of the immunoglobulin superfamily, which is found in many different animals. This gene encodes a Thy1 protein (CD90) which measures 25–37 kDa. Although the function of the Thy1 gene is unknown, it is thought to play a crucial role in the immunological response, particularly in the activation of T cells. The goal of this study was to find a variation in the Thy1 gene in IPB D2  chickens that might be utilized as candidates for immunoresistance genetic markers. A total of 123 samples from IPB D2 chickens consisting of 38 samples from IPB D2 G0 (basic generation) chickens and 85 samples from IPB D2 G2 (second generation) chickens were used in this study. PCR methods and direct-DNA sequencing were used in this study. Data analysis in this study consisted of genotype frequency, allele frequency, observed heterozigosity, expected heterozygosity, and Hardy-Weinberg equilibrium. The results of the study  discovered two SNPs on the intron (g.2139 CT and g.2542 AG). Four haplotypes were created by combining two SNPs. The g.2139 CTand g.2542 AG were polymorphic, but not in the Hardy-Weinberg equilibrium.

Abstract

Studies on the role of domestic animals in the transmission of schistosomiasis japonica in the West Lore Sub-district, Poso District, are still limited despite its importance as zoonosis. This study used a cross-sectional design to determine schistosomiasis prevalence in domestic mammals and identify the relative contribution of each mammalian species’ schistosomiasis transmission in the West Lore Sub-District. Fecal samples were obtained from 209 animals (seven buffaloes, 70 dogs, 44 cattle, 86 pigs, and two horses). The Danish Bilharziasis Laboratory technique was used to detect both the occurrence of S. japonicum egg in feces and the intensity of schistosomiasis infection. The examination of 1852 human fecal samples using the Kato-Katz method was carried out by the Laboratory of Schistosomiasis Lengkeka. The measurement of environmental pollution with S. japonicum eggs and the relative contribution of each species in the transmission was performed. The highest prevalence of S. japonicum infection in animals was in horses (100%; 2/2), cattle (54.55%; 24/44), and pigs (51.16%; 44/86). The prevalence in buffaloes and dogs was 28.57% (2/7) and 32.86% (23/70). Cattle (69.74%) were the main contributors to S. japonicum eggs contamination in the environment, followed by pigs (21.95%) and buffaloes (4.71%). This study reported a high prevalence of schistosomiasis in animals (45.46%), while low human schistosomiasis prevalence (0.59%).

Abstract

The Mx gene is an antiviral gene used to determine the resistance or the susceptibility to different types of viruses, including the Avian Influenza (AI) virus subtype H5N1. The AI virus subtype H5N1 infection in chickens causes Mx gene polymorphism. The Mx+ gene shows resistant to the AI virus subtype H5N1, whereas the Mxgene shows signs of susceptible. The objective of this research was to detect the Mx gene in wild aquatic birds using the Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) method with the primer pairs F2 and NE-R2/R and the RsaI restriction enzyme. DNA samples were obtained from eight species of wild waterbirds with positive and negative exposure to the AI virus subtype H5N1. DNA amplification results showed that the Mx gene in wild aquatic birds is found in a 100 bp fragment, which is the same as the Mx gene found in chickens. However, unlike chickens, the Mx gene in wild aquatic birds did not show any polymorphism. This study proves that Mx- based resistance to AI virus subtype H5N1 in different in wild birds than in chickens.

Abstract

Further detailed research is required to obtain deeper information on the role of wild birds on the distribution of Avian influenza in Asia. A research has been carried out on February–June 2007 focused on blood sampling (serosurveillance) of wild birds in Pulau Dua Nature Reserves (CAPD), Serang, Banten. The research is aimed to investigate the infection of AI virus sub-tye H5N1 on the studied wild birds. The blood samples were taken from studied aquatic birds, followed by HI (haemagglutination-inhibition) test. A total of 183 samples represent 7 water bird species were taken i.e Cattle egret Bubulcus ibis, Javan pond-heron Ardeola speciosa, Little egret Egretta garzetta, Intermediate egret Egretta intermedia, Black-crowned night heron Nycticorax nycticorax, Great egret Casmerodius albus and Grey heron Ardea cinerea. The result revealed that 41 (23.27%) samples showed the present of AIV antibodies serotype H5N1 which is identified as positive. Data showed 5 positive-test species, including B. ibis (29.27%), E. garzetta (29.27%), E. intermedia (4.88%), Ardeola speciosa (7.32%), and N. nycticorax (29.27%). A total of 41.46% were infected adult individual, whereas 58.54% were juveniles.
