Abstract

The aim of this study was to evaluate the effect of different IgY concentrations on the production, reproduction and immunity of Sentul chicken. This research used 20 month-old Sentul chickens which consisting of 10 chickens with high IgY concentration and 10 chickens with low IgY concentration Parameters observed included prelayer performance (feed consumption, weight gain, feed conversion, morbidity and mortality), layer performance (feed consumption, egg production, egg weight, feed conversion, hatchability, morbidity and mortality), and immunity at chicks (IgY concentration of eggs and chicks serum). The experiment was conducted as a completely randomized design. The treatments were high and low IgY concentration. All treatments were repeated 10 times. The data obtained were analyzed using the SAS program t-test. Morbidity and mortality were analyzed descriptively. The results of this research indicated that the prelayer period, chickens with high IgY concentration had a higher performance than chicken with low IgY concentration. This is caused by about 30% chickens with low IgY were in sick condition for approximately seven days. Further, the layer period showed that the chickens with high IgY concentration had lower production performance than the chicken with low IgY concentration. The study concluded that chickens at low IgY concentrations are susceptible to exposure to antigens that lead to decreased productivity. In normal conditions hens, Sentul with high IgY concentration resulted in low production. The observation of chicken immunity showed that concentration of IgY from chicks and eggs did not differ between hens with high IgY concentrations and hens with low IgY concentrations.

Abstract

Escherichia coli (E. coli) K-99 infection cause acute diarrhea in calves which may reduce electrolyte, such as sodium and potassium. The experiment was conducted to study the sodium and potassium profiles in calves infected by E. coli and received immunoglobulin G anti E. coli K99 microcapsules. Sixteen calves, 4 – 6 days years old, were grouped into four groups, consists of four calves, i.e. negative control/NC group (the calves were not infected by E. coli K-99 and no treatments); positive control/PC group (the calves were infected by E. coli K-99 and no treatments); treatment 1/P1 group (the calves were infected by E. coli K-99 and received colostrum suspension of IgG anti E. coli K-99); and treatment 2/P2 group (the calves were infected by E. coli K-99 and received microcapsules of IgG anti E. coli K-99). Escherichia coli K-99 were infected to all calves except the NC group orally with the dosage of 5 × 1010 cfu/ml. Suspension and microcapsules of IgG anti E. coli K-99 for each calf were given orally with dosage of 1.76 gram IgG anti E. coli K-99 per day. Serum were collected from jugular vein at 0, 24, 48, 72, and 168 hours after E. coli K99 infection. The sodium and potassium concentrastion were analyzed using a spectrophotometer. Results of the experiment indicated that the concentration of sodium and potassium were not significantly different among groups. Sodium concentration of P2 group was relatively higher than PC or P1 and showed increased concentration 168 hours after infection. Potassium concentration of P2 were higher than the PC group, but lower than NC and P1 group. In conclusion, the administration of IgG anti E. coli K-99 microcapsules was not significantly change the sodium and potassium concentration, but may maintain the stability of sodium level in calves infected by E. coli K-99.

Abstract

Immunoglobulin Y (IgY) in yolk has been shown in several studies to prevent both bacterial and viralinfections. This research was conducted to find evidence that IgY specific against avian influenza virus(AIV) of H5N1 subtype can be produced in a large quantity in egg yolk. Laying hens were vaccinated withAI killed-vaccine (IPB-Shigeta). The IgY was purified using affinity chromatograpy technique, and anti-H5activity was measured using a standard haemaglutination inhibition (HI) and agar gel immunodifusion.The concentration of IgY was calculated, and the protein pattern was detected using polyacrilamid gel(AGID) electrophoresis (PAGE). Anti H5 antibody as high as 27 – 29 HI units was detected and produce aspecific line of precipitation in AGID. The concentration of IgY was 7.89 mg/ml. Purified specific IgY consistof 6 main protein bands with molecular weights ranging from 35 to 225 kD. These proteins were sensitiveto heat treatment (75oC for 30 minutes), to acid condition (pH2) as well as the pepsin and trypsin. Theseresults indicated the possibility of using specific IgY for passive immunisation to prevent AIV infection oras immunotherapeutic applications for AI treatment in humans.

Abstract

Kerbau lumpur merupakan ternak multiguna yang memiliki kedekatan dengan kehidupan masyarakat di kawasan lumbung ternak khususnya di Kabupaten Brebes, Jawa Tengah. Infeksi parasit saluran pencernaan pada kerbau lumpur bedampak negatif terhadap kesehatan ternak sekaligus menyebabkan kerugian ekonomi. Data epidemiologi infeksi saluran pencernaan pada kerbau lumpur di Kabupaten Brebes belum tersedia. Penelitian ini bertujuan untuk mengukur prevalensi, derajat infeksi serta analisis faktor risiko infeksi parasit saluran pencernaan pada kerbau lumpur di Kabupaten Brebes. Sebanyak 240 sampel tinja diperoleh dari kerbau di empat kecamatan, yaitu; Salem, Bantarkawung, Tonjong dan Brebes. Hasil pemeriksaan mikroskopik dengan teknik Mc Master menunjukkan bahwa 27.5% kerbau terinfeksi oleh parasit saluran pencernaan. Infeksi disebabkan oleh Toxocara vitulorum (1.67%; telur tiap gram tinja [TTGT] 174.4), cacing Strongyle (Cooperia, Nematodirrus, Trichostrongylus) (4.17%; TTGT 140.18), Trichuris spp. (3.33%; TTGT 64.84), Moniezia expansa (9.17%; TTGT 122.47) dan Eimeria spp (21.25%; TTGT 162.37). Infeksi lebih tinggi tercatat pada kelompok jantan, umur muda (>12–30 bulan), kerbau di Kecamatan Salem dan yang dipelihara pada daerah bertipe iklim D4. Variabel umur merupakan satu-satunya faktor risiko infeksi yang ditunjukkan dengan ­P-value 0.03. Nilai odd ratio kelompok muda dan pedet adalah 3.424 dan 2.529, artinya, kelompok muda dan pedet memiliki risiko 3.424 dan 2.529 lebih tinggi dibandingkan kerbau dewasa (referensi). Dari penelitian ini dapat disimpulkan, baik prevalensi maupun derajat infeksi parasit saluran pencernaan di Kabupaten Brebes terjadi dalam taraf yang rendah dan dipengaruhi oleh faktor umur ternak.

Abstract

Immunoglobulin G (IgG) from colostrum of hyperimmune colostrum obtained from vaccinated cattle with H5N1 vaccine is potential to be developed into passive immunotherapy product for “bird flu” infection. The administration of passive therapy can be done orally, but the environmental conditions of the digestive tract can cause IgG damage, so the microencapsulation technique was used in this study to protect IgG. This study aimed to evaluate the absobrption of H5N1 IgG anti-Avian Influenza which was microencapsulated using rats (Rattus novergicus) as animals model. This study was conducted using 108 adult male white rats (Rattus norvegicus), Wistar strain, approx. 250 grams of body weight. Rats were divided into three treatment groups (N = 36), i.e. control group (K) were given aqua distilled water, suspension group (S) were given anti-H5N1 IgG suspension, and microcapsules group (M) were given microcapsules equivalent to suspension with the titer of 25. Necropsy was carried out at four observation points (2, 4, 6 and 8 hours) to collected blood through the heart and collected in vacutainer without anticoagulants to obtain serum. The blood serum then analyzed for total protein and globulin and albumin/globulin ratio (A/ G). The results showed that the level of total protein and globulin group M was higher and significantly different (P> 0.05) compared to K and S at the end point of observation. The A/G ratio in group K showed a significant increase (P> 0.05) at the end point of observation while in group M there was a marked decrease (P> 0.05) at the time point of 2 hours to 4 hours and then constant until the end of observation. These results showed that administration of anti-H5N1 IgG microcapsules can increase total levels of protein and globulin in the blood.

Abstract

Faculty of Animal Science, IPB University has developed new trait of chicken named IPB D1 traits. The chicken is a crossbreed chicken between male F1 PS (Pelung x Sentul) with F1 female (Kampung x parent stock Cobb). The IPB D1 chickens aims to produce the local commercial meat type, which adapted to traditional or semi intensive management (back yard). Salmonellosis is one of the bacterial disease that commonly infected poultry, therefore a study was conducted to determine the immune response of IPB D1 chickens to these disease using genetic markers, namely TLR4 gene (Toll-Like Receptor 4). TLR4 gene is one of the genes that control chicken resistance to Gram negative bacteria infection, through non-specific immune responses, one of Gram negative bacteria is Salmonella enteritidis. The aim of this study was to determine of TLR4 gene and their role in immunity of IPB D1 chickens against S. enteritidis infection. As much as 11 chickens were use in this study. This research was carried out through several stages of i.e determination of the TLR4 gene by PCR and sequencing and observing its immune response through the number of leukocyte, leukocyte differentiation and other hematology profile. Humoral immunity response against S. enteritidis were observe by clearance test before and after challenged with S. enteritidis. The results of this study indicate that IPB D1 chickens which have TLR4 genes (genotype GG and AG) were resistance against S. enteritidis based on the leukocyte, hematology profile and humoral immunity.

Abstract

Rabies is an infectious disease, zoonotic, caused by virus from the genus Lyssa virus and generally transmitted by the bite of rabid animal, especially rabies infected dog. Rabies is preventable but is always fatal to humans if the central nervous system (CNS) is infected. Vaccination has been used as one of rabies prevention programmed. A total of 83 samples were tested using an Indirect ELISA (Enzyme Linked Immunosorbent Assay) to identify post-vaccination rabies antibody titer. Antibody titres correlated with absorbance values and standard solutions concentrations. Absorbance value can be determined using ELISA reader and mobile phone camera. Absorbance were read at 450 nm and 620 nm as reference using ELISA reader and image from mobile phone camera using image processing software (ImageJ). The aim of this study is to compared between ELISA reader as gold standard and mobile phone camera through validity testing such sensitivity, specificity, and accuracy. There is no significant difference between gold standard and alternative test equipment. The mobile phone camera has sensitivity 98,6%, specificity 88.8 % and accuracy 97,5%. The image processing method using ELISA reader is relatively expensive and difficult to hold in laboratory with minimum funds. Image processing method using a mobile phone camera with ImageJ application is expected to be an alternative tool to read the result of ELISA.

Abstract

Newcastle disease (ND) or Tetelo is an endemic poultry disease in Indonesia, and caused significant economic losses. Instead of diseases control programme has been carried out, ND outbreaks still occurs among poultry flock. IPB D1 chicken is a type of cross-breeding chickens between pelung-sentul crossbred chicken male and kampung-broiler crossbred chicken female, this chicken is genetically selected based on on body resistance to disease. The aim of our research is to study immune response of IPB D1 against ND infection. Fourty IPB D1 chickens were used in this study, and divided into three groups : vaccinated group, unvaccinated group, control group. Vaccinated group received two time ND vaccination at age 10 weeks and booster at age 12 weeks. Unvaccinated and control group were remain not vaccinated. At age 13 weeks, vaccinated and unvaccinated group were challenge with ND virus (107 EID50/ml). Immune response were determined based on antibody titer which is measured by haemagglutination inhibition (HI) test and the profile of white blood cell of the chicken. Our result showed that both vaccinated and unvaccinated group has ND antibody titer e” log 24 at challenge time, and survived after challenge. White blood cell profile among all groups showed that lymphocites has the higher number than other leukocyte type. It can be concluded that IPB D1 chickens showed a good response against ND virus.

Abstract

Paramphistomum spp. and Fasciola spp. are the most common trematodes that infect cattle throughout the world including in Manokwari, West Papua. During their life cycle, freshwater snails act as the intermediate host for the larval. This study aims to provide information about the presence of cercariae –the larval form of trematode- within the body of Lymnaea spp. snail. The study was conducted in August-September 2019 in Prafi District, Manokwari, West Papua. Lymnaea spp. were collected from 15 permanent and seasonal drainage around a water dam, rice field irrigation, and palm oil plantations. Snails are collected by hand using a quadrant or using the "man per minute" method. Cercaria within the snail's body is examined using the "crushing method" under a light microscope. Snails that have cercaria trematodes are categorized as infected snails. The percentage of snails infected with trematodes in August and September were 15.92% (53 out of 333 snails) and 38.05% (78 out of 205 snails), respectively. Cercariae that were identified were longifurcate-pharyngeate cercariae (Strigea cercaria), Cercariaeum cercaria, and Virgulate cercaria. The adult phase of these cercaria are generally exist in the intestine of mammals, amphibians, birds and fish. Further research is needed to provide more accurate information about the epidemiology and morphology of trematodes in freshwater snails.

Abstract

ABSTRACT Cockroaches are insects that act as mechanical vectors of pathogenic agents to humans such as Salmonella sp. The aim of this study was to identify the presence of Salmonella sp. in cockroach bodies on cargo and passenger ships. This research was conducted at Baubau Port from October to December 2017. A total of 24 ships were examined consisting of 12 passenger ships and 12 cargo ships. Cockroaches were collected from rooms inside each ship namely galley, bridge, deck and bathroom. A total of 3 196 cockroaches caught consisting of Periplaneta americana (69.50%), Blattella germanica (29.60%), Periplaneta brunnea (0.66%), Pseudophorapsis sp. (0.03%), Pycnoscelus surinamensis (0.03%), Periplaneta australasiae (0.03%), and Neuphoeta cinerea (0.03%). The presence of Salmonella sp. were examined for cockroach species with a large number of population, namely Periplaneta americana and Blattella germanica. A total of 95.3% of the 42 cockroaches examined in the laboratory contained Salmonella sp.. Based on this research it can be concluded that the cockroaches collected from the ships leaning at Baubau Port can serve as a vector of salmonellosis for both ship crew and passengers. Keywords: Cockroaches, Ships, Salmonella sp, Salmonellosis
