Abstract

Sociocultural aspect is considered as important consideration in pre-service science teachers’ educational program. The objective of the study was to improve the ability of pre-service science teachers in reconstructing local community indigenous knowledge. Subjects were 87 pre-service science teachers from three universities in Central and East Java with Dieng Plateau’ community indigenous knowledge in breastfeeding as knowledge that they have to reconstruct. Results suggested that pre-service science teachers were able to reconstruct the indigenous knowledge well. Implication to pre-service science teacher’s educational program is discussed.

Abstract

Karakterisasi Biokimia Enzim Fitase Rekombinan (phyK) dari Klebsiella sp. ASR1 Yang Dienkapsulasi Dengan AlginatEnzim fitase melepas molekul fosfor pada atom C dari benzena Inositol fitat. Tetapi fitase memiliki kelemahan tidak mampu bertahan terhadap kondisi ekstrim dalam lambung nonruminansia. Solusi dalam penelitian ini yaitu fitase dienkapsulasi menggunakan alginat. Penelitian ini bertujuan mengkarakterisasi fitase setelah dienkapsulasi menggunakan alginate. Hasil penelitian ini yaitu fitase yang dienkapsulasi memiliki aktivitas tertinggi pada pH 6,0, sedangkan fitase tanpa enkapsulasi pada pH 5,0. Suhu optimum untuk aktivitas tertinggi fitase yang dienkapsulasi yaitu 70ºC, sedangkan fitase tanpa enkapsulasi 37ºC. Untuk perlakuan penambahan ion logam, aktivitas tertinggi fitase yang dienkapsulasi terjadi dengan penambahan 0,1 mM Fe2+ dan 1,0 mM Ca2+, sedangkan fitase tanpa enkapsulasi dengan penambahan 0,1 mM Fe2+. Berdasarkan hasil penelitian ini, fitase yang dienkapsulasi memiliki keunggulan lebih banyak dibandingkan dengan fitase tanpa enkapsulasi, karena mampu bertahan pada pH dan suhu tinggi, dan beberapa efek ion logam.Kata Kunci: alginat, asam fitat, enkapsulasi, fitase, fitase rekombinanABSTRACTPhytase enzymes release phosphorus molecules on the C atom from benzene inositol phytate. This study aims to characterize phytase after being encapsulated using alginate. The results of this study were the encapsulated phytase had the highest activity at pH 6.0, while the unencapsulated phytase at pH 5.0. The optimum temperature for the highest activity of the encapsulated phytase was 70ºC, while the unencapsulated phytase 37ºC. For treatment of metal ion addition, the highest activity of the encapsulated phytase occurred with the addition of 0.1 mM Fe2+ and 1.0 mM Ca2+, while the unencapsulated phytase with the addition of 0.1 mM Fe2+. Based on the results of this study, the encapsulated phytase had more advantages compared to the unencapsulated phytase, as the former withstand high pH and temperature, and some metal ion effects.

Abstract

This research was aimed at producing a crude intracellular phytase characterized from recombinantbacteria. The recombinant bacteria (pEAS1AMP) was produced by way of transforming pET-22b(+)+pEAS1 into competent E. coli BL21 and E. coli BL21(DE3) cells. Crude intracellular phytaseproduction was induced using 1,5 mM Isopropyl-β-D-thiogalactopyranosid (IPTG). Recombinantbacteria product and enzyme activity test followed the Sajidan method. E. coli BL21(+)pEAS1 and E.coli BL21 (DE3)(+)pEAS1 recombinant bacteria showed growth after 20 hours and 10 hours oftransformation. Phytase activity of E. coli BL21 (DE3)(+)+pEAS1 showed higher than those of E. coliBL21(+)+pEAS1. Crude intracellular phytase of pEAS1AMP recombinant bacteria has an optimumactivity at pH 5, 40oC, incubation period of 60 minutes, substrate concentration of 2%, molecular weight(MW) of 47.3 kDa, Km = 15.91 υM and Vm = 2.41 υM/second. Mg2+ acts as a cofactor but Fe3+ (10-4M) acts as an inhibitor.

Abstract

Penelitian ditujukan untuk memberikan pelatihan dan pengembangan usaha masyarakat non produktif melalui UMKM berbasis bahan dasar bekatul. Kegiatan pengabdian yaitu pemberian materi tentang kandungan bekatul, cara pengolahan bekatul sebagai pakan ternak berkualitas dan makanan bergizi, pengemasan dan pemasaran produk. Jumlah peserta yang diikutsertakan dalam program ini yakni sebanyak 35 masyarakat dengan teknik pengumpulan data penelitian dengan cara: observasi potensi, wawancara, analisis data sekunder, dan dokumentasi. Metode kegiatan pengabdian dilakukan dengan cara pemaparan materi oleh ahli, fact finding (analisis kebutuhan), discussion (diskusi antara pemateri), asking and answering (tanya jawab dan diskusi), dan recitation (resitasi atau pembagian tugas), practice and experiment (praktik dan eksperimen). Hasil kegiatan ini berupa temuan bahwa saat ini masyarakat memiliki pemahaman pengolahan bekatul terbatas pada produk olahan untuk pakan ternak. Kegiatan diskusi memberikan paradigma baru terhadap masyarakat untuk mengolah berbagai macam produk makanan berbahan bekatul. Pelatihan ini memberikan bekal kepada masyarakat untuk menganalisis produk berbahan bekatul untuk dikembangkan menjadi produk olahan makanan sehingga memiliki nilai jual yang lebih tinggi. Kegiatan pengabdian membekali masyarakat untuk memiliki pengetahuan dasar pengolahan makanan hingga ke tahap pemasaran sehingga diharapkan dapat meningkatkan perekonomian masyarakat.

Abstract

ABSTRAKPenelitian ini dilakukan dengan tujuan untuk mengevaluasi kemampuan menulis argumentatif calon guru biologi meningkat setelah mereka mendapatkan pembelajaran inkuiri berjenjang. Partisipan dalam penelitian tindakan kelas ini adalah mahasiswa semester 4 di Program Studi Pendidikan Biologi Fakultas Keguruan dan Ilmu Pendidikan Universitas Sebelas Maret Surakarta yang menempuh mata kuliah Embriologi Tumbuhan. Data dianalisis secara deskriptif melalui wawancara, pengamatan kelas dan portofolio. Kemampuan menulis diukur menggunakan Toulmin’s Argument Pattern (TAP) yang meliputi empat komponen, yaitu data, warrants, backings, dan claims. Hasil penelitian menunjukkan bahwa pada akhir siklus kedua terjadi peningkatan skor keterampilan berpikir argumentatif pada setiap aspeknya di semua tingkatan jenjang inkuiri. Calon guru biologi yang telah berpengalaman dalam inkuiri berjenjang menunjukkan kemampuan menulis argumentasi yang lebih baik sehingga disimpulkan bahwa pembelajaran dengan inkuiri berjenjang sesuai diterapkan untuk mendukung berbagai aktivitas inkuiri dan meningkatkan kemampuan menulis argumentatif. ABSTRACTThe purpose of this study was to evaluate biology teacher candidates’s argumentative writing skill after they participated in hierarchy of inquiry learning. The participants of this action research are the 4th semester students of Biology Teacher Education Program Faculty of Teacher Training and Education Sebelas Maret University who took the course of Plant Embryology. Data were analyzed descriptively through interviews with experts, classroom observations, and student portfolios. The argumentative writing was measured using Toulmin’s Argument Pattern (TAP) with four kinds of argument elements, which include data, warrants, backings, and claims. The results showed that the argumentative writing skill was increased, in every aspect and levels of inquiry. Biology teacher candidates who had well-experienced in the hierarchy of inquiry outperformed argumentative writing skill, thus it can be concluded that Hierarchy of Inquiry Learning is suitable in fostering teacher candidates to do some inquiry activities, and positively impacted argumentative writing skill.

Abstract

This study aims to design a Scientific Reading-based Inquiry (SRbI) model that supports argumentation skills development. The assessment of these skills refers to the Toulmin Argument Pattern (TAP), and the participants were Biology Education students in a state university. Furthermore, the Design-based Research (DBR) approach was adopted by combining exploratory studies, trials, and case studies as part of an iterative process. The intervention was formed based on design principles derived from literature review and findings from exploratory studies. Also, observations were made during the trial and intervention process. Data in assessments and observations of written and oral arguments were collected and descriptively analyzed. The study, in three iterations, produced a framework as the basis for the SRbI learning model, with five phases: Reading Orientation, Recapturing, Processing, Communicating, and Reviewing. Therefore, the application of this learning model had a significant impact on the development of students’ argumentation skills.

Abstract

This study aims to describe the perceptions of SD Angkasa Colomadu teachers on online science learning during the pandemic. This research used descriptive qualitative research methods and a case study approach. The subjects of this study were three teachers of grades IV, V, and VI SD Angkasa Colomadu in the academic year 2020/2021. Data collection techniques used interviews and documentation. The data validity test techniques used the triangulation technique. Data analysis techniques used interactive analysis techniques Miles and Huberman consisting of data reduction, data display, and conclusions. The results show that the teachers felt that learning sciences in the network was not optimal. Teachers do not optimally apply basic process skills. The teachers feel the limitation in the communication process.  They also have a restriction on directing the students to formulate hypotheses, conclude the material, guide experimental activities, attract students to ask questions, guide the measurement process, guide observation activities, classify, interpret, and predict processes due to media limitations. On the other hand, teachers feel that online learning makes teachers more innovative, creative, and able to take advantage of cellphones, computers, and the internet in learning activities, especially in learning science online during the pandemic period.

Abstract

Abstract: Although classroom action research (CAR) is considered useful for teacher development but this activity has not been implanted yet effectively in Indonesia. In this paper, we report the alternative way to overcome this problem. It addresses how blended learning support self-directed learning and enhance teacher competency research. Through questionnaire, interview and document analysis, the result showed that blended learning-based self-learning can improve teachers performance in conducting classroom action research (CAR) and enhance their ability in compiling the research report.

Abstract

Based on the need’s analysis done, the empowerment of higher-order cognitive skills of the students in Surakarta was not optimal yet. This research addressed to determine and describe the feasibility of biology module which was arranged based on Stimulating Higher-Order Thinking Skills (Stim-HOTS) model. This Research and Development (R&D) involved six expert validators, including material validator, module development validator, language validator, learning instrument validator, and school practitioners. The results showed that the percentages of the module feasibility were between 83% and 98% which were categorized as very good. Based on the results of the analysis, it can be implied that the Stim-HOTS-based module can be used as learning resource.
