Abstract

AbstrakAvicelase dan CMCase mewakili aktivitas eksoselulase dan endoselulase, termasuk dua dari tiga jenis selulase dalam sistem selulase yang dihasilkan oleh Bacillus circulans. Ekspresi keduanya sangat dipengaruhi oleh jumlah dan jenis sumber karbon dalam media produksi. Penelitian ini bertujuan mengetahui jenis dan konsentrasi limbah pertanian terbaik untuk menghasilkan avicelase dan CMCase dari B. circulans serta kemampuan sistem selulase tersebut untuk mendegradasi substrat lignoselulosa. Limbah pertanian yang diteliti berupa ampas tebu, bonggol jagung dan sekam. Limbah pertanian tersebut digunakan sebagai satu-satunya sumber karbon pada media produksi. Avicel digunakan sebagai sumber karbon pembanding. Aktivitas avicelase dan CMCase ditentukan berdasar jumlah gula pereduksi yang dihasilkan dengan menggunakan metoda Somogy-Nelson. Hasil penelitian menunjukkan bahwa semua limbah pertanian yang diteliti berpotensi sebagai sumber karbon yang lebih baik dibandingkan dengan avicel untuk produksi avicelase dan CMCase dari B. circulans. Limbah pertanian yang terbaik adalah ampas tebu. Penggunaan ampas tebu pada konsentrasi 0,5 % menghasilkan aviselase dan CMCase sebesar 309,39 dan 405,48 U/mL. Sistem selulase yang dihasilkan mampu mendegradasi ampas tebu, bonggol jagung dan sekam.Kata kunci : avicelase, CMCase, limbah pertanian, sumber kabon, Bacillus circulansAbstractAvicelase and CMCase activity represent exocellulase and endocellulase, including two of the three types of cellulase in the cellulase system produced by Bacillus circulans. Expression both of them strongly influenced by the amount and type of carbon source in the production medium. The aims of this research were to determine the type and concentration of the best agricultural waste to generate avicelase and CMCase from B. circulans and their ability to degrade lignocellulosic substrates. Studied agricultural wastes are bagasse, corn stalks and husks. Agricultural waste is used as the sole carbon source in media production. Avicel is used as a comparable carbon source. Avicel and CMCase activity are determined based on the amount of the reducing sugars is generated by using the method of Somogy - Nelson. The results showed that all the studied agricultural waste was a potential carbon source better than ethical for avicelase and CMCase production from B. circulans. Agricultural waste bagasse was the best one. The use of 0.5 % bagasse produce CMCase and aviselase at 309.39 and 405.48 U / mL respectively. Abstract

The production of ligninase by wood rot fungus (WRF) is determined by carbon source and growth condition. The goal of this study is to determine the ligninase profile produced by WRF KLUM2 in Kirk Medium using teak wood alkaline lignin as a carbon source known as Kirk Medium-Alkali lignin Kayu Jati (MK-ALKJ), optimization of dominant ligninase production in the MK-ALKJ compared to the one that is produced in the Kirk’s medium with glucose as a carbon source (MK-Glucose). This research was conducted in an experimental laboratory consisting of: (1) spore suspension preparation, (2) ligninase profiling at various growth times, (3) ligninase profiling at various temperature variations, (4) optimization of laccase production including pH and the amount of nitrogen source. Growth was identified based on the specific activity of lignin peroxidase (LiP), manganese peroxidase (MnP), and laccase. The results showed that relatively the three types of ligninase, namely LiP, MnP, and laccase, were produced in the same amount by the wood rotting fungus isolates KLUM2 in MK-ALJK. All three were produced with the highest yield of respectively 55.65; 52.48; 57.64 U/mg. Laccase as the dominant ligninase can be optimized to reach 83.52 U/mg by inoculating 2.107 spore cells in MK-ALKJ in 37 °C, pH = 3.5, and a nitrogen source of 20mM (NH4)2SO4 for 6 days. Therefore, it can be concluded that the ligninase activity of indigenous WRF KLUM2 in MK-ALJK medium is higher than in the MK-Glucose.

Abstract

Soil bacteria from tropical environments play a significant role in resolving various environmental issues, including biodegradation. Exploratory research on biodiversity is crucial to develop and harness the potential of different types of soil bacteria that are highly abundant. The bacterial diversity in landfills is typically high due to the decomposition of organic and inorganic waste, creating a favorable medium for the growth and development of soil bacteria. This study aims to assess the candidates of xenobiotic degrading indigenous bacteria from the Probolinggo City landfill using Next Generation Sequencing (NGS) method. The research stages include: 1) sampling, 2) isolation of genomic DNA from samples using the ZymoBIOMICS DNA MiniPrep Kit from Zymo Research, 3) amplification of isolated DNA with primers 16S 27F – 1429R, 4) sequencing the results of DNA amplification with NGS, 5) downstream analysis of the results using software Pavian Krona Tools, and 6) narrative analysis review to identify the candidates of xenobiotic degrading indigenous bacteria. The results show that soil samples from the Probolinggo City landfill exhibited a high diversity of bacterial communities. Based on NGS analysis, 2400 bacterial species were identified, comprising 56 genera, 17 orders, 4 classes, and 4 phyla, with respective abundances of Proteobacteria (70%), Firmicutes (15%), Planctomycetes (2%), and Cyanobacteria (0,3%). Based on the narrative analysis review, several bacteria in the Probolinggo City landfill exhibited potential as: 1) polypropylene-degrading bacteria, including Bacillus cereus, B. licheniformis and B. thuringiensis. 2) styrofoam degrading bacteria, namely Bacillus amyloliquefaciens, Bacillus cereus, Bacillus firmus and Pseudomonas aeruginosa. 3) total ammonia nitrogen (TAN) reducing bacteria, including Bacillus megaterium. 4) pesticide degrading bacteria Profenofos and Chlorantraniliprole, including Bacillus stearothermophilus. and 5) tannic acid degrading bacteria, including Pantoea dispersa. These results indicate that the Probolinggo City landfill is a good habitat for various xenobiotic-degrading bacteria, then the isolation of specific bacteria can be designed using an appropriate selective medium.

Abstract

This research aimed to ascertain the impact of alkaline lignin of teak sawdust on the synthesis of ligninase and decolorization of Reactive Black 5 (RB5) by M. thermophilia KLUM1, also to elucidate the mechanism of mycoremediation. This research was conducted within the cultivation of the spore suspension in a co-culture setting. The RB5 dye percentage decolorization and ligninase activity were produced by varying the addition of alkaline lignin. The results show that the addition of 0.5% alkaline lignin accelerates the production of ligninase in the Kirk medium. However, adding more than 0.5% alkaline lignin does not affect the highest amount of ligninase produced. The increase in RB5 dye decolorization by M. thermophila KLUM1 is proportional to the increase in alkaline lignin in the medium. The percentage of wood-rot fungi decolorization in Kirk medium with and without lignin with the addition of 0.5%, 1%, and 2% lignin on the 10th day was 26.29%, 29.51%, 34.74%, and 40.43%, respectively. The decolorization process of RB5 by M. thermophilia KLUM1 in Kirk medium with alkaline lignin occurs through several mechanisms, i.e., enzymatic degradation, adsorption by the growing mycelia, and alkaline lignin present in the medium.
