Abstract

The specialized epithelial outer layer, including the skin and mucosal surfaces, is relatively resistant to most environmental hazards and resistant to infection by disease-causing microorganisms. This literature review aimed to describe the role of natural physical, mechanical, and biochemical barriers in innate immunity. The physical barrier that protects against damage and infection consists of closely related epithelial cells, including the skin and the sheet membranes that line the digestive tract, genitourinary, and breathing. The epithelial surface also provides a biochemical barrier by synthesizing and secreting substances intended to trap or destroy microorganisms (chemicals derived from the epithelium). Mucus, sweat, saliva, tears, and earwax are examples of biochemical secretions that can trap and kill potential disease-causing microorganisms. Microorganisms in the microbiome do not usually cause disease, although some are opportunistic in that they can cause disease if the integrity of the body surface is compromised or the individual's immune or inflammatory systems are damaged. In conclusion, natural barriers include physical, mechanical, and biochemical on the surface of the body and are present from birth to prevent damage by substances in the environment and prevent infection by pathogenic microorganisms.

Abstract

Each cell has receptors on the cell surface that specifically bind to solutes (ligands) produced during tissue damage or infection. This review aimed to describe the role of PRR in the human body's defense system. The binding of the ligand to its receptor results in the activation of intracellular signaling pathways and cell activation. The B and T lymphocytes of the adaptive immune system have developed surface receptors (that is, the T-cell receptor, or TCR, and the B-cell receptor, or BCR) that bind a broad spectrum of antigens. The cells involved in innate resistance have developed a distinct set of receptors that recognize a much more limited array of specific molecules. These are called pattern recognition receptor (PRR), and they recognize the molecular patterns in infectious agents or their products (pathogen-associated molecular patterns, or PAMP) or products of cellular damage (necrosis or apoptosis; molecular pattern-associated damage, or DAMPs). In conclusion, the pattern recognition receptor (PRR) is a receptor complex that interacts with various molecules, such as PAMP and DAMPs. PRR bonds with these various molecules and play a role in various actions of innate immunity and adaptive immunity.

Abstract

The inflammatory response is the rapid initiation and interplay of humoral (dissolved in the blood) and cellular systems designed to limit the degree of tissue damage, destroy infectious microorganisms, initiate an adaptive immune response, and initiate the healing process. This review aimed to comprehensively describe the role of the inflammatory response in innate immunity. Three key plasma protein systems are essential for an effective inflammatory response. These are the complement system, clotting system, and kinin system. Although each system has a unique role in inflammation, they also share many similarities. Each system consists of several proteins in the blood. To prevent activation in unnecessary situations, each protein is normally in an inactive form. Some proteins are enzymes that circulate in an inactive form as proenzymes. Each system contains several proteins that can be activated at the start of inflammation. Cells of the innate and acquired immune systems are recruited and activated by biochemical mediators produced at sites of cell damage. These molecules originate from destroyed or damaged cells, contaminating microbes, activation of plasma protein systems, or secretion by other cells of the innate or acquired immune system. Activation may result in the cell acquiring a function essential for the inflammatory response or inducing the release of additional cellular products that promote inflammation, or both. In conclusion, inflammatory cells and various protein systems (complement, kinin, and clotting), together with the substances they produce, act at the site of tissue injury to limit the extent of damage, kill microorganisms, and remove debris in preparation for healing: tissue regeneration or repair.

Abstract

The prevalence of broken homes, characterized by parental separation, divorce, or single-parent households, poses significant challenges to adolescents' development and academic outcomes. This research explores the intricate relationship. Through a mixed-methods approach encompassing quantitative surveys and qualitative interviews, the study examines the mechanisms through which family disruptions influence academic functioning among adolescents. Quantitative analyses reveal significant associations between broken home conditions and lower levels of attentional control, task persistence, and academic engagement among adolescents. These findings persist even after controlling for demographic variables such as socioeconomic status and parental involvement, underscoring the independent contribution of family disruptions to academic outcomes. Qualitative insights provide rich contextual understanding of the lived experiences of adolescents in broken homes, highlighting the emotional turmoil, insecurity, and challenges they face in navigating academic demands amidst familial upheaval. The implications of these findings are far-reaching, with significant implications for educational practice, policy, and intervention efforts. By recognizing the unique needs of adolescents from broken homes and developing evidence-based strategies to support their holistic development and academic success, educators, policymakers, and stakeholders can create nurturing and inclusive environments where all adolescents have the opportunity to thrive.

Abstract

Background: The most prevalent joint condition and leading contributor to disability is osteoarthritis (OA), resulting in a significant socio-economic impact. It occurs due to an imbalance of pro-inflammatory and anti-inflammatory mediators, influencing anabolic and catabolic activities. This study aims to determine the effect of activated growth factor (AGF) on alpha-SMA levels. Methods: This research is an in vivo post-test-only control group experimental study conducted at the Eureka Research Laboratory. Thirty male Wistar rats were divided into five treatment groups: normal control group, negative control group, AGF I group (TGF-β 100 pg/mL), AGF II group (TGF-β 1000 pg/mL), and AGF III group (TGF-β 10,000 pg/mL). AGF was obtained from rat blood intravenously and centrifuged at a predetermined speed. Growth factor activation was performed by adding 10% CaCl2, and then TGF-β levels were measured. All groups of rats were acclimatized for 7 days. After that, osteoarthritis was induced with intra-articular injection of monoiodoacetate (MIA) 4.8 mg/60μL in all groups of rats except the normal control group. Next, rats were given treatment according to the group for 21 days. On the 21st day, rats were euthanized, and alpha-SMA levels were measured using the sandwich method ELISA kit. Results: Sequentially, the mean Alpha-SMA levels for each group in pg/mL were: 91.495 ± 2.36; 10.682 ± 1.09; 30.502 ± 2.00; 52.892 ± 1.29; and 76.180 ± 2.65. In the AGF group, there was an increase in alpha-SMA levels directly proportional to the dose of TGF-β injected. Conclusion: AGF has an effect on increasing alpha-SMA levels in the joints of rats with a model of osteoarthritis.

Abstract

This study aims to analyze the da'wah strategy of the Pendakwah Keren Community (KPK) through the slogan "Ikhlas Ruhnya Dakwah" (Sincerity is the Spirit of Da'wah) in South Sulawesi. The focus of this study is on how this community interprets and implements the value of sincerity as the spirit of da'wah in its activities, and how this strategy shapes a positive image in the eyes of the community. This study uses a qualitative approach with a case study method. Data were obtained through in-depth interviews, participatory observation, and documentation of KPK activities in various regions in South Sulawesi. The results show that the KPK implements six main strategic steps in carrying out the slogan "Ikhlas Ruhnya Dakwah": internalization of the value of sincerity through spiritual regeneration, implementation of social da'wah without expecting anything in return, program selection based on the value of sincerity, leadership that exemplifies sincerity, evaluation based on intention rather than outward achievements, and maintenance of sincerity through dhikr and muhasabah. The urgency of developing the KPK's da'wah strategy is driven by socio-cultural changes in society, internal community challenges, the need to strengthen the implementation of the slogan's values, expansion into marginalized areas, and the professionalization of da'wah without losing its spiritual essence. The strengths of the KPK's strategy include an emphasis on spiritual values that distinguish it from commercial da'wah, a non-competitive and inclusive approach, spirituality-based cadre development, and flexibility in da'wah methods. Its weaknesses include the absence of objective measures to assess sincerity, high dependence on individual motivation, and a weak organizational management system, all of which lead to program inconsistency. The slogan "Ikhlas Ruhnya Dakwah" (Sincerity in Preaching) serves as the philosophical foundation guiding all of the KPK's preaching activities, ensuring they remain focused on the ultimate goal of pleasing Allah, without getting caught up in materialism or popularity. This study is expected to contribute to contemporary preaching strategies and serve as a reference for other preaching communities in developing strategies that are relevant, creative, and rooted in the value of sincerity.
