Abstract

Abstract

The basic immunization program is a strategic effort to reduce morbidity and mortality in infants from preventable diseases. This study aimed to analyze the management of the DPT and Polio immunization program for infants in the working areas of Health Centers in South Central Timor Regency, covering aspects of planning, implementation, human resources, and digital reporting systems. The study used a qualitative approach with purposive sampling at four health centers, namely Kota, Taneotop, Lotas, and Siso. Data were collected through in-depth interviews, observation, and document review, then analyzed using the Miles and Huberman model, which includes data reduction, presentation, and conclusion drawing. The results showed that most health centers had conducted vaccine and logistics planning according to standards, but were not yet fully capable of independently managing the cold chain. Immunization was carried out routinely and through mass campaigns, although challenges remained related to electricity, vaccine distribution, and limited storage facilities. In terms of human resources, only some staff had received technical training, while others had not fully developed competencies in logistics management and digital reporting. The implementation of the ASIK and SMILE applications improved the accuracy and efficiency of reporting but was not yet uniformly applied across all areas due to infrastructure and network limitations. In conclusion, the success of the immunization program in South Central Timor Regency heavily relies on data-driven planning, capacity building for staff, and the strengthening of an integrated and sustainable digital system.

Abstract

Alzheimer's disease is a neurodegenerative disorder marked by cognitive decline and memory loss, primarily caused by the aggregation of amyloid-beta and tau proteins in the brain. Conventional treatments offer limited benefits, emphasizing the need for new therapeutic strategies. To design and evaluate short peptides as targeted protein inhibitors to prevent the aggregation of amyloid-beta and tau proteins, aiming to halt or reverse the progression of Alzheimer's disease. The study employed computational modeling to design peptides, followed by in vitro assays for initial screening, and in vivo tests using transgenic mouse models to assess therapeutic efficacy and safety. Techniques included mass spectrometry, HPLC, and behavioral tests for cognitive function. Designed peptides demonstrated high binding affinity and specificity for amyloid-beta and tau proteins, reducing aggregation by 70% in vitro. In vivo studies showed significant reductions in amyloid plaques and tau tangles, with improved cognitive performance in treated mice. Peptides effectively crossed the blood-brain barrier and accumulated in target brain regions. The findings support the potential of short peptides as a novel therapeutic approach for Alzheimer's disease, warranting further research and clinical trials to validate their efficacy and safety in human subjects.

Abstract

Magnetic nanoparticles (MNPs) have emerged as promising materials for biomedical applications, particularly as contrast agents in tumor imaging. Early and accurate tumor detection is critical for improving treatment outcomes, yet current imaging techniques often lack sensitivity and specificity. This study aimed to synthesize and characterize magnetic nanoparticles for their potential as contrast agents in tumor imaging. The nanoparticles were synthesized using a co-precipitation method, followed by surface modification with organic compounds to enhance stability and targeting specificity. Characterization included transmission electron microscopy (TEM), X-ray diffraction (XRD), vibrating sample magnetometer (VSM), and dynamic light scattering (DLS). Cytotoxicity and targeting efficiency were evaluated in vitro using cultured human tumor cells. The results demonstrated that the synthesized nanoparticles had an average size of 25 ± 5 nm, superparamagnetic properties with a saturation magnetization of 55 emu/g, and high colloidal stability due to surface modifications. Fluorescence imaging revealed significant accumulation of the nanoparticles in tumor cells, while cytotoxicity tests showed cell viability above 85% at concentrations up to 100 ?g/mL. These findings indicate the nanoparticles are safe and effective for tumor imaging. This study highlights the importance of integrating synthesis, characterization, and biological evaluation to optimize nanoparticle design for biomedical applications. While the results are promising, further in vivo studies are needed to evaluate nanoparticle distribution, accumulation, and clearance in complex biological systems. The findings provide a foundation for future research and development of advanced contrast agents for tumor imaging

Abstract

The background of this study focuses on diabetes, a chronic metabolic disease characterized by impaired glucose and insulin regulation. Although existing therapies are available, insulin resistance and other complications remain a major challenge. The purpose of this study is to identify new drug targets through the analysis of protein interactions that play a role in the pathogenesis of diabetes. The method used is protein interaction network analysis (PPI) using public databases such as STRING and BioGRID to map the interaction between proteins related to glucose metabolism and insulin. The results of this study identified more than 150 proteins that interact with each other in the regulatory pathways of glucose and insulin metabolism, with several new proteins found to have the potential to be drug targets to overcome insulin resistance. The study concludes that a protein interaction-based approach can open up opportunities to develop new therapies that are more specific and effective in managing diabetes. Further development should be undertaken for the validation of these findings in animal models and clinical trials to confirm their effectiveness as a diabetes therapy.

Abstract

Influenza virus remains a significant global health challenge, causing seasonal epidemics and potential pandemics with high morbidity and mortality rates. This study aims to develop a recombinant vaccine as a safer and more effective alternative to traditional influenza vaccines, which often suffer from limited efficacy and lengthy production timelines. Utilizing a recombinant DNA technology approach, this research employed the baculovirus expression system to produce hemagglutinin (HA) antigens derived from the influenza A virus. Experimental methods included antigen purification, immunogenicity assays in murine models, and neutralizing antibody titration. Results revealed that the recombinant HA vaccine elicited a robust immune response, with a significant increase in hemagglutination inhibition titers compared to control groups. Furthermore, the vaccinated subjects exhibited substantial protection against viral challenge, evidenced by reduced viral load and minimized lung pathology. The findings suggest that recombinant vaccine platforms offer promising avenues for rapid and scalable vaccine development. This study underscores the potential of recombinant influenza vaccines in mitigating future influenza outbreaks with improved safety, efficacy, and production agility.
