Abstract

The production of ligninase by wood rot fungus (WRF) is determined by carbon source and growth condition. The goal of this study is to determine the ligninase profile produced by WRF KLUM2 in Kirk Medium using teak wood alkaline lignin as a carbon source known as Kirk Medium-Alkali lignin Kayu Jati (MK-ALKJ), optimization of dominant ligninase production in the MK-ALKJ compared to the one that is produced in the Kirk’s medium with glucose as a carbon source (MK-Glucose). This research was conducted in an experimental laboratory consisting of: (1) spore suspension preparation, (2) ligninase profiling at various growth times, (3) ligninase profiling at various temperature variations, (4) optimization of laccase production including pH and the amount of nitrogen source. Growth was identified based on the specific activity of lignin peroxidase (LiP), manganese peroxidase (MnP), and laccase. The results showed that relatively the three types of ligninase, namely LiP, MnP, and laccase, were produced in the same amount by the wood rotting fungus isolates KLUM2 in MK-ALJK. All three were produced with the highest yield of respectively 55.65; 52.48; 57.64 U/mg. Laccase as the dominant ligninase can be optimized to reach 83.52 U/mg by inoculating 2.107 spore cells in MK-ALKJ in 37 °C, pH = 3.5, and a nitrogen source of 20mM (NH4)2SO4 for 6 days. Therefore, it can be concluded that the ligninase activity of indigenous WRF KLUM2 in MK-ALJK medium is higher than in the MK-Glucose.

Abstract

AbstrakPenelitian ini bertujuan untuk  mengetahui sumber nitrogen, kadar sumber nitrogen dan pH optimum untuk produksi protease isolat HTcUM6.2.2. Penelitian ini merupakan penelitian eksperimental laboratoris menggunakan Rancangan Acak Lengkap (RAL) satu faktorial masing-masing yaitu jenis, kadar sumber nitrogen, serta pH medium dengan tahap penelitian terdiri dari peremajaan, validasi kemurnian isolat HTcUM6.2.2, produksi ekstrak kasar protease isolat HTcUM6.2.2,  uji aktivitas protease. Hasil penelitian menunjukkan sumber nitrogen optimum untuk menghasilkan protease dari isolat HTcUM6.2.2 adalah susu skim dan limbah cair tahu. Kadar limbah cair tahu optimum untuk produksi protease dari isolat HTcUM6.2.2 sebesar 10 %. Produksi protease cukup tinggi dan  relatif konstan antara pH 6 sampai 8.  Aktivitas protease tertinggi sebesar 0,817 U/ml dicapai dengan penggunaan 10 % limbah cair tahu pada medium produksi, pH = 7 selama 3 hari. Kata kunci: protease, tauco, isolat, nitrogen, limbah cair tahu   AbstractThis study aims to determine the optimum of the nitrogen source, percentage of nitrogen source and pH for the production of protease from isolate HTcUM6.2.2. This research is a laboratory experimental with a research stage comprising inoculation and validation of the purity of the isolates HTcUM6.2.2, production of crude extract of protease to determine source of nitrogen, percentage of nitrogen source and pH optimum, determination of protease activity. The results showed that the optimum source of nitrogen to produce proteases from the HTcUM6.2.2 isolate was skim milk and tofu wastewater. Percentage of nitrogen source optimum to produce protease of  HTcUM6.2.2 isolate was 10 % of tofu wastewater. Protease production is relatively high and constantance at pH 6 to 8. The highest protease activity was achieved by the use of 10 % tofu wastewater at production medium, pH = 7 for 3 days was 0,817 U/mL. The keywords: protease, tauco, isolate, nitrogen, tofu wastewater

Abstract

AbstrakTujuan penelitian ini adalah untuk mengetahui tingkat kemurnian hasil pengendapan ekstrak kasar selulase Bacillus circulans pada berbagai kejenuhan aseton;   pengaruh waktu pengendapan terhadap tingkat kemurnian hasil pengendapan pada kejenuhan aseton optimum; dan stabilitas waktu penyimpanan fraksi optimum pada suhu 0°C selama 0,1,4 dan 7 hari.Aktivitas selulase diukur berdasarkan kenaikan kadar gula pereduksi yang dihasilkan pada substrat kertas Whatman no 1. Penentuan kadar gula pereduksi menggunakan metode Somogy-Nelson. Kadar protein ditentukan dengan metode Lowry. Tingkat kemurnian enzim merupakan perbandingan aktivitas spesifik enzim setelah pemurnian dengan sebelum pemurnian.Tingkat kemurnian hasil pengendapan ekstrak kasarBacillus circulans mengalami peningkatan pada kejenuhan aseton ( 33 %), (41,1 %) dan (50 %) kemudian menurun pada kejenuhan (60 %) dan (67 %). Fraksi yang aktivitas spesifik tertinggi adalah pada tingkat kejenuhan aseton 50 %, dengan tingkat kemurnian rata-rata 63,61 ± 0,141 kali ekstrak kasar dan perolehan protein kembali rata-rata 76,85 ± 7,99 %. Waktu pengendapan tidak berpengaruh terhadap tingkat kemurnian hasil pengendapan ekstrak kasar selulase Bacillus circulans pada kejenuhan aseton optimum.Selulase hasil pengendapan dengan aseton memiliki stabilitas yang kurang baik pada suhu ± 0°C karena menurun sebesar 41,1 % pada hari pertama dan 88,2 % pada hari ketujuh setelah penyimpanan. Kata kunci: selulase, Bacillus circulans, pengendapan, aseton.    AbstractThe aims of this study were to determine the purity level of precipitations of  cellulases of Bacillus circulans at various saturation of acetone; the effect of precipitation time on the purity level of the precipitations at optimum acetone saturation; and the stability of the storage time of the optimum fraction at 0 ° C for 0,1,4 and 7 days. The activity of cellulase was measured based on the increased of reducing sugar levels produced from hydrolysed of Whatman paper as substrate. The reducing sugar levels were determined with  the Somogy-Nelson method. Protein levels are determined with the Lowry method. The purity level of the enzyme is a comparison of specific enzyme activity after and before purification. The purity level of precipitations of cellulose of Bacillus circulans has increased in saturation of acetone (33%), (41.1%) and (50%) then decreases in saturation (60%) and (67%). The highest cellulase specific activity fraction was at 50% acetone saturation level, with an average purity level of 63.61 ± 0.141 times crude extract and yield protein recovery an average of 76.85 ± 7.99%. The precipitation time did not affect the purity level of the precipitations at optimum acetone saturation. The precipitations had low storage stability at ± 0 oC because its cellulase activity decreased by 41.1% on the first day and  88.2 % on the seventh day after storage The keywords: cellulose, Bacillus circulans, precipitation, aceton

Abstract

Limbah Taman Kampus berupa rumput pahitan berpotensi sebagai pakan ternak ruminansia karena kandungan selulosanya yang tinggi, namun selulosa tersebut masih dalam bentuk lignoselulosa yang menurunkan ketercernaannya pada rumen ruminansia sehingga bentuk lignoselulosa tersebut perlu didegradasi untuk menurunkan kadar lignin sebagai zat anti nutrisi. Fermentasi limbah rumput pahitan menggunakan Kapang Pelapuk Kayu Indigenous Thermothelomyces guttulata KLUM2 merupakan metode alternatif untuk menurunkan kadar lignin dalam menciptakan kadidat pakan ternak yang mudah dicerna dari limbah rumput pahitan. Tujuan penelitian ini adalah mengamati pengaruh jumlah spora dan waktu fermentasi terhadap kandungan lignin, lemak kasar, dan protein kasar selama fermentasi rumput pahitan oleh kapang pelapuk kayu Thermothelomyces guttulata KLUM2. Penelitian ini merupakan penelitian eksperimen laboratories menggunakan metode RAL dengan variasi jumlah spora (1,23×107; 3,07×107; dan 6,14×107sel) dan waktu fermentasi (14 dan 21 hari). Tahapan penelitian ini: (1) Preparasi limbah rumput, (2) Preparasi suspensi spora Thermothelomyces guttulata KLUM2, (3) Fermentasi limbah rumput pahitan oleh Thermothelomyces guttulata KLUM2, dan (4) Analisis kadar lignin, protein kasar, dan lemak kasar. Hasil penelitian yang dilakukan menunjukkan bahwa fermentasi limbah rumput pahitan dengan Thermothelomyces guttulata KLUM2 mampu menurunkan kadar lignin dan meningkatkan kadar lemak serta protein kasar, namun secara umum tidak dipengaruhi oleh variasi lama dan jumlah spora. Kondisi optimum yang diperoleh adalah kombinasi jumlah spora 6,14x107 sel dan waktu fermentasi 21 hari menghasilkan kadar lignin, kadar lemak kasar dan kadar protein kasar berturut-turut sebesar 12,20±0,25%; 2,51±0,38%; dan 11,17±0,08%. Berdasarkan hal tersebut maka fermentasi menggunakan KPK Indigenous Thermothelomyces guttulata KLUM2 berpotensi dalam rangka menciptakan pakan ternak yang mudah dicerna oleh ruminansia.

Abstract

Exploration of aerobic bacteria which can produce cellulase system is very important to discover potential celulase resourse which can hydrolysis lignocelulose waste into glucose. The aimed of this researched were determine optimum condition of cellulase system production in Berg’s media with avicel as inducer and it’s characterization. Bacillus circulans local strain from laboratory of Microbiology ITB produced consist of CMC- ase(111.11U/mL)andavicelas (55.56U/mL) in Berg’s medium contain 0.5% avicel, pH=9.0 and 5 days of incubation. Characteristic of this cellulase system were: (1) optimum level of CMC-ase (129.97 U/mL) and avicelase (87.96 U/mL) was obtained at pH= 7.0, temperature 50oC and 2 hours incubation, (2) Vmaks and Km of CMC-ase was 1000 μg glucose/hour and 5%, Vmaks and Km of avicelase was 200 μg glucose/hour and 1.2%, (3) capable of hidrolizing sugarcane, corn cob and rice bran during optimum condition and released glucose 262 ppm, 81 ppm and 78 ppm. This research encouraged that Bacillus circulans capable of producing cellulose system with high activity and suggested to degradated lignocelluloses as feedstocks of bioetanol.

Abstract

Diare merupakan salah jenis penyakit yang banyak diderita oleh masyarakat di Indonesia. Diare telah menjadi penyebab kematian terutama pada bayi.Diare adalah penyakit yang disebabkan oleh infeksi mikroorganisme termasuk bakteri, virus serta parasit lainnya seperti jamur, cacing dan protozoa.Pengetahuan ibu tentang hygiene makanan merupakan salah satu faktor yang cukup krusial dalam kejadian diare pada bayi. Tujuan penelitian ini untuk mengetahui adakah hubungan pengetahuan ibu tentang hygiene makanan dengan kejadian diare pada bayi. Desain penelitian ini menggunakan analitik korelasional dengan pendekatan cross sectional. Total sampel berjumlah 30 orang yang diperoleh melalui teknik total sampling. Data diperoleh melalui pengisian kuesioner. Data kemudian dianalisis menggunakan uji coefficient contingency. Hasil penelitian menunjukkan bahwa sebanyak 50% ibu mempunyai pengetahuan baik, sebanyak 40% bayi mengalami diare dan sebanyak 60% bayi yang tidak diare. Tidak ada hubungan antara pengetahuan ibu tentang hygiene makanan dengan kejadian diare pada bayi (p= 0,163). Ada berbagai faktor yang mempengaruhi hubungan pengetahuan ibu dengan kejadian diare pada bayi. Faktor sikap dan perilaku ibu dalam memberikan makanan juga mungkin penting untuk diketahui. Kata kunci: Diare pada bayi, hygiene, makanan, pengetahuan ibu.

Abstract

The determinants of customer satisfaction are service quality and price. Therefore, this study aimed to determine the effects of service quality and price on customer satisfaction. This research was conducted at a Forwarding Company, with a total sample of 78 respondents. Meanwhile, the data analysis method used was descriptive and multiple linear regression using SPSS 25. The results showed that service quality and price has a significant effect on customer satisfaction with a determination coefficient of 50%. Meanwhile, the remaining 50% is influenced by other variables, which have been widely studied. However, research in forwarding service companies is rarely conducted.

Abstract

The research purpose is to examine customer loyalty from the variable of mobile banking usability mediated by customer satisfaction. Data were collected in a survey of M-Banking customers conducted from February to August 2020 via a questionnaire involving 105 respondents. The results showed that the ease of using M-Banking lead to customer satisfaction and also lead to bank loyalty, respectively. Considering the change in customer consumption patterns to using technology-based services, banks as service providers need to deliver service in easily, safely, and fast ways, such as internet banking and M-Banking. The research originality examines the direct effect of usability on customer loyalty, especially in conventional banks in Jakarta Province.

Abstract

Tujuan penelitian ini untuk mengetahui pengaruh jumlah spora Paecilomyces inflatus PnUM dan waktu fermentasi terhadap kadar lignin, protein kasar, dan lemak kasar sehingga limbah taman kampus dapat diolah menjadi pakan ternak ruminansia. Penelitian ini merupakan penelitian eksperimen laboratoris menggunakan metode RAL dengan variasi jumlah spora (8,08 x 106; 2,02 x 107; dan 4,04 x 107 sel) dan waktu fermentasi (7, 14 dan 21 hari). Tahapan penelitian ini: (1) Preparasi limbah rumput, (2) Preparasi suspensi spora isolat KPK Indigenous Paecilomyces inflatus PnUM, (3) Fermentasi limbah taman oleh KPK Indigenous Paecilomyces inflatus PnUM, dan (4) Analisis kadar lignin, protein kasar, dan lemak kasar. Pengujian dilakukan sebanyak dua ulangan. Hasil penelitian menunjukkan fermentasi limbah rumput pahitan menggunakan KPK Paecilomyces inflatus PnUM dapat mendegradasi lignoselulosa dan menurunkan kadar lignin, tetapi perlakuan jumlah spora dan waktu fermentasi yang diberikan tidak menunjukkan pengaruh yang signifikan. Perlakuan paling baik diperoleh pada pemakaian  spora sebesar 2,02 x 107 sel dan waktu fermentasi selama 7 hari menghasilkan rumput pahitan yang potensial digunakan sebagai alternatif pakan hijauan ruminansia dengan kadar lignin 14,13%±0,04; kadar lemak kasar 2,75%, dan protein  kasar 11,71%±0,1. Berdasarkan hal tersebut maka fermentasi menggunakan KPK Indigenous Paecilomyces inflatus PnUM sebagai alternatif pengolahan limbah rumput pahitan cukup potensial untuk menyediakan alternatif pakan hijauan ruminansia dan mendukung program Green Campus UM.

Abstract

Good human resource management can be leveraged to ensure a company's success. This study aims to determine the relationship between psychological capital, subjective well-being, work engagement, and employee performance. Data was collected by distributing questionnaires to 407 employees of steel billet and iron rod manufacturing companies in Jakarta. The collected data was then analyzed using Structural Equation Modeling (SEM). This study found that high psychological capital is able to improve work engagement and employee performance, but not subjective well-being; high work engagement is able to improve employee performance and subjective well-being, and high employee performance is able to improve subjective well-being. Furthermore, this study found that psychological capital affects subjective well-being mediated by work engagement, while psychological capital does not affect employee performance mediated by work engagement.
