Abstract

Basic laboratory skills are essential for supporting the quality of learning outcomes and processes, and they are also very beneficial as preparation for the workplace. The goal of this activity is for students and graduates of the Faculty of Mathematics and Natural Sciences (FMIPA) to refresh and develop their basic abilities in working in a microbiological laboratory in preparation for their final assignment and the workplace. During COVID-19 pandemic, the agenda is carried out in two methods, namely online and offline systems. The training was conducted online using a video conferencing platform and an online academic information system, as well as offline at the microbiology laboratory, Department of Biology, FMIPA, Universitas Negeri Malang. The participants' comprehension of basic microbiology concepts and basic abilities for working in a microbiology laboratory improved as a result of this exercise according to the pretest and posttest result.

Abstract

Transactions using e-commerce are a form of changing the buying and selling culture, especially after the Covid 19 Pandemic. This transaction can facilitate and benefit both parties. This study focuses on consumer repurchase intentions from electronic word of mouth (e-Wom, trust, and risk. The research was conducted from February to July 2021 with e-commerce as the research object. A conditional sample was used to ensure that the data obtained was valid and could be accounted for, namely the age group of generation Z and millennials who use e-commerce and are domiciled in DKI Jakarta Province, with a total sample of 120. The results show that generation Z and millennial consumers' repurchase intention is influenced by e-Wom and trust. It is also a consideration for consumers to make repeat transactions. The results of this study also prove that generation Z and millennial consumers prefer to use E-commerce in making purchases. It shows that to attract the attention of this generation, companies can build repeat purchases on e-commerce by maintaining positive e-Wom to create the same trust very high, and the risk of buying online is shallow.

Abstract

This study was aimed to characterize lignin peroxidase (LiP) obtained from Phanerochaete chrysosporium ITB isolate. The characterizations included molecular weight, the pH and optimum working temperature of the crude extract of the enzyme, the temperature stability, the effect of metal ions and inhibitors, their precipitation with ethanol, and the storage stability. The LiP of P. chrysosporium ITB isolates was 34 kDa. The crude extract of LiP displays high activity at pH between 3 until 5 and 26–32 °C, has good thermal stability at 26–32 °C for 20 h. The activity is affected by Pb2+, K+, Co2+, Fe2+, Cd2+, Mg2+, and Cu2+ EDTA, Na+, Cr3+, Hg2+, NaN3, Ni2+, and Ca2+ ions, is not affected by Mn2+ and Zn2+ ions, precipitated with the optimum ethanol at 64% ethanol saturation which results in an increase in specific activity of 2.3 times. The crude extract storage at 0 °C is more stable than the precipitate resulting from ethanol precipitation and resuspension from ethanol precipitation. These results strengthen that LiP from P. chrysosporium is another LiP isoenzyme that can be used for bioremediation processes. Unfortunately, the concentration using the ethanol precipitation method has not been effective, so further studies using other methods should be required.

Abstract

Produktivitas tebu dapat menurun karena serangan hama dan keberadaan gulma di kebun. Pengendalian penggunaan pestisida masih menjadi pilihan ketika serangan hama dan populasi gulma tidak dapat dikendalikan secara manual. Aplikasi pestisida pada tanaman tebu tidak menutup kemungkinan bersamaan dengan parasitoid Trichogramma sebagai agen pengendali hayati hama penggerek tebu. Oleh karena itu tujuan dari penelitian ini adalah untuk mengetahui pengaruh aplikasi pestisida berbahan aktif klorantraniliprol serta paraquat diklorida dan diuron terhadap proporsi penetasan parasitoid, proporsi malformasi, dan lama hidup parasitoid Trichogramma japonicum. Percobaan Menggunakan Rancangan Acak Lengkap (RAL) dengan 5 perlakuan dan 4 ulangan. Perlakuan yang diuji adalah kontrol (tanpa perlakuan), infestasi pias T. japonicum pada daun tebu yang terbuka insektisida/herbisida hari ke-0, ke-1, ke-2, dan ke-3. Hasil pengamatan menunjukkan bahwa aplikasi insektisida berbahan aktif klorantraniliprol tidak berpengaruh nyata terhadap persentase penetasan dan kematian parasitoid tetapi berpengaruh nyata pada persentase malformasi serangga. Adapun aplikasi herbisida berbahan aktif paraquat diklorida dan diuron berpengaruh nyata terhadap proporsi penetasan, malformasi serangga dan kematian parasitoid T. japonicum .Kata kunci : insektisida, herbisida, klorantraniliprol, paraquat diklorida, diuron, Trichogramma  ABSTRACTSugarcane productivity decreases due to the presence of weeds and pest incursion in the field. Pesticide is still an option in controlling pest and weed populations when they cannot be controlled manually. Sometimes pesticide application coincides with the release of parasitoid Trichogramma, a biological control agent of sugarcane borer pests. Therefore, the purpose of this study was to determine the effect of the pesticide application with active ingredients chlorantraniliprole, paraquat dichloride and diuron on the percentage of parasitoid hatching, malformations, and the life span of the parasitoid Trichogramma japonicum.The experiment used a completely randomized design (CRD) with 5 treatments and 4 replications. The treatments tested were control (untreated), T. japonicum infestation on sugarcane leaves exposed to insecticides/herbicides on the 0, 1, 2, and 3 days. The results showed that the application of insecticides with the active ingredient chlorantraniliprole had no significant effect on hatching and parasitoid deaths percentage, but it was a significant different in the percentage of insect malformations. Meanwhile the application of herbicides with the active ingredients of paraquat dichloride and diuron had a significant effect on hatching, malformation and parasitoid deaths percentage of T. japonicum.Key word :   insekticide, herbicide, clorantraniliprol, paraquat diclorida, diuron, Trichogramma

Abstract

Red House Coffee merupakan salah satu UMKM Aceh Tengah Desa Berghendal Kecamatan Meria Satu Kabupaten Bener Meriah yang dibangun pada 2016 oleh Isra Failani. Usaha ini telah  menerima surat perizinan Besertifikasi Pemenuhan Komitmen Produksi Rumah Tangga (SPP-IRT) dan NIB. Red House Coffee menyediakan coffee Honey, wine, natural, luwak, robusta, dan speacility.  Omzed Red House Coffee pada tahun 2022 terbilang rendah dibandingkan pengeluaran, yakni mencapai 150 juta/bulan dengan pengeluaran 40 juta/bulan. Tujuan penelitian ini adalah untuk mengetahui efektivitas penyusunan SOP Red House Coffee Gayo. Di sini SOP sangat menunjang kesuksesan sebuah usaha. Metode penelitian yang digunakan berupa kualitatif bersifat deskriptif dengan sumber data  primer dan sekunder dan teknik pengumpulan datanya secara kuesioner serta wawancara.  Berdasarkan hasil penelitian diketahui bahwa SOP Red House Coffee Gayo tidak memenuhi prosedur penyusunan SOP yang benar. Dalam hal ini SOP Red House Coffee belum berbentuk dokumen, hanya berupa perintah secara lisan dari atasan kepada bawahan. Hal ini disebebakan oleh faktor administrasi, kepegawaiaan, dan harga penyusunan SOP yang mahal.

Abstract

Desa Sumberejo merupakan salah satu desa binaan Universitas Negeri Malang yang berada di Kecamatan Gedangan Kabupaten Malang. Desa ini terdiri dari enam dusun yakni Dusun Sumbersari, Mulyosari, Krajan, Dokosari, Tlekung dan Sumberwangi. Desa Sumberejo dikenal sebagai desa penghasil batik, dengan branding “Batik Sujo”. Produksi Batik Sujo melibatkan ibu-ibu rumah tangga yang berasal dari berbagai dusun di Desa Sumberejo. UMKM ini memproduksi beberapa lembar kain batik dalam sehari dan setiap produksi menghasilkan limbah, khususnya limbah cair yang dapat menimbulkan pencemaran pada lingkungan. Edukasi mengenai dampak dan pengelolaan limbah cair batik perlu diketahui oleh semua pembatik, sehingga perlu dilakukan sosialisasi mengenai topik tersebut. Tujuan kegiatan ini untuk mengetahui dampak sosialisasi terhadap tingkat pemahaman mengenai bahaya limbah batik dan cara pengelolaannya. Metode penelitian menggunakan Quasi Experimental dengan One Group Pretest Posttest Design, dengan pengambilan subjek menggunakan metode Quota Sampling. Hasil penelitian menunjukkan bahwa sosialisasi berpengaruh terhadap Pembatik Sujo, karena terjadi peningkatan pemahaman mengenai dampak dan pengelolaan limbah batik meningkat 15%.

Abstract

Siwalan fiber waste (SFW) has the potential as a source of lignin (7-25%) which is used as a green inhibitor to inhibit the corrosion rate of low carbon steel (LCS). The aims of this study were: (1) determine the optimum conditions (types of HCl and H2SO4, Cinhibitor and Cmedia, inhibition temperature, and coating time) for LCS coating process using the inhibitory method using lignin extract from SFW, and (2) knowing the resistance of LCS coated with inhibitor at optimum conditions. Empirical research through laboratory experiments explains the relation between variables by analyzing numerical data. 185g of SFW produced 52.289g (28.26%) fine brown powder. IR spectrum analysis on extract showed the wavelength was almost the same as the standard. IR spectrum analysis was also carried out on the extracted compounds that were adsorbed, and the absorption peaks confirmed the presence of lignin in the adsorbed compounds. Based on the results of the analysis of the weighing data of the specimen before and after inhibition, the inhibitor concentration of 0.2–0.3 g/L in H2SO4 at 313K is the optimum condition to suppress the corrosion rate on the surface of LCS by 76.2%.

Abstract

Halal food assurance is becoming more important with the growth of the halal industry globally. Adulteration of halal meat products using non-halal sources such as pork, dog, boar, and even frog meat has become a major problem for moslems. The purpose of this study is to initiate the method for frog meat identification using polymerase chain reaction (PCR) technique. In this study, three primer pairs (Fk1-Rk1, Fk2-Rk2, Fk3-Rk3) were analyzed for their specificity toward frog meat against other common halal meat sources such as beef, chicken, shrimp, squid, and mackerel. The visualization of DNA amplification showed that primer pair Fk1-Rk1 produced primer-dimer, thus cannot be used for this circumstance. Primer pair Fk2-Rk2 showed a better result where DNA amplicon was produced at ~100 bp for frog meat and no amplicons for other meat. Primer pair Fk3-Rk3 showed a different pattern of DNA amplification for all the meat tested, where the amplicon of frog meat was shown at ~100 bp, while the other meat showed multiple amplicons or none. In conclusion, primer pairs Fk2-Rk2 and Fk3-Rk3 showed their potential as primer pairs for frog meat identification using PCR for implementing halal food assurance, although sensitivity analysis needs to be investigated.

Abstract

Terdapat 9314 pedagang makanan kaki lima di kota Malang, dan jumlah terbanyak (717 pedagang) adalah “nasi lalapan” yang berjualan mulai sore hari (jam 17.00) sampai malam hari. Usaha ini cukup diminati dan tidak pernah sepi dari pengunjung khususnya konsumen mahasiswa dan masyarakat kota Malang. Namun, ada beberapa kebiasaan buruk yang sering dilakukan oleh para pedagang pada umumnya yaitu: sudah mulai menggunakan minyak goreng bekas di awal penggorengan, dan melakukan pengulangan penggorengan sampai 20 – 30 kali penggorengan. Kondisi ini tentu saja akan sangat membahayakan bagi konsumen. Solusi yang tepat untuk mengatasi ini adalah pelatihan penggunaan arang aktif untuk pemurnian minyak goreng jelantah agar tetap dapat menjaga kesehatan dan keamanan pangannya dengan memperhatikan nilai ekonomisnya. Metode pengabdian dilakukan melalui penyuluhan tentang bahaya penggunaan minyak goreng jelantah melalui video motion grafis yang menarik, pelatihan pemurnian minyak goreng, dan pendampingan ke 10 lokasi pedagang “ayam lalapan” di lingkar kampus. Pemberian edukasi ini sangat dirasakan kebermanfaatannya, dan langsung dapat dipraktekan oleh para pedagang karena bisa menekan biaya produksi, mudah diperoleh, dan mudah pengerjaannya serta tetap memperhatikan kesehatan konsumennya. Pemberian pelatihan ini juga bisa dilakukan kepada para pedagang yang lainnya karena memiliki masalah yang sama.Kata kunci—pemurnian, minyak jelantah, arang aktif, pedagang kaki lima
