Abstract

Indonesian coffee and cocoa commodities are constrained by low product quality problem due to contamination of fungal metabolites which  producing ochratoxin A (OTA). Ochratoxin is neprotoxic, immunogenic, carcinogenic and teratogenic to the human health. Early detection method on site detection should be developed  because of  those negative effects. The aim of this study  was to produce antibody to develop a method for  OTA detection. Antibody was produced by immunization of egg laying hen. Antibody-produced was sepatared and analyzed using ELISA (Enzyme-Linked Immunosorbent Assay) and DBIA (dot blot immunoassay),and tested its composition using HPLC and SDS PAGE. The results showed that anti-OTA polyclonal antibodies had been obtained already from chicken eggs in the 4th period (7 weeks after initial immunization). These antibodies showed anti-OTA reactivity by DBIA method and still showed anti-OTA reactivity up to 9th period (12 weeks after initial immunization). The anti-BSA antibodies produced should be removed to increase the sensitivity of antibodies againts ochratoxin A. The separation of BSA antibodies can be conducted by the absorption of the protein.  [Keywords: ochratoxin A; early detection; antibody IgY]. AbstrakKomoditas kopi dan kakao Indonesia terkendala masalah mutu produk yang rendah akibat kontaminasi cendawan penghasil okratoksin A. Okratoksin A (OTA) bersifat neprotoksik, imunogenik, karsinogenik dan teratogenik yang membahayakan kesehatan manusia. Karena efek negatif yang diakibatkan oleh mikotoksin ini, maka perlu dikembangkan deteksi dini kontaminasi okratoksin langsung di lokasi. Penelitian ini bertujuan menghasilkan antibodi imunoglobulin Y (IgY) untuk mengembangkan metode perakitan perangkat deteksi cepat berbasis imunologi untuk deteksi OTA. Antibodi dihasilkan menggunakan uji ayam petelur. Antibodi yang dihasilkan dipisahkan dan dianalisis aktivitasnya dengan ELISA (Enzyme-Linked Immunosorbent Assay) dan DBIA (dot blot immunoassay), serta diuji komposisinya dengan HPLC dan SDS PAGE. Hasil penelitian menunjukkan bahwa antibodi poliklonal anti-OTA sudah diperoleh dari telur ayam pada periode ke-4 (7 minggu setelah imunisasi awal). Antibodi ini menunjukkan reaktivitas anti-OTA dengan metode DBIA dan masih menunjukkan reaktivitas anti-OTA sampai periode 9 (12 minggu setelah imunisasi awal). Komposisi asam amino antibodi anti-OTA menunjukkan perbedaan dengan komposisi asam amino IgY di database. Antibodi anti BSA yang dihasilkan harus dihilangkan terlebih dahulu untuk meningkatkan sensitivitas antibodi terhadap okratoksin A dan pemisahan dapat dilakukan dengan penyerapan antibodi BSA.[Kata Kunci:  okratoksin A;  deteksi dini; antibodi IgY].

Abstract

Meat refers to parts of an animal's body, such as liver, kidneys, brain, and other muscle tissues that are edible, and meat has a fairly complete nutritional content. Meatballs are a type of food popular in Indonesia, but are prone to spoilage due to the activity of microorganisms. The production of beef meatballs with the addition of moringa leaf powder (Moringa oleifera) can inhibit microbial growth and affect the quality of beef meatballs. This study aims to evaluate the effect of Moringa oleifera (Moringa leaf flour, MLF) addition on total microbes and organoleptic of beef meatballs, including color, texture, taste, and preference for color, aroma/smell, texture, and flavor. The research method used a completely randomized design (CRD) with four treatments: meatballs without the addition of MLF (T0) and meatballs with the addition of MLF 1% (T1), 2% (T2), and 3% (T3). The parameters observed included measurements, total microbes, and organoleptic characteristics, which included hedonic and sensory quality tests of beef meatballs. The results showed that the addition of moringa flour had a significant effect (P<0.05) on the results of hedonic and organoleptic quality tests but had no significant effect (P>0.05) on total microbes. The study results show that adding MLF at different levels does not effectively suppress the microbial growth of beef meatballs. Adding MLF with different levels in beef meatballs does not effectively suppress microbial growth. The addition of MLF in the hedonic and organoleptic quality tests significantly affected color, texture, smell, and taste, and was still acceptable to the panelists.

Abstract

Purpose: This research investigates the specific language needs of esports professionals in Indonesia, particularly assessing whether conventional English classes effectively support their international success. It also explores language barriers that hinder team dynamics and performance. As the global esports industry continues to expand, effective communication among players is increasingly recognized as a critical factor in achieving team success. Methodology: This study employed a mixed-methods approach, combining both quantitative and qualitative data. The participants consisted of 17 esports professionals, including players, coaches, managers, casters, streamers, and brand ambassadors. Data were gathered through structured questionnaires, in-depth interviews, and direct observations during esports events organized by local esports bodies. Results: The findings indicate that conventional English classes do not meet the specialized communication needs of esports professionals. Participants reported difficulties in understanding contractual agreements, following instructions from international coaches, and communicating effectively during international matches. These communication gaps negatively impact team cohesion and performance. Applications/Originality/Value: The study underscores the urgent need for English for Specific Purposes (ESP) programs tailored to the esports context. The results are valuable for educators, policymakers, and esports organizations in designing targeted language training initiatives that align with the unique demands of the esports industry and enhance the global competitiveness of Indonesian esports professionals.

Abstract

Purpose: This research aims to identify the learner profiles of economics and medical students at a university in Indonesia. The goal is to use these profiles as a foundational reference for customizing English for Specific Purposes (ESP) programs to suit students' contextual and disciplinary needs. Methodology: A qualitative approach was employed in this study. Data were collected through semi-structured interviews conducted in three rounds, each lasting 30 minutes. The participants consisted of 12 ESP lecturers from higher education institutions in Indonesia. Results: The findings revealed that medical students generally have high English proficiency, are hardworking, diligent, highly motivated, well-mannered, active, and goal-oriented. In contrast, economics students tend to be passive, sociable, polite, less motivated, lower in English proficiency, and more dependent on contextual learning. Applications/Originality/Value: This research highlights the necessity of considering students’ distinct characteristics in the development and implementation of ESP programs. Tailoring instructional strategies based on student profiles can significantly enhance the effectiveness and relevance of ESP teaching in higher education contexts.

Abstract

Purpose: This study aims to explore the significance of English language proficiency among batik sellers in Beringharjo Market, Yogyakarta, in response to the rise of international tourism. It focuses on identifying specific English language needs that support effective communication with foreign tourists in daily business transactions. Methodology: This qualitative research involved four batik sellers as participants. Data were gathered through in-depth interviews to gain insights into their experiences, language use, and perceptions regarding the importance of English in their business activities. Results: The findings revealed varying perspectives among participants. Some sellers viewed English as essential to customer engagement, while others considered it less critical. The participants also demonstrated diverse levels of English proficiency, with speaking and listening as the most utilized skills. Common challenges included pronunciation difficulties and limited vocabulary. Applications/Originality/Value: This research offers practical insights for designing English for Specific Purposes (ESP) programs tailored to traditional market vendors. By aligning language training with real communicative needs, particularly in high-tourism areas, such initiatives can empower small business owners to interact confidently with international customers and enhance the competitiveness of local cultural economies.

Abstract

Purpose: This study aims to examine the English language requirements of tour guides at Mangkunegaran Palace to support the preparation of tourism students in meeting the practical demands of the guiding profession. English proficiency is essential in traditional tourism contexts such as Mangkunegaran Palace, which receives a considerable number of foreign visitors. Methodology: An exploratory qualitative approach was employed to investigate the English needs of professional tour guides. Data were collected through interviews with five selected tour guides based on their experience and length of service. The interviews focused on the English tasks they perform and the specific language demands of their roles. Results: The findings indicate that most tour guides have an educational background in tourism or a related field. They frequently utilize their speaking and listening skills when guiding tourists, and the English training they received in school has been beneficial. A notable challenge they face is understanding the diverse accents of international tourists, which can hinder effective communication. Applications/Originality/Value: The study suggests that well-designed English for Specific Purposes (ESP) courses are essential to support tour guide professionals. These courses should emphasize the development of practical communication skills tailored to real-world tourism contexts, particularly in handling cross-cultural and multilingual interactions.

Abstract

Purpose: Speaking skill is an integral part of language learning, yet many students face speaking difficulties, including language anxiety, negative expectations, and lack of motivation. These issues hinder vocabulary retention, slow speaking progress, and lead to insecurity. This study aims to examine students’ perceptions of storytelling as a method to improve speaking skills in English as a Foreign Language (EFL) classrooms. Methodology: This research employed a qualitative approach, using semi-structured interviews and direct observation during storytelling activities as primary data collection methods. The data were analyzed using two theoretical frameworks: the cognitive theory of George Lakoff and Mark Johnson, which integrates cognition with perceptual and motor experiences; and Miles and Huberman’s qualitative data analysis theory to construct the conceptual framework. Secondary data were also obtained from journals and articles. Results: The findings revealed that students showed noticeable improvement in their speaking skills after participating in storytelling activities. They engaged enthusiastically and demonstrated increased confidence and vocabulary use. Storytelling was also found to be an effective pedagogical approach for enhancing speaking skills. Applications/Originality/Value: This research provides valuable insights for future studies in EFL teaching strategies, particularly in the use of storytelling. It also offers practical implications for teachers to adopt storytelling as a method to evaluate and improve students’ English-speaking abilities.

Abstract

Sabila Florist merupakan salah satu usaha mikro kecil dan menengah (UMKM) yang bergerak di bidang florist dan berlokasi di Jakarta. Usaha ini menghadapi kendala dalam memperluas jangkauan pasar serta meningkatkan daya saing di era digital karena keterbatasan dalam pemanfaatan media pemasaran online. Kegiatan pengabdian ini bertujuan untuk meningkatkan kompetensi pelaku usaha dalam memahami dan menerapkan strategi pemasaran digital sebagai upaya mendukung pertumbuhan dan nilai ekonomi usaha. Metode pelaksanaan dilakukan melalui pendekatan pelatihan dan pendampingan langsung terhadap satu mitra utama, yaitu Sabila Florist, dengan melibatkan dua orang pengelola inti. Materi yang diberikan mencakup pelatihan penggunaan media sosial, aktivasi Google My Business, pembuatan konten promosi digital, serta strategi pemasaran melalui platform marketplace. Hasil kegiatan menunjukkan adanya peningkatan pemahaman strategi digital marketing, peningkatan kemampuan komunikasi daring (softskill), serta potensi peningkatan nilai ekonomi usaha melalui transaksi berbasis online.

Abstract

Transformasi digital telah menjadi prioritas dalam pembangunan sektor Usaha Mikro, Kecil, dan Menengah (UMKM), mengingat kontribusinya yang signifikan terhadap Produk Domestik Bruto dan penyerapan tenaga kerja di Indonesia. E-business, sebagai bagian dari digitalisasi proses bisnis, dipandang sebagai solusi strategis untuk meningkatkan daya saing UMKM di era industri 4.0. Namun demikian, adopsi e-business tidak dapat dilepaskan dari kesiapan teknologi dan kompetensi sumber daya manusia (SDM) yang memadai. Penelitian ini bertujuan untuk menganalisis secara sistematis kesiapan teknologi dan SDM terhadap implementasi e-business pada UMKM di Indonesia melalui pendekatan Systematic Literature Review (SLR). Penelusuran literatur dilakukan terhadap 20 artikel berbahasa Indonesia yang diterbitkan antara tahun 2020–2025. Data dianalisis dengan teknik tematik berdasarkan kerangka Technology-Organization-Environment (TOE) dan indikator kompetensi SDM. Hasil studi menunjukkan bahwa kesiapan teknologi memainkan peran penting dalam mendukung transformasi digital, namun belum semua UMKM memiliki infrastruktur dan literasi digital yang memadai. Di sisi lain, kompetensi SDM, terutama dalam hal adaptasi terhadap teknologi dan kemampuan kolaboratif, menjadi faktor penentu dalam keberhasilan e-business. Temuan juga mengindikasikan bahwa interaksi antara teknologi, organisasi, dan lingkungan menentukan tingkat kesiapan UMKM secara menyeluruh. Studi ini merekomendasikan perlunya strategi penguatan SDM dan dukungan infrastruktur teknologi yang berkelanjutan, agar implementasi e-business di sektor UMKM dapat dilakukan secara inklusif dan berkelanjutan. Abstract

Meningkatnya tren investasi aset digital di Indonesia menunjukkan antusiasme masyarakat terhadap instrumen keuangan berbasis teknologi. Namun, masih banyak calon pengguna yang ragu untuk terlibat karena keterbatasan pemahaman, keraguan terhadap platform digital, dan persepsi terhadap keamanan transaksi. Penelitian ini bertujuan untuk menganalisis pengaruh promosi digital, kemudahan penggunaan, dan kepercayaan terhadap minat pembelian cryptocurrency pada platform Indodax. Penelitian ini menggunakan pendekatan kuantitatif dengan metode survei. Data dikumpulkan melalui penyebaran kuesioner kepada 100 responden yang merupakan pengguna aktif Indodax. Teknik analisis data yang digunakan adalah regresi linier berganda dengan bantuan perangkat lunak SPSS versi 26. Hasil penelitian menunjukkan bahwa ketiga variabel independen, yaitu promosi digital, kemudahan penggunaan, dan kepercayaan, secara parsial maupun simultan berpengaruh positif dan signifikan terhadap minat pembelian cryptocurrency. Dari ketiga variabel tersebut, kepercayaan menjadi faktor yang paling dominan memengaruhi minat pengguna. Implikasi dari penelitian ini menunjukkan bahwa peningkatan literasi digital, penyampaian informasi promosi yang lebih jelas, serta edukasi tentang keamanan sistem perlu diperhatikan guna membangun kepercayaan dan meningkatkan partisipasi pengguna terhadap investasi cryptocurrency melalui platform Indodax.
