Abstract

One way to produce good seedlings is to use appropriate media for seedling growth so that they can produce healthy seedlings with optimal growth. The use of soil mixed with organic matter compost of Lamtoro (Leucaena leucocephala) leaves with certain amounts is expected to increase the growth of Yellow Cempaka seedlings (Michelia champaca L). This study aims to determine the effect of various doses of compost of Lamtoro (Leucaena leucocephala) media on the growth of yellow Cempaka (Michelia Champaca L) seedlings. The research was carried out for three months, namely from February to May 2019, at the BPDAS Palu-Poso Permanent Nursery, Tadulako University, Palu. The study used a Completely Randomized Design (CRD) consisting of 4 treatments namely L0 = soil (control), L1 = soil and lamtoro leaf compost 1: 1, L2 = soil and leaf compost Lamtoro 1: 1/2, L3 = soil and compost Lamtoro leaf 1: 1/3. The parameters observed were height increase, an increase in the number of leaves, an increase in diameter and robustness of seedlings. Data analysis using analysis of variance (F-Test). The results showed that the different doses of compost of Lamtoro (Leucaena leucocephala L) leave had a very significant effect on height increase, increase in the number of leaves of seedling robustness and had no significant effect on seedling stem diameter. The soil treatment and 1:1/2 lamtoro leaf compost (L2) gave the best effect on the growth namely height increase (4.18 cm), leaf number increase (4.2 strands), and robustness of seedlings (93.71).

Abstract

Chicken manure compost contains high levels of nitrogen, phosphorus, and potassium nutrients that are useful for plants, the provision of chicken manure compost can improve soil structure on lands that lack on organicmatter. The research was conducted in may-juli 2019 at the palu – poso bpdas permanent seedbed, tadulako university, central Sulawesi. This research used completely randomized design method with four treatments, namely S0 = soil without chicken manure compost; S1 = 1000g of soil : 50g of compost; S2 = 1000g of soil : 100g of compost; S3 = 1000g of soil : 150g of compost. Each treatment was repeated 10 times so that there are a total of 40 experiment units. Growth parameters observed ware seedling height, stem diameter, number of leavers, wet weight, shoot dry weight and seedling root, and seed quality indeks. The results show that chicken manure compost significantly ofected the growth of glodokan seedling growth. Treatment S1 = 1000g of soil : 50g chicken manure compost significantly increases the growth of glodokan seedling, as follows growth of seedling height (5,68 cm), stem diameter (1,44 mm),  number of leaves (5 leaves), shoot wet weight (2,96 g), root wet weight (0,64g), canopy dry weight (1,19g), root dry weight (0,55g), and seed quality indeks 0,20Kata Kunci: Chicken Manure, Compost, Glodokan, Polyathia longifolia Sonn

Abstract

Tectona grandis is an annual woody plant that has high economic value in trade commodities. Tectona grandis were usually produced in a conventional way using seed. However, large quantities of seed production in a given time become limited because of an outer layer of a hard seed. The success of tissue cultures is influenced by some factors, among which nutrients composition and plant growth regulator balance as a media component. Plant growth regulator which is used as a media component of tissue culture is the cytokinin  and auksin. The aim of this research is to identify the concentration of kinetin and BAP (Benzyl Amino Purine) concentration is the best for Tectona grandis. The research was carried out for three months, from October to December 2019, located in The Biotechnology Laboratory of Agriculture, Faculty of Agriculture, Tadulako University, Palu. The research was utilizes completely randomized design, with 6 handling of BAP 1 mg, BAP 2 mg, BAP 3 mg, Kinetin 1 mg, Kinetin 2 mg dan Kinetin 3 mg. The result of observation shown giving handles BAP and Kinetin have a distinct effect on seedling Tectona grandis growth by in vitro. Besides the other treatment, kinetin 2 mg provides the best response that is emerge bud (5.1) of the day, the number of shoots (1.6) of shoots and the number of leaves (4) of sheets. Keywords: Tissue Culture, Tectona grandis, Kinetin
