Abstract

This study aims to identify the issues raised by the community in protesting coal transportation policies along the Trans-Sumatra Highway that passes through Jambi Province. Coal transportation along public roads often causes issues that interfere with community activities, ranging from traffic congestion and accidents to pollution and road damage. This study examines the issues that significantly impact social life along the Sumatra Trans-Road passing through Jambi Province. The study employs a literature review method, utilizing data from online news sources from 2023 to 2024, which were then analyzed using NVIVO 14. The results of this study indicate that each issue arising from coal transportation activities is interconnected, with traffic congestion and accidents being the most dominant, followed by road damage, community pressure for the completion of a dedicated coal road, and issues related to pollution and welfare.

Abstract

The hematological profile and blood metabolite status are indicators of the physiological condition of livestock. This study aimed to determine the correlation between blood metabolite profiles (blood glucose and cholesterol) and sperm quality (motility and abnormality) in fresh Bali bull semen. Blood collection and semen sampling were conducted twice a week for 5 weeks. The collected blood and fresh semen were evaluated from two treatments: before and after the addition of micronutrients (including Zn, Se, and Vitamin E) in the feed. The parameters observed in this study were blood metabolite profiles (blood glucose and cholesterol levels) and sperm quality (percentage (%) motility and % abnormality of spermatozoa) before and after the addition of micronutrients. The data obtained were analyzed using simple linear regression and correlation tests. The results of this study showed that both treatments had a correlation between blood metabolite profiles and sperm quality in fresh Bali bull semen. Blood glucose levels correlated with sperm motility in fresh Bali bull semen, and good sperm motility values were obtained in all treatments before and after micronutrient administration in the feed. Cholesterol levels correlated with sperm abnormalities in fresh Bali bull semen, and all treatments before and after micronutrient addition in the feed showed abnormality values above the standard value. The blood metabolite profile (glucose and cholesterol) correlates with spermatozoa quality (motility and abnormalities) in fresh semen from Bali bull, where the addition of micronutrients in feed can increase spermatozoa motility but is not effective in suppressing the decrease in spermatozoa abnormalities. Keywords: Blood metabolite, blood sugar, cholesterol, spermatozoa quality

Abstract

This study was conducted to determine the contamination of microorganisms, pH, water activity (aw), as well as sensory tests (color, aroma, flavor, and texture) with the presentation of different cheeses. This research was conducted in an Laboratorium Terpadu and Laboratorium Phatogen, Teknologi Hasil Ternak, Program Study Ilmu Produksi dan Teknologi Peternakan, Fakultas Peternakan, Bogor Agricultural University. Data from the observations in doing calculations using t-test mathematical model of this design is t =  with 2 times each replicate. The variables measured were the value of Total Plate Count (TPC), Potato Dextrote Agar (PDA), pH, aw, and a sensory test. Data were analyzed using t-test model. The results of the analysis of the t-test (α = 5%) of cheddar cheese block and cheddar cheese slice stem not give significant differences (P > 0,05) on the TPC and PDA yeasts or fungi. t-test (α = 5%) to pH and aw on each individual cheese does not give significant differences (P> 0,05). From the results of sensory analysis on the color of the cheese is seen that the panelists really liked the cheese sticks. Rate on cheese stem higher with the average value of which is 4 compared with cheddar cheese shaped slice which has an average value is 3,7. as well as on the assessment of texture and flavor, the rod-shaped cheddar cheese has a value - average higher than shaped slice of cheddar cheese. while the aroma cheddar cheese slice has a higher average value than the rod-shaped cheddar cheese. However this is seen not provide a significant difference due to differences in cheese and cheese slice stem relatively small. On the results of the analysis (t-test) showed that the difference in assessment of color, texture, aroma and taste of the cheese slice and cheese stems not significant or not significant. This is most likely because of the cheese slice and rods produced by the company that produced simultaneously.

Abstract

Lactobacillus plantarum IIA-1A5 is an Indonesian probiotic bacterium isolated from Ongole grade cattle. This strain has demonstrated series of benefical properties and promising to be further applied in development of functional foods. Attempts to further exploit this strain require comprehensive understanding on its molecular properties, in particular is the genes responsible for its probiotic properties.Whole genome sequencing (WGS) is currently an affordable tool to fully decipher genetic organization of the bacteria, yet often limited by the requirement of high amount of high quality of genomic DNA ofsample. This study aims to develop an optimum method for obtaining high quality genomic DNA of L. plantarum IIA-1A5 in sufficient amount of WGS study. To address, genomic DNA of L. plantarum IIA-1A5 was extracted from growth phase of adaptation, exponential and stationary. The result showed that exponential and stationary phases were able to yield genomic DNA higher than minimum requirement for WGS. Nevertheless, the quality of genomic DNA from both phases were considerably low, according to their 260/280 and 260/230 ratio. The worst quality was found on 260/280 ratio referring to thepresence of high amount of proteinaceous compounds in the sample. Further optimization was done on genomic DNA from stationary phase by adding proteinase K at various incubation time. It showed that the incubation for 7.5 to 10 hours yielded acceptable purity and amount of genomic DNA for WGS. In addition, we also observed a trade-off phenomenon of yield-purity genomic DNA of L. plantarum IIA-1A5. Altogether, the optimized method here should contribute to further molecular studies of this probiotic strain.
