Abstract

Tunjuk Langit Plant (Helminthostachys zeylanica (L.) Hook) contains a lot of secondary metabolite compounds, one of which is found in the root which contain the flavonoids ugonin and quercetin. The purpose of this study was to determine the antibacterial activity of compound 4 isolates IFATL-05 endophytic fungi from Tunjuk Langit (Helminthostachys zeylanica (L.) Hook) root against bacterial gastrointestinal tract infection. The results of purification of endophytic fungi with IFATL-05 code on macroscopic examination showed blackish-white fungus color, round with scalloped colony shapes, wavy edges and drop-like elevation angles. The process of separating the compounds using KLTP with n-hexane:ethyl acetate (1:1) as an eluent resulted in four bands of active compounds in UV lamps of 254 and 366 nm (TL-1, TL-2, TL-3 and TL-4). Test results for compound 4 isolates of IFATL05 endophytic fungi from Tunjuk Langit root (Helminthostachys zeylanica (L.) Hook) using the Bioautographic TLC method using n-hexane:ethyl acetate (6:1) as eluent on Escherichia coli, Salmonella thypi, Shigella dysenteriae and Vibrio cholerae with Rf value = 0.90

Abstract

Isolation of bacterial cellulose from fruits in traditional market of Makassar have been done. This objective of this research was to determine optimum temperature of isolate bacteria that produce cellulose from Dragon fruit (Hylocereus polyrhizus) using dilution agar method. Isolation of bacteria produced cellulose using Hestrin-Schramm agar (HSA) obtain 5 (five) isolate NB02, NB03, NS01 and NS02. The result of screening activity showed that isolate NB04 has a potential as bacteria produce cellulose. The temperature optimize of NB04 has been done at 25oC and 40oC. Based on research results showed that optimum temperature for NB04 was at 25oC and has highest cellulose content at 1,29 g.

Abstract

One common disease found in tropical countries is skin infection. Inappropriate use of antibiotics to treat skin infections can lead to bacterial resistance, making it necessary to explore the source of bioactive compounds in natural materials. The Bidara plant (Ziziphus mauritiana Lam.) is known to have antibacterial properties. This research aimed to determine the antibacterial activity of endophytic fungal isolated from the parts of the Bidara plant against bacteria that cause skin infections using the TLC-Bioautography method. In this research, the isolate with the code IFAZ-6 was purified to obtain a pure endophytic fungal isolate, and macroscopic and microscopic examinations were conducted on that isolate. The obtained isolate was fermented for 21 days at room temperature using MYB (Maltose Yeast Broth) medium. The fermentation products were extracted using ethyl acetate (1:1). The obtained extract was further tested using TLC with a chloroform: methanol eluent (7:1). The TLC-Bioautography results showed Rf values of 0.81 and 0.58 indicating antibacterial activity against Staphylococcus aureus, Pseudomonas aeruginosa, Staphylococcus epidermidis, and Propionibacterium acnes bacteria. The identification of chemical components in the endophytic fungal isolate from the Bidara plant confirmed the presence of flavonoids, alkaloids, and anthraquinones. The extract from the endophytic fungal isolate with the code IFAZ-6 indicated antibacterial activity.

Abstract

White weed (Ageratum conyzoides L.) is known to have antibacterial elements containing chemical compound such as saponin and flavonoids. This study aimed to examine the antibiotic activity of endophytic fungi isolates of white weed leaves by the TLC-Bioautography method. The results of endophytic fungi isolation of white weed leaves obtained 10 isolates consisting of IFDP 1, IFDP 2, IFDP 3, IFDP 4, IFDP 5, IFDP 6, IFDP 7, IFDP 8, IFDP 9, and IFDP 10. The results of the macroscopic examination of the ten isolates of endophytic fungi found different characteristics. The results of the screening using 9 samples obtained isolates that found activity showing high inhibitory power, such as IFDP 1, IFDP 2, and IFDP 4 isolates. Isolates of IFDP 1, IFDP 2 and IFDP 4 were fermented on MYB medium for 14 days. Then filtering and evaporation were carried out to produce an extract. Isolate fermentate extracts were identified using Thin Layer Chromatography with chloroform eluent: methanol (4:2). The antibiotic activity test was done using TLC-Bioautography method, and obtained an Rf value in isolate 1, at Rf 0.74 on Vibrio cholerae. Isolate 2 resulted rf value of 0.67 on Salmonella thypi, Vibrio cholerae, Eschericia coli, disentriae, Staphylococcus epidermis. Isolate 4 generated an Rf value of 0.74 on Pseudomonas aeruginosa and Vibrio cholerae. While the group of active chemical components contained in the endophytic fungi isolates of white weed leaves was lavonoids by using AlCl3 reagents, AICI3 reagent alkaloids, and Sulfuric acid for saponin identification

Abstract

Endophytic fungi are fungi that grow and colonize plant tissues and are capable of producing bioactive compounds similar to those produced by their host. This study aims to determine the concentration of endophytic fungal isolates from Bidara roots (Ziziphus mauritiana Lam) that have antibacterial activity against bacteria that cause skin infections using the agar diffusion method. The research method begins with the rejuvenation of IFAZ-6 isolates, and microscopic and macroscopic observations are carried out. The isolates were fermented using Maltose Yeast Broth medium at 200 rpm for 21 days. The fermentate was extracted with an ethyl acetate solvent to obtain a thick extract. The tests carried out were minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and antibacterial activity tests carried out using the agar diffusion method. The results of the antibacterial activity test showed that the IFAZ-6 isolate extract at a concentration of 800 ppm showed antibacterial activity against Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, and Propionibacterium acnes. The statistical analysis results showed that the extract of isolate IFAZ-6 against Staphylococcus aureus and Staphylococcus epidermidis at concentrations of 800-1600 ppm and 1600–3200 ppm exhibited the same antibacterial activity. The bacterium Pseudomonas aeruginosa showed the same antibacterial activity at all tested concentrations, and the bacterium Propionibacterium acnes at concentrations of 1600–3200 ppm also exhibited the same antibacterial activity.

Abstract

Research on optimization time for antibacterial production of endophytic fungal isolated from bidara root (Ziziphus mauritiana Lam.) has been conducted, with the aim of determining the optimum production time for antibacterial compounds. Endophytic fungi are a group of fungi that live within plant tissues without causing harm to their host plants. In this research, the roots of bidara were isolated and purified using PDA media, isolates were followed by screening endophytic fungi isolates against test bacteria. The results of the endophytic fungal isolation from the bidara roots yielded 10 endophytic fungal isolates. The screening test against the test bacteria showed the largest inhibitory zone for IFAZ-6. Subsequently, the growth curve of these isolates was determined through fermentation using MYB media using a shaker speed of 200 rpm at a temperature of 25-28°C for 27 days. Then, mycelia and supernatant were collected on days 11, 13, 15, 17, 19, 21, 23, 25, and 27 with a 48-hour interval. The mycelia were weighed until a constant weight was obtained, while the supernatant was used to test the activity against the test bacteria using MHA media. The growth curve results indicated a stationary phase on day 21. The antibacterial activity test showed that Isolate IFAZ-6 reached optimum activity on day 27 with an inhibitory zone of 12.33 mm against Salmonella thypi.

Abstract

The Indian Costus plant (Saussurea lappa) possesses the ability as an antibacterial, which contains alkaloid compounds, whereas this study aims to determine the antibacterial activity of ethanol extract of Indian Costus plant root (Saussurea lappa). The extraction process of Indian Costus plant root simplicia (Saussurea lappa) was carried out using the Soxhlet method with 96% ethanol as a solvent, and then evaporated with a rotavapor to obtain a viscous extract. Screening tests were carried out by giving ethanol extract of Indian Costus plant root (Saussurea lappa) with a concentration of 0.1% to 2 test bacteria, namely Staphylococcus aureus and Escherichia coli. Thereafter, the Thin Layer Chromatography-Bioautography test was carried out, followed by identification of chemical compounds. The screening test results obtained showed that the ethanol extract of Indian Costus plant root (Saussurea lappa) provided activity against 2 test bacteria at a concentration of 0.1%. The Thin Layer Chromatography-Bioautography test results indicated that there were 3 spots that were formed with Rf values of 0.67, 0.60, 0.54, 0.45, 0.43 providing activity on Staphylococcus aureus and Escherichia coli bacteria. The chemical identification of ethanol extract of Indian Costus plant roots (Saussurea lappa) yeilded positive result for the presence of alkaloids. Based on the activity of the ethanol extract of Indian Costus plant root (Saussurea lappa), it is known that it has potential as an antibacterial.

Abstract

Nona makan sirih (Clerodendrum thomsoniae) contains secondary metabolites that have the potential as antibacterials. The TLC-Bioautography method will determine the antibacterial activity and chemical components of endophytic fungal isolates of nona makan sirih. This research involved the isolation of endophytic fungi from leaves of nona makan sirih using PDAC media, followed by purification and macroscopic and microscopic tests. In addition, a screening test its effect againts pathogenic bacteria. The chosen isolates were fermented for 21 days. Then supernatant and mycelia were seperated. The supernatant was extracted by dissolving ethyl acetate using the liquid-liquid ectraction method. Then extract obtained was processed by TLC with the mobile phase of chloroform:methanol (8:1) and TLC-Bioautography was carried out on bacterial tests and chemical components. The results of isolation and purification obtained as many as 13 isolates. Screening test results with IFDNMS codes 6, 8 and 11 showed activity against the test bacteria. TLC-Bioautography result isolate 6 obtained 5 spots with an Rf value of 0.89; 0.65; 0.45; 0.23; 0.05 was active against all test bacteria. Isolate 8 obtained 5 spots with an Rf value of 0.89; 0.65; 0.05 was active against all tested bacteria, while 0.45 was active against P.acnes, S.aureus, and S.epidermidis bacteria and 0.23 against S.aureus bacteria. Isolate 11 obtained 5 spots with an Rf value of 0.89; 0.61; 0.45; 0.23; 0.05 against all test bacteria. The findings indicate that flavonoids and phenolics are chemical components with antibacterial activity.

Abstract

This research aimed to determine whether the use of the Plotagon Application is effective in enhancing students’ English Presentation skills. The researcher used a pre-experimental as a research design. This research focuses on a single group pre-test and post-test which asks students to present in front of the class on the topic of daily activities. This research was conducted at SMP Telkom Makassar in the eighth grade with a total population of 230 students. The sample was class VIII B with a total of 30 students selected using a random sampling technique. The findings from this study showed that the mean score of pre-tests was 68.11 and post-test was 84.55, this means that the score was increased after the sample received treatment and based on the paired sample on sig. value (2-tailed) was 0.00 < 0.05. It can be concluded that the use of the Plotagon application is effective in enhancing students' English presentation skills.

Abstract

PT.X merupakan perusahaan yang bergerak di industri percetakan dan pengemasan offset yang memproduksi inner pack karton, gift box, dan outor box dengan dukungan teknologi terkini untuk memenuhi permintaan customer dengan kualitas seperti yang diharapkan. Pada penjadwaalan, diketahui bahwa persentase miss (produk yang tidak tercetak) masih dalam kategori tinggi yang pada akhirnya mengakibatkan keterlambatan proses produksi. Dari hasil penelitian yang dilakukan dengan metode OEE diketahui bahwa rata-rata masing-masing OEE bulan desember dan januari adalah 41,11% dan 68,03%. Jika nilai OEE<60% produksi diperlukan adanya ruang yang besar untuk improvement.. Salah satu faktor yang menyebabkan produktivitas mesin menurun adalah adanya downtime yang cukup lama dan frekuensinya sering. Oleh karenanya, perusahaan harus menjaga keandalaan peralatan produksi untuk kelancaran dalam sistem produksinya. Sehingga perlu dilakukan analisa mengenai keandalan mesin menggunakan metode FMEA. Hasil penelitian menunjukkan, Overall Availability (Ao) pada mesin ini masih rendah yaitu 84,99% yang artinya masih dibawah world class standard (≥ 90%).
