Abstract

Induksi embrio somatik pada kopi arabika (Coffea arabica) dengan menggunakan beberapa zat pengatur tumbuh (ZPT) telah berhasil dilakukan. Pengaruh komposisi media terutama kombinasi antara jenis ZPT yang berbeda dan tanggap genotipe tanaman dilaporkan sangat bervariasi. Tujuan penelitian untuk mengkaji pengaruh pemberian 2,4-D dan kinetin dalam proses pembentukan dan pertumbuhan kalus embriogenik asal daun. Penelitian dilakukan di Laboratorium Pusat Penelitian dan Pengembangan Perkebunan (Puslitbangbun) Agustus 2011 sampai Januari 2012. Bahan tanaman yang digunakan adalah daun dari kopi arabika varietas Sigarar Utang yang merupakan tanaman koleksi Balai Penelitian Tanaman Industri dan Penyegar (Balittri). Penelitian menggunakan rancangan acak lengkap dengan 5 ulangan, masing-masing ulangan terdiri dari 5 eksplan. Peubah yang diamati, meliputi persentasi kalus yang terbentuk, morfologi kalus, berat basah kalus, dan jumlah globular. Hasil menunjukkan semua perlakuan dapat membentuk kalus, pertambahan berat eksplan tertinggi diperoleh pada media kombinasi 2,4-D 1 mg/l atau 2 mg/l dan kinetin 1 sampai 4 mg/l. Embrio somatik terbanyak diperoleh pada media yang diberi 2,4-D 0,5 mg/l dan kinetin 1 mg/l. Selain kalus, massa proembrio dan embrio, juga terbentuk akar adventif yang jumlahnya tidak nyata antar perlakuan.  The Effect of Composition Media to Callus Formation of Somaticembryogenesis of Arabica Coffee (Coffea arabica)ABSTRACT Induction of somatic embryos with plant growth regulators (PGR) has successfully performed in arabica coffee. However, the influence of media composition combined with different PGR, explants and genotype of plants is widely various in response yields. The objective of this study was to examine the effects of 2,4-D and kinetine in process of formation and growth of embryogenic callus developed from leaves of arabica coffee. The studiy was carried out at a laboratory of Indonesian Research Center for Estate Crops (Puslitbangbun) from August 2011 to January 2012. Plant materials used are coffee leaves var. Sigarar Utang taken from a germplasm collection of the crop grown at Pakuwon Research Station, Indonesian Research Institute for Industry Crops (Balittri) located at Sukabumi, West Java. A completely randomized design with 5 replications and plot size of five explants was used. Parameters observed are percentage of callus formation, morphology of the callus, fresh weight of callus, and number of globular. Results show that all treatments examined are able to form callus. The highest increase in weight of explants was obtained from the media treated with 2,4-D (conc. of 1mg/l or 2 mg/l) and kinetin (conc. of 1 to 4 mg/l). While, the most number of somatic embryo formed was obtained from those of treated with 2,4-D 0.5 mg/l and kinetin 1 mg/l. In addition to callus formation, proembryo mass, embryo and adventive roots were also formed in spite of not significant between different the treatments.

Abstract

Kopyor coconut is a natural mutant that has abnormal endosperm development. For the first time several genes that were suspected to be related to kopyor trait were identified based on the chemical compounds of the endosperm that different from that of normal coconut. Sucrose synthase (SUS), Stearoyl acyl carrier protein desaturase (SACPD), and Absicid acid insensitive (ABI) genes were isolated and analyzed. Four DNA fragments with length of 746, 738, 780, and 687 bp (CnSus1A, CnSus1B, CnSus2A, and CnSus2B) were obtained from SUS gene. Sequence analysis at DNA and amino acid level showed that CnSus1A, CnSus1B, CnSus2A, and CnSus2B were classified into monocot SUS group with nongrass SUS type. Isolation of SACPD gene resulted in one DNA fragment with DNA length of 716 bp. CnSacpd shared a high homology with SACPD gene of oil palm and soybean. Isolation of ABI gene resulted in two DNA fragments, CnAbi3A and CnAbi3B, with DNA length of 760 and 728 bp, respectively. CnAbi3A and CnAbi3B showed a high homology with ABI3 gene of several plants. All DNA fragment obtained from SUS, SACPD, ABI genes were used as templates to design spesific markers for each corresponding gene. There were 7 specific primer sets designed, i.e., CnSUS1A, CnSUS1B, CnSUS2A, CnSUS2B, CnSACPD, CnABI3A, and CnABI3B.<

Abstract

The aims of this research were to (1) compare the effectiveness of established Agro bacterium mediated transformation and bombardment mediated transformation on peanut (2) evaluate the integration of green fluorescent protein (GFP) and hygromycin resistance (hygr) gene on the peanut embryosomatics. To achieve those objectives we shoot 1200 explants using bombardment and transform 1000 explants using Agrobacterium mediated transformation. Both of Agrobacterium mediated transformation and particle bombardment could give positive embryos transformed.  Particle bombardment gave 4.5% transformation efficiency while Agrobacterium mediated transformation gave only 1.8%. From this research, we concluded that particle bombardment more efficient and gave more transgenic explants than Agrobacterium mediated transformation. Keywords: agrobacterium, bombardment, GFP.

Abstract

Salah satu penyakit pada tanaman karet adalah penyakit gugur daun Corynespora (PGDC) yang disebabkan oleh jamur Corynespora cassiicola. Patogen tersebut dapat menyerang semua tahap pertumbuhan tanaman karet yang menyebabkan terjadinya penurunan hasil lateks yang cukup signifikan dan bahkan dapat menyebabkan kematian tanaman. Penggunaan klon-klon tahan sebagai bahan tanam merupakan cara yang paling efektif dan ekonomis untuk mencegah terjadinya serangan PGDC. Karena itu identifikasi klon-klon resisten merupakan strategi utama dalam manajemen PGDC. Penelitian ini bertujuan untuk mengidentifikasi tingkat ketahanan 50 genotipe terpilih plasma nutfah IRRDB 1981 (PN’81) terhadap PGDC. Enam klon karet yang berasal dari populasi Wickham digunakan sebagai pembanding (BPM 24, BPM 1, GT 1, RRIC 600, PB 260 dan RRIM 600). Empat isolat C. cassiicola (CC-01, CC-20, CC-22, dan CC-23) diinokulasikan masing-masing pada daun muda tahap pertumbuhan B2C, dan intensitas kelayuan daun dihitung berdasarkan estimasi kehilangan air akibat aktifitas patogenisitas toksin. Hasil penelitian menunjukkan bahwa 12 genotipe tergolong sangat tahan, 13 genotipe tergolong tahan, 23 genotipe tergolong rentan dan 8 genotipe tergolong sangat rentan. Abstract

Transcription factors have an important function in regulating gene expression and plant responses to stresses. The ERF, bZIP, WRKY, MYB, and NAC are stress inducible transcription factors. The OsNAC6 is a member of the NAC transcription factor family in rice and its expression is induced by abiotic stresses, wounding and blast disease. Characterization of OsNAC6 gene sequences would give a better undersanding on how OsNAC gene functions biologically. The objectives of this research are to isolate the OsNAC6 cDNA from Nipponbare, Batutegi, and Rojolele cultivars, to characterize their DNA sequences, and to compare their sequences to other NAC genes from other plants available in GenBank DNA databases. Isolated cDNA and sequencing of the fragments resulted in a 912 bp DNA sequences. Translation of the sequences yielded a protein consisted of 303 amino acid residue. Blast analysis of amino acid sequences indicated identity of isolated cDNA from three Indonesian rice cultivars are the OsNAC6 gene. Deduced amino acid residues from amplified cDNAs of Nipponbare, Batutegi, and Rojolele cultivars shared 100% sequence identities to rice OsNAC6 (Acc. # BAA89800), 71-100% sequence identity to a number of OsNAC protein from Oryza sativa and 63-83% sequence identity to NAC protein from other plants.

Abstract

WEB-based bioinformatic analysis on partial genome sequence of Sago (Metroxylon sagu Rottb.)ABSTRACTSago genome sequencing analysis is still very limited. This study is a preliminary study of sago sequence analysis obtained from NGS technology to understand and identify new genetic sequences that have homology to genes in the NCBI database. Sequences were analyzed using Blast2Go to determine the genetic function annotation, putative gene identification was performed on the Arabidopsis database using the BLASTx program with a 10-3 e-value limit on The Arabidopsis Information Resource (TAIR) (http://www.arabidopsis.org/index.jsp). Gene interactions were analyzed using DAVID and GeneMania programs. Based on sequence analysis with Blast2Go, 33 sequences with Blastx hit consisting of: 29 sequences had a high homology. The sago sequences with a similarity of ≥ 90% are glutamate decarboxylase and HT1-like serine threonine kinase with hit number 10. The distribution of interactions between genes from GeneMania analysis is known to be mostly interconnected in the 65.13% protein domain, predicted 19.83%, genes with 14.47% shared expression and the remaining 0.57% had localization together.Keywords: bioinformatics, gene annotation, gene ontology, genome sequence, Metroxylon sagu ABSTRAKKajian analisis sekuen genom sagu hingga saat ini masih amat terbatas. Penelitian ini merupakan riset pendahuluan analisis sekuen sagu yang diperoleh dari teknologi NGS untuk mengetahui dan mengidentifikasi sekuen gen baru yang memiliki homologi dengan gen pada database NCBI. Sekuen dianalisis menggunakan perangkat Blast2Go untuk mengetahui anotasi fungsional gen, identifikasi gen putatif dilakukan terhadap database Arabidopsis menggunakan program BLASTx dengan batas e-value 10-3 pada The Arabidopsis Information Resource (TAIR). Interaksi gen dianalisis menggunakan program DAVID dan GeneMania. Berdasarkan analisis sekuen dengan Blast2Go, diperoleh 33 sekuen dengan Blastx hit yang terdiri atas: 29 sekuen memiliki homologi yang tinggi. Gen dengan rataan kemiripan ≥ 90% adalah glutamate decarboxylase dan serine threonine-kinase HT1-like dengan jumlah hit 10. Persebaran interaksi antar gen hasil analisis GeneMania diketahui sebagian besar saling terkait pada domain protein 65,13%, koneksi yang berhasil diprediksi 19,83%, gen dengan ekspresi bersama 14,47% dan sisanya 0,57% memiliki peranan bersama. Kata Kunci: anotasi gen, bioinformatika, Metroxylon sagu, ontologi gen, sekuen genome

Abstract

The primary and secondary somatic embryogenesis can be used to propagate Coffea arabica L clonally.  However, the success of this propagation was depended on plant growth regulator and varieties. This study aimed to examine the possibility of 2,4-D and thidiazuron application to form primary and secondary somatic embryo to support Arabica coffee clonal propagation. The study consisted of two activities (1) 2,4-D and thidiazuron Application to Induce Primary Somatic Embryogenesis of Arabica Coffee and (2) The Application of thidiazuron in Solid and Semi-Solid Media to Induce Secondary Somatic Embryos.  The results indicated significant effect of varieties and plant growth regulator on fresh weight, number of torpedo and germinated embryo.  However, it showed no significant effect on callus formation percentage. The best medium to induce primary somatic embryogenesis depending on variety, on the treatment of 4.52 μM 2,4 -D +18.16 μM thidiazuron was the best for AS2K and Sigarar Utang varieties, S 795 at 4.52 μM 2,4-D + 9.08 μM thidiazuron, whereas Kartika at 4.52 μM 2.4-D + 13.62 μM thidiazuron.  The morphology of coffee somatic embryo was normal.  Primary somatic embryo was developed indirectly, whereas the secondary somatic embryo was directly.  The application of 9.08 μM thidiazuron  increased the percentage and number of secondary somatic embryos, hence enhancing number of Arabica coffee planlet. Keywords : Coffea arabica L, 2,4-D, thidiazuron, semi-solid media, Indirect somatic embryogenesis

Abstract

Penyakit bilur kacang tanah yang disebabkan oleh peanut stripe virus (PStV)merupakan salah satu penyakit utama pada pertanaman kacang tanah (Arachishypogaea L.). Penggunaan varietas tahan merupakan alternatif paling efektif untukmengatasi penyakit tersebut. Rekayasa genetika merupakan metode efektif untukmendapatkan varietas kacang tanah yang resisten PStV. Tujuan penelitian ini adalahuntuk (1) mengetahui respons tanaman kacang tanah transgenik yang membawa gen cpPStV terhadap infeksi PStV dan (2) menguji stabilitas transgen sampai tujuh generasisilang-dalam. Tanaman kacang tanah transgenik cv. Gajah generasi T0, T1, T2, T3, T5,T6, dan T7 diinokulasi secara mekanik dengan PStV. Terdapat tiga jenis respons kacangtanah transgenik terhadap infeksi PStV, yaitu resisten, recovery, dan rentan. Padatanaman resisten gejala tidak muncul. Pada tanaman recovery, gejala chlorotic ringmottle muncul pada satu daun atau lebih, selanjutnya gejala tidak tampak pada daundaunyang tumbuh kemudian. Pada tanaman rentan, gejala severe blotch muncul padasuatu daun, selanjutnya gejala tersebut tetap muncul pada seluruh daun yang tumbuhkemudian. Transgen cp PStV tetap stabil setelah mengalami tujuh generasi silangdalam.Sejumlah galur murni kacang tanah transgenik yang resisten PStV telahdiidentifikasi.

Abstract

Phalaenopsis amabilis is an Indonesia native orchid species having large, white flowers with yellow labellum coloration. This studies aimed to develop Phal. amabilis micropropagation methods and evaluate the regenerated plantlet fidelity. Media supplemented with Thidiazuron (TDZ) and Polyvinylpyrrolidone (PVP) and medium pH adjustment effects to induce protocorm-like bodies (PLBs) from leaf explants and proliferate secondary PLBs were investigated. Clonal fidelity among regenerated plantlets was evaluated using eighteen SNAP marker loci. The results showed that the ½ MS medium supplemented with 3 mg L-1 TDZ and 0.5 g L-1 PVP was the best for PLB induction while the ½ MS medium supplemented with 0.5 mg L-1 TDZ was the best for PLB proliferation. For PLB induction, the media pH was adjusted into pH=7 for efficient PLB regeneration. Based on the assessment using 18 SNAP marker loci, four variant alleles in three loci (11.8%) out of a total 34 plantlets were detected. The mutation frequency at the evaluated SNAP marker loci was 2.5 x 103 (0.25%). Changes in SNP alleles may not always result in phenotype changes and allele variant occurrences may not affect phenotype fidelity of micro-propagated Phal. amabilis plantlets. Therefore, further studies about the phenotype fidelity among plantlets are necessary.

Abstract

The aim of this research is to analyze and examine the inheritance of stomatal density trait and RWL as a variable in drought tolerance ofpeanut. The experiment was conducted by using cv. Kelinci that is sensitive genotype as female parent and US 605 which is tolerantgenotype as male parent, including population off spring from hybrid cv. Kelinci (P1) with US 605 (P2). Stomatal density was determinedby making leaf imprint and by observing leaf imprint under microscope. Relative water loss was determined by dipping peanut leaf in PEG40% for 48 hours. Result of the analysis showed that stomatal density and RWL were not only controlled by qualitative characters of majorgene, but also controlled by quantitative character of minor gene by polygenic with the complex gene action. Both characters seem toinfluence more as genetic factor and have high level fixation additive varians which can give the opportunity to obtain the tolerant offspring.
