Abstract

Bacterial canker, caused by Clavibacter michiganensis subsp. michiganensis is a newly introduced disease of tomatoes in Indonesia. Its existence was first officially reported in 2004. The objective of this research was to monitor the existence of C. michiganensis subsp. michiganensis in various tomato production centers in Sumatra and Java. Tomato samples showing symptoms of C. michiganensis subsp. michiganensis infection were collected from various tomato production centers in Sumatera and Java and the causal agents were isolated from these samples. Based on the occurrences of typical symptoms of C. michiganensis subsp. michiganensis infection in tomato, the incidence of suspected C. michiganensis subsp. Michiganensis infection ranged from 1-20%. From a total of 74 tomato plants sampled, 24 bacterial isolates exhibiting similar colony morphology to C. michiganensis subsp. michiganensis were obtained. After various physiological, hypersensitive response, and pathogenicity tests, 18 isolates derived from 14 tomato production centers in eight provinces in Sumatera and Java were identified as C. michiganensis subsp. michiganensis. Although the incidence was still low, results of these experiments further indicated that C. michiganensis subsp. michiganensis existed in Indonesia and had spread in a number of tomato production centers in Sumatra and Java. Positive results of C. michiganensis subsp. Michiganensis identification from suspected tomato samples should be a warning sign for all tomato production stakeholders in Indonesia, especially those responsible in regulating the seed trade, importation, and plant quarantine

Abstract

Transgene Identity and Number of Integration Sites and Their Correlation with Resistance To PStV in Transgenic Peanuts Carrying Peanut Stripe Virus (PStV) Coat Protein Gene. This research aimed to determine (1) the identity and copy number of PStV cp gene in transgenic peanut plants carrying PStV cp gene and (2) correlation between the identity and the number of integration sites and resistance to PStV infection. One T0 transgenic peanut was selfed up to five generations. T2, T3, and T5 plants were mechanically inoculated with PStV. Samples of T5 plants derived from several different T4 plants were subjected to Southern analysis to confirm the integration of PStV cp gene and to determine its identity and copy number. The Southern analysis showed three bands of different size, i.e. 1.1 kb, 1.3 kb, and 5.8 kb. Most of the lines of T5 generation have one insertion site, suggesting that the three insertion sites were located in different loci. Based on the phenotypic data, the transgenes of 1.1 kb and 1.3 kb were functional, resulting in resistant or recovery phenotype, while that of 5.8 kb was not functional. Copy number apparently had no effects on the phenotypes.

Abstract

Early Identification of Sclerotium rolfsii Resistant Lines of Peanut Derived from In Vitro Selection Using S. rolfsii Culture Filtrates. The objectives of this experiment was to evaluate methods for early identification of Sclerotium rolfsii resistant lines derived from fungal culture filtrate (CF)-insensitive somatic embryos (SE) and to correlate the results of early identification with that of direct S. rolfsii inoculation in the plastic house. In the first method, shoots regenerated from CF-insensitive SE were planted onto selective medium containing 30% of S. rolfsii CF for one month and then responses were evaluated. In the second method, leaves and leaflets of R0 peanut lines derived from CF-insensitive SE were inoculated with S. rolfsii by detached leaf-dual culture test. The damages of the inoculated leaves and leaflets were assessed 6 days after inoculation. A number of R1 plants were grown in the plastic house from seeds harvested from R0 peanut lines, and they were inoculated with S. rolfsii at 30 days after planting (dap). Responses of the inoculated R1 plants were observed at 44 dap. Results of the experiment indicated that shoots from CF-insensitive SE showed less damage scores (DS=1.2), when cultured on CF-containing selective medium than ones from non-selected SE (DS=2.9). Most of the detached leaves and leaflets inoculated with S. rolfsii totally necrosed. However, leaves and leaflets from 7 out of 23 R0 lines showed variegation between necrosed and healthy tissues, indicating the presence of S. rolfsii resistant tissue among the tested leaves and leaflets. Direct inoculation of R1 plants in the plastic house showed 10 out of 18 progenies derived from 6 R0 lines showed less disease severity scores (DSS=2 or 3) and they were able to survive and produce seeds. On the other hand, seed derived (original) peanut plants cv. Kelinci inoculated with S. rolfsii all showed DSS=5 and died after inoculation. The overall results of the experiment indicated the usefullness of in vitro selection using S. rolfsii culture filtrates for regenerating S. rolfsii resistant peanut variants and the possible use of methods for early identification of desirable variants.

Abstract

Peanut somaclonal variants resistant to Sclerotium rolfsii derived from in vitro selection with fungal culture filtrates. Sclerotium stem rot is one of the most important peanut disease which often caused significant yield loss. The use of peanut cultivars resistant to Sclerotium rolfsii infection is the most efficient way to control the disease. Attempts to obtain peanut lines with tolerance or resistance to Sclerotium stem rot through induction of somaclonal variation and in vitro selection using fungal culture filtrates (CF) have been conducted previously. Somatic embryo (SE) clumps that had been maitained in culture for approximately one year in regeneration medium were exposed onto selective medium containing 30% S. rolfsii CF for three consecutive 1 month –passages, and the SE formed after selection periods were considered to be insensitive to the fungal CF. A number R0 peanut lines have been regenerated from CF-insensitive SE, and their R1 and R2 progenies were grown in a plastic house for evaluation on qualitative and quantitative variant characters. Early identification for tolerance to S. rolfsii was also conducted among R0 peanut lines, and the results have shown enhanced resistance when compared to the original non-selected cultivar. However, further evaluation is needed to study responses of the R1 and R2 progenies of the somaclones against S. rolfsii infection. The objective of this particular study was to evaluate responses of R1 and R2 peanut somaclones derived from fungal CF-insensitif SE against S. rolfsii infection in the plastic house. Results of this experiment showed, a number of resistant somaclonal variants were obtained among R1 and R2 population, which segregated as resistance and susceptible to S. rolfsii infection. The resistance somaclonal variants initially showed stem rot symptoms after inoculation with S. rolfsii with disease score (DS) of 1 to 3. However, as the plants grew and developed, they showed wound-healing process at the lesio and they were able to produce the same or higher number of filled pod as the original non-inoculated peanut plants.

Abstract

Inheritance of Resistance to PStV in Transgenic Peanuts Containing cp PStV Gene. We have obtained transgenic peanut lines containing coat protein gene of PStV. To get maximal use of the transgenic character in a breeding program, it is necessary that the transgene is also stably inherited and expressed. This experiment was conducted from June 2002 – January 2005 at Plant Molecular Biology Laboratory, Bogor Agricultural University, and Queensland Agricultural Biotechnology Center, The University of Queensland, Australia. The research aimed (1) to test whether PStV cp transgene was functional in progenies derived from crosses between transgenic peaanut plants containing PStV cp gene and non-transgenic ones and (2) to determine pattern of inheritance of resistance to PStV as a result of PStV cp gene action. Several crosses were made between trangenic peanut cv.Gajah resistant to PStV (T4 generation) and non-transgenic peanut line WS susceptible to PStV. The F1 and F2 populations were mechanically inoculated with PStV two weeks after planting. The experiment showed that all plants in the F1 population were less susceptible to PStV, suggesting that the transgene was partially dominant. Phenotipic segregation in F2 population was not Mendelian with the appearance of quick and slow recovery plants and the number of resistant plants being more than expected. However, the proportion of transgenic and non-transgenic plants followed 3:1 ratio, which was Mendelian.

Abstract

The objective of this study was to analyze the relationship between working burden with time and motion study. The research’s design was used cross sectional studies with 56 samples of nurse which are taken by using purposive sampling technique. Data were collected for working burden by using observational with time and motion study, for nurses motivation and nursing care quality by using questioners with multiple choices. Data were analyzed by using Spearman’s Rho test with value level of  p<0.05. The result showed that there were no relationship between working burden and nursing care quality with value level of  p>0.05.

Abstract

The objectives of the experiment were to evaluate effectiveness of seed treatments to eliminate Cmm of infected tomato seed lot, evaluate physiological quality of Cmm infected tomato seed lot after treatments with plant extracts. The research used curcuma rhizomes, betel vine leaf extract, cinnamon and clove oil to eliminate Cmm in tomato seed. Part of the seeds were dipped in either suspension of curcuma rhizomes, betel vine leaf extract, cinnamon and clove oil for 20 minutes and the others were matriconditioned using a mixture of burned rice hull (at 22 0C and RH 60-70%) plus either of plant extracts or oils. Elimination of Cmm from infected seeds was observed 10 days after treatments, and physiological seed qualities were evaluated before and after seed treatments. Results of the experiment indicated, dipping infected seeds in either 5% of curcuma, betel vine extract, cinnamon oil or 0.5% of clove oil suspension or matriconditioning plus either these extracts, eliminated 99% of infected Cmm on tomato seeds. Seed treatments using plant extracts for elimination of Cmm did not reduce seed germination, germination rate, vigor index, and did not increase time to reach 50% total germination (T50) of infected tomato seed except the seed treatment by using 5% cinnamon oil with or without matriconditioning.Key words: plant extracts, physiological quality, seed viability, vigor

Abstract

Tawas ut tuber (Ampelocissus rubiginosa Lauterb.) contains of flavonoid, tanin, compound and saponin which is potentially anti-inflammatory. This objective of this research is to prove by effect giving extrak ethanol tawas ut tuber about effect anti-inflammatory topical based on consideration the fold of the skin and amount of cell neutrofil to mice which induced by carageenan. Thirty mice ware divided into 6 groups. They were normal control group (palacebo gel), positive control (Voltaren®), negative control (karagenin), and testing groups with gel extract concentrations of 0,5%, 1%, and 2%. Ethanol extract of Tawas ut tuber leaf applied, after back of test animals was induced by 0,2 ml of 3% carageenan, then every hour was measured for middosal skin fold thickness during 6 hours observation. The animal’s skin was cut off to perform test at 24 hours after treatment. The neutrophils migration was observed using hematoxylin and eosin (HE) stining method. The percentage result of inhibition (%PI) ethanol extrak of tawas ut tuber with concentration 0,5%, 1%, and 2% were 20,00%; 23,51%; and 24,92% respetively. The average result of netrofil total from ethanol extrak of tawas ut tuber with concentration 0,5%, 1% and 2% were 28,28; 20,88; and 16,24 respecively. Gel 2% posesses the most effective topical anti-inflammatory. Based on the result, it can be concluded that ethanol extrak of tawas ut tuber posesses anti-inflammatory in skin oedema of back of mice induced by carageenan.

Abstract

Quantitative analysis of zeatin is needed to determine correlation between callus and embryo formation with the presence of zeatin itself on explant and interaction between zeatin content on explant and synthetic plant growth regulators (PGRs) which is added to oil palm in vitro culture media. This research was conducted to obtain method of zeatin content analysis that is easy and fast to reproduce, so it can be applied to research studies on phytohormones, especially zeatin. The analysis was carried out using ultra high performance liquid chromatography (UHPLC) with C18 column and isocratically 100% methanol mobile phase and 0.2 mL / minute flow rate with UV detector at length λ = 280 nm. Analyzed explants were taken from individuals representing La Me, Avros, PPKS 239 and Simalungun varieties. Results of system suitability test indicated that the method fulfilled its requirements to be used for analysis with relative standard deviation (RSD) value was ≤ 2. Method validation gaveresults of calibration curve with the coefficient of determination r2 = 0,9996 and precision of instrument and method obtained relative standard deviation by 0,56 and 1,03, respectively. LoD and LoQ values ​​obtained were 2,457 ppm and 7,446 ppm respectively.

Abstract

Detecting the availability of sufficient water in tidal land management is important for agriculture. Tidal flood pattern provides an information of area where has tidal flood as well as drought by time and places. The objectives of this research are 1) to study the tidal flood spatial pattern by using real time data and digital data image processing, 2) to determine a tidal flood classification based on ground water level on a specific locationand time, and 3) to study the hydrological factors affecting the flood. The tidal flood pattern can be classified within a block by their ground water fluctuation. The visual analysis with four image of different time, gives a clear differences between tidal flood pattern change and tidal fluctuation. The classification on the moist Landsat TM image after a Tesseled Cap transformation can produce classified image with similar flooded condition with the same range of ground water level. After the test, the tidalflooded pattern was found similar with the classification results. The tidal flood pattern is also recognized having a specific relationship with land use or land utilization type. The land use and land management affect the pattern. The application of geographical information system, especially remote sensing digital image data analysis, will help in determining tidal flood pattern as well as the ground water spatial pattern.
