Abstract

Identifi kasi ketahanan genotipe adalah langkah awal dalam pengembangan kultivar tahan terhadap serangan patogen. Tujuan penelitian ini adalah (i) mendapatkan cara inokulasi dengan jumlah dan konsentrasi inokulum Cmm yang efektif untuk mengevaluasi ketahanan tomat terhadap Cmm di rumah kaca, (ii)  mendeterminasi reaksi ketahanan berbagai genotipe tomat akibat inokulasi Cmm. Percobaan ini menggunakan 38 genotipe tomat yang terdiri dari 7 genotipe tomat lokal, 15 genotipe tomat komersial, dan 16 genotipe koleksi Pusat Studi Pemuliaan Tanaman IPB Bogor (PSPT/IPB). Agen penyebab penyakit yang digunakan adalah 6 isolat Cmm hasil percobaan sebelumnya. Cara inokulasi Cmm yang efektif terhadap tomat cv. Marta (sangat rentan),  uji reaksi ketahanan berbagai genotipe tomat terhadap Cmm telah dilakukan di rumah kaca.  Kesimpulan dari penelitian ini adalah (i) inokulasi dengan menyuntikkan inokulum Cmm 5 µl konsentrasi 106 cfu/ml pada beberapa tempat di ketiak daun (daun pertama, daun tengah dan pucuk) merupakan cara yang paling efektif mengevaluasi ketahanan tomat terhadap Cmm, (ii) berbagai genotipe tomat yang diuji belum ada yang tahan teradap Cmm, genotipe tomat lokal ada yang agak rentan dan agak tahan.Kata kunci: kultivar tahan, suspensi bakteri, tomat

Abstract

Chitinase and peroxydase are important bioactive proteins or are specific enzymes that are related to plant resistance to pathogens. The aims of the research were to analyze the chitinase and peroxidase activities of crude protein extract from calli, stem, leaves and roots of T. cucumerinavar. anguina. In the first experiment, chitinase and peroxydase activities were analyzed from in vitro calli, leaves and roots obtained from 2-month-old of field grown plants. The media for calli induction were Murashige and Skoog medium with addition of 1 µM NAA + 1 µM BA, 2 µM NAA + 2 µM BA, 3 µM NAA + 3 µM BA, or 4 µM NAA + 4 µM BA. In the second experiment, the chitinase and peroxydase activities from crude protein extract of roots, stems and leaves were analyzed. The extracts were from 3-week-old seedling (less than a month), 1-month and 2-month-old plants. The first and the second experiment results showed that crude protein extracts of plant roots from the field grown plants had the highest chitinase and peroxidase activities. Stem of field grown plants had the similar level of chitinase activities with the plant roots. Chitinase activities of in vitro calli were not significantly different from those of plant roots so that it could be used as an alternative for plant roots in studying chitinase from T. cucumerinavar. anguina. Chitinase activities in crude protein extracts of roots appeared constant whereas peroxidase tend to increase with plant age. Keywords: calli, enzyme activities, leaves, roots, stem

Abstract

The objective of the study was to determine the effect of seed treatment on growth, seed quality, P fertilizer utilization, and rice production in the glasshouse. The experiment was conducted in a split plot design with three replications from January to June 2010.The main plots were P fertilizer rates (0, 50, and 100 kg SP-36 ha-1), and sub plots were (1) negative control (seeds were not inoculated by Xanthomonas oryzaepv. oryzae(Xoo) and without seed treatment); (2) positive control (seeds were inoculated by Xoo); (3) seeds were inoculated by Xoo and immersed in Pseudomonas diminutaA6 suspension; (4) seeds were inoculated by Xoo and immersed in Bacillus subtilis5/B suspension; (5) seeds were inoculated by Xoo and immersed in P. diminutaA6 + B. subtilis5/B suspension; (6) matriconditioning + P. diminutaA6, (7) matriconditioning + B. subtilis5/B, and (8) matriconditioning + P. diminutaA6 + B. subtilis5/B. The results showed that seed treatment with biological agents P. diminutaA6, seed treatment with P. diminutaA6 + B. subtilis5/B were the best seed treatments to improve plant growth and yield of rice. Treatment with matriconditioning + P. diminutaA6 + B. subtilis5/B was the best treatment to enhance seed germination. Soaking the seeds in B. subtilis5/B suspension or P. diminutaA6 + B. subtilis5/B suspension reduced the use of P fertilizer, based on the number of grain, total number of grain, percentage of grain per panicle. Abstract

Bacterial leaf blight, the disease caused by Xanthomonas oryzae pv. oryzae (Xoo), a seedborne pathogen, has been known to reduce rice yield by 50%. In this study, two consecutive experiments were conducted in order to investigate the effect of various biological seed treatments applied on rice seeds artifi cially infected by Xoo on seed quality and seedling growth. Laboratory experiment was arranged in a completely randomized design while greenhouse experiment was arranged in a completely randomized block design. In both experiments, 12 seed treatments were applied: un-infected seeds (negative control) without seed treatment, Xoo infected seeds (positive control) without seed treatment, infected seeds soaked in bactericide Agrept 0.2%, infected seeds soaked in biological agent suspension isolate A6, infected seeds soaked in A54 isolate, infected seeds soaked in 5/B isolate, infected seeds soaked in 11/C isolate, infected seeds matriconditioned + A6 isolate, infected seeds matriconditioned + A54 isolate, infected seeds matriconditioned + 5/B isolate, and infected seeds matriconditioned + 11/C isolate. Soaking seeds either in Agrept 0.2% or in biological agent suspension was conducted for 30 h. Matriconditioning was conducted using ratio of seeds to carrier (burned rice hull 32 mesh) to biological agent suspension or bactericide solution of 1.0 (g) :0.8 (g): 1.2 (mL) for 30 h in air-conditioned room ca. 25 oC. Results of experiments showed that all biological seed treatments could suppress Xoo in rice seeds. Matriconditioning plus biological agent (isolate A6), biopriming with isolate A6 or isolate A54 were the best seed treatments to improve seed viability and vigor. In greenhouse experiment, matriconditioning plus isolate A54 was the best seed treatment to increase seedling growth.Keywords: biopriming, matriconditioning, rhizobacteria, seed health, viability

Abstract

Wortel (Daucus carota L.) merupakan komoditas sayur yang dikenal secara umum oleh masyarakat. Kualitas wortel yang baik diharapkan mampu meningkatkan konsumsi masyarakat. Salah satu cara meningkatkan kualitas wortel adalah meningkatkan rasa manis dengan cara meningkatkan dosis pupuk KCl yang diberikan. Penelitian bertujuan menguji hubungan dosis pupuk KCl terhadap tingkat kemanisan wortel. Penelitian dilaksanakan pada bulan Mei hingga Agustus 2019 di Kebun Percobaan Pasir Sarongge IPB, Kecamatan Pacet, Kabupaten Cianjur dan Laboratorium Pascapanen Departemen Agronomi dan Hortikultura IPB. Rancangan percobaan yang digunakan adalah rancangan petak terbagi dengan dua faktor. Faktor pertama adalah varietas (Kuroda dan Cipanas) sebagai petak utama dan faktor kedua adalah dosis pupuk KCl (60, 90, 120, dan 150 kg ha-1) sebagai anak petak. Hasil penelitian menunjukkan bahwa perlakuan varietas berbeda nyata pada semua parameter, kecuali daya berkecambah dan diameter umbi. Perlakuan dosis pupuk KCl pada tanaman wortel tidak meningkatkan kadar padatan terlarut total. Wortel Kuroda memberikan hasil terbaik pada uji tingkat kemanisan, sedangkan wortel Cipanas memberikan hasil terbaik pada produksi dan produktivitas panen. Karakter hasil tanaman yang berkorelasi nyata secara positif terhadap masing-masing karakter adalah produksi, produktivitas, panjang umbi, dan bobot umbi. Kadar padatan terlarut total (PTT) berkorelasi nyata secara negatif terhadap karakter produksi, produktivitas, panjang umbi dan tinggi tanaman. Abstract

VO2max is a component that affects the performance of a soccer player on the field. Research was conducted to examine the extent to which SIT can provide significant benefits in increasing VO2max for soccer players. The study used a quasi-experiment with a quantitative approach. The samples involved were Persikab Bandung soccer players in 2023 as many as 18 players who were given treatment in the form of Sprint Interval Training (SIT). The instrument uses the MFT (Multistage Fitness Test) which is used to measure the player's VO2max. The results showed that the average VO2max before treatment was 48,678 ± 7,883, and the average VO2max after training was 51,417 ± 4,944. Based on the results of the analysis, it is concluded that Sprint Interval Training (SIT) can effectively increase the VO2max capacity of soccer players. The results of this study are expected to contribute to the development of football training programs.

Abstract

Penelitian ini bertujuan untuk (1) Mengetahui Dampak Positif minimarket terhadap pola konsumsi masyarakat lokal di Kabupaten Barru (2) Mengetahui dampak negatif minimarket misi pasar raya terhadap pola konsumsi masyarakat lokal di Kabupaten Barru. Jenis penelitian ini adalah penelitian kualitatif deskriptif, dengan lokasi penelitian pada Misi pasar raya di kecamatan tanete rilau kabupaten barru. Penentuan informan secara purposive sampling yaitu teknik yang mencakup orang-orang yang diseleksi atas dasar kriteria, sedangkan orang-orang dalam populasi yang tidak sesuai dengan kriteria tersebut tidak dijadikan sampel. Metode yang digunakan dalam mengumpulkan data wawancra, observasi, dan dokumentasi. Metode analisis data yang menggunakan tiga tahap yaitu analisis reduksi data, penyajian data dan verifikasi data. Hasil penelitian menunjukan bahwa pola konsumsi masyarakat lokal Tananete rilau dengan hadirnya minimarket. Dampak sosial yang terjadi pada Masyarakat dengan hadirnya minimarket berefek pada proses transaksi, pola konsumsi Masyarakat berkembang dan menunjukan perilaku konsumtif yang lebih memilih berbelanja pada minimarket, peran pemerintah sangat penting untuk meminimalisir persaingan dagang pasar tradisional dan modern. Dampak Negatif persaingan warung kecil dengan Keberadaan minimarket menjadi ancaman serta saingan yang berat bagi para pemilik warung kecil, Pasar modern (minimarket) merupakan pesaing dan akan mengancam keberadaan pedagang di pasar tradisional. penyebabnya antara lain perubahan gaya hidup Masyarakat, harga, kondisi pasar tradisional. Abstract

Transcription factors have an important function in regulating gene expression and plant responses to stresses. The ERF, bZIP, WRKY, MYB, and NAC are stress inducible transcription factors. The OsNAC6 is a member of the NAC transcription factor family in rice and its expression is induced by abiotic stresses, wounding and blast disease. Characterization of OsNAC6 gene sequences would give a better understanding on how OsNAC gene functions biologically. The objectives of this research are to isolate the OsNAC6 cDNA from Nipponbare, Batutegi, and Rojolele cultivars, to characterize their DNA sequences, and to compare their sequences to other NAC genes from other plants available in GenBank DNA databases. Isolated cDNA and sequencing of the fragments resulted in a 912 bp DNA sequences. Translation of the sequences yielded a protein consisted of 303 amino acid residue. Blast analysis of amino acid sequences indicated identity of isolated cDNA from three Indonesian rice cultivars are the OsNAC6 gene. Deduced amino acid residues from amplified cDNAs of Nipponbare, Batutegi, and Rojolele cultivars shared 100% sequence identities to rice OsNAC6 (Acc. # BAA89800), 71-100% sequence identity to a number of OsNAC protein from Oryza sativa and 63-83% sequence identity to NAC protein from other plants.

Abstract

Abstract: Phalaenopsis orchids are one of the ornamental plants that are very popular with the public because they have attractive flower colors and shapes. One of the obstacles in the success of orchid cultivation is the attack of soft rot disease caused by Dickeya dadantii. Therefore, efforts are needed to find Phalaenopsis clones that are resistant to soft rot disease. This study aims to identify the resistance response of the hybrid orchid genotype Phal. Salu Spot Phal. bellina no. 1102-44. The plant material used was the hybrid orchid Phal. Salu Spot Phal. bellina no. 1102-44 inoculated with Dickeya dadantii bacteria. Identification of the resistance response was carried out through the infection method using the leaf disk technique. The results showed that the clone Phal. Salu Spot Phal. bellina no. 1102-44 was successfully grouped into five resistance classes at 24 JSI (hours after inoculation). A total of three accessions were identified as having resistance to soft rot disease, with two accessions originating from the 22.19 M media treatment (T2-15 and T2-17) and one other accession originating from the 44.39 M media treatment (T4-23).Keywords: orchid, Dickeya dadantii, plant resistance, leaf disk technique Abstrak: Anggrek Phalaenopsis merupakan salah satu tanaman hias yang sangat diminati masyarakat karena memiliki warna dan bentuk bunga yang menarik. Salah satu hambatan dalam keberhasilan budidaya tanaman anggrek adalah serangan penyakit busuk lunak yang disebabkan oleh Dickeya dadantii. Oleh karena itu, diperlukan upaya untuk menemukan klon Phalaenopsis yang memiliki ketahanan terhadap penyakit busuk lunak. Penelitian ini bertujuan mengidentifikasi respons ketahanan genotipe anggrek hibrida Phal. Salu Spot Phal. bellina no. 1102-44. Bahan tanaman yang digunakan berupa anggrek hibrida Phal. Salu Spot Phal. bellina no. 1102-44 yang diinokulasi dengan bakteri Dickeya dadantii. Identifikasi respons ketahanan dilakukan melalui metode infeksi menggunakan teknik leaf disk. Hasil penelitian menunjukkan bahwa klon Phal. Salu Spot Phal. bellina no. 1102-44 berhasil dikelompokkan ke dalam lima kelas ketahanan pada 24 JSI (jam setelah inokulasi). Sebanyak tiga aksesi teridentifikasi memiliki ketahanan terhadap penyakit busuk lunak, dengan dua aksesi berasal dari perlakuan media 22,19 M (T2-15 dan T2-17) dan satu aksesi lainnya berasal dari perlakuan media 44,39 M (T4-23).Kata kunci: Anggrek, Dickeya dadantii, ketahanan tanaman, tekhnik leaf disk

Abstract

Penelitian ini mengkaji strategi Kepala Madrasah MTs Miftahul ’Ulum Denpasar dalam merancang strategi yang efektif guna meningkatkan kinerja pendidik agar proses pembelajaran berjalan profesional dan bermakna. Tujuannya: (1) mengetahui strategi kepala madrasah dalam meningkatkan kinerja pendidik di MTs Miftahul ’Ulum Denpasar, (2) menelaah sejauh mana strategi tersebut berdampak pada peningkatan kinerja pendidik, dan (3) mengidentifikasi faktor penghambat serta solusi yang ditempuh. Metode yang digunakan adalah kualitatif deskriptif naratif melalui observasi, wawancara, dan dokumentasi, dengan triangulasi untuk uji keabsahan data. Hasil penelitian menunjukkan: (1)    Strategi kepala madrasah dilakukan melalui pemberian motivasi, keteladanan, pelatihan, serta komunikasi harmonis. (2) Strategi ini terbukti meningkatkan variasi metode pembelajaran, keterlibatan guru dalam kegiatan sekolah, serta hasil belajar siswa. (3) Hambatan yang muncul antara lain keterbatasan waktu guru dan kurangnya proaktivitas sebagian pendidik. Solusi yang ditempuh berupa pembinaan berkelanjutan dan dorongan untuk terus berinovasi. Dengan demikian, strategi kepala madrasah berkontribusi positif dalam meningkatkan kinerja pendidik dan mutu pembelajaran di madrasah.
