Abstract

The coastal waters of Semarang Bay have been polluted by heavy metal lead (Pb). The research was conducted from September 2015 to March 2016 with 3 stations at the Tambaklorok green mussel cultivation location. Analysis of the concentration of heavy metal Pb using the AAS (Atomic Absorption Spectrophotometer) method. Data processing using SPSS and Excel. The results showed that the average concentration of heavy metal Pb in the air ranged from 0.51 to 0.76 mg/L. The average concentration of heavy metal Pb in three observations obtained an average concentration of Pb between 4.975-20.43 mg/kg. Green mussels taken in the waters were sorted and separated according to the length of the shell and weighed 1 kg each according to the length of the shell. Small (2-3 cm), medium (3-4 cm) and large shells with long shells (5-6 cm). The results of the measurement of the concentration of heavy metal Pb in P. viridis showed that the average green mussel shell size 4-5 cm at the location of station III had the lowest concentration of heavy metal Pb at 0.24 mg/kg. Meanwhile, the highest average concentration of Pb in P. viridis was found at station I at 0.89 mg/kg with a shell length of 2-3 cm. The correlation equation between shell length and Pb concentration is Y= - 0.131 + 1.13 X. Standard error of the Estimate (SEE) is 0.19. While the standard deviation of 0.20 means that the predictor variable (water) to predict the dependent variable (shellfish) is feasible because SEE is greater than the standard deviation. The average value of the Bioconcentration Factor (BCF) of green mussels is smaller than that of medium-sized green mussels and large green mussels. Abstract

Udang merupakan sumber protein hewani yang banyak diminati masyarakat karena citarasa enak dan bergizi. Udang merupakan komoditas perikanan ekspor. Upaya pemenuhan kebutuhan ekspor udang saat ini telah dilakukan budidaya intensif. Budidaya intensif telah terjadi pemberian pakan yang berlebih sehingga terbentuk limbah organik di dasar tambak. Limbah pakan mengakibatkan terbentuknya senyawa nitrit dan amoniak yang toksik bagi udang. Komponen utama limbah ini adalah protein. Protein harus di dekomposisi menjadi asam amino oleh bakteri proteolitik. Salah satu bakteri yang memiliki aktivitas proteolitik adalah bakteri Bacillus Firmus dari simbion sponge Chalinula Pseudomolitba. Tujuan penelitian adalah melakukan optimasi substrat dan pemilihan co-substrat media kultur yang sesuai terhadap pertumbuhan dan aktivitas protease ektraseluler Bacillus Firmus. Penelitian menggunakan metode eksperimen laboratoris. Penelitian terdiri dari uji optimasi substrat, pemilihan Co-substrat C dan N. Hasil penelitian menunjukkan bahwa bakteri Bacillus firmus memiliki laju pertumbuah dan aktivitas protease terbaik pada media kultur dengan subtrat susu skim 1% dengan nilai 0,110 c/jamdan 12,566 c IU/ml. Sumber C terbaik adalah glukosa dengan nilai 0,124 c/jamdan 16,094 cIU/ml. Sumber N terbaik adalah amonium nitrat dengan nilai sebesar 0,137 c/jamdan 19,046 c IU/ml. Shrimp is a source of protein and have good taste. Shrimp is an export commodity. Shrimp cultivation is developing to be intensive. Intensive cultivation uses large amounts of feed. not all of the feed is eaten. leftover feed becomes waste. Feed waste produces nitrite and ammonia. Ammonia is toxic. Feed waste must be cleaned. Decomposition process by proteolytic bacteria. Bacillus Firmus is a proteolytic bacterium from the symbiont sponge Chalinula Pseudomolitba. The research aims to optimize the substrate and select the co-substrate. The study used a laboratory experimental method. The research consisted of a substrate optimization test, selection of Co-substrate C and N. The results showed that Bacillus Firmus had the best growth rate and protease activity on 1% skimmed milk substrate with values of 0.110 c/hour and 12,566 c IU/ml. The best source of C is glucose with a value of 0.124 c/hour and 16,094 c IU/ml. The best source of N as ammonium nitrate with values of 0.137 c/hour and 19,046 c IU/ml.

Abstract

Tujuan penelitian ini untuk menganalisis faktor yang mempengaruhi kinerja bidan dalam meningkatkan pelayanan antenatal care di puskesmas kabupaten Halmahera Tengah. Metode penelitian ini bersifat survei analitik dengan rancangan penelitan cross sectional yaitu suatu penelitian dimana variabel-variabel yang termasuk faktor resiko dan variabelvariabel yang termasuk efek di observasi sekaligus pada waktu yang sama dengan instrument penelitian yaitu kuesioner. Hasil penelitian ini adalah tidak ada hubungan antara masa kerja dengan kinerja bidan. Dengan nilai p = 1,000 > 0,05 yang artinya Ha ditolak. Tidak ada hubungan antara kemampuan dan keterampilan dengan kinerja bidan dengan nilai p = 0,003 0,05 yang artinya Ha ditolak. Ada hubungan antara imbalan dengan kinerja bidan dengan nilai p = 0,037 < 0,05 yang artinya Ha diterima. Kata Kunci: Kinerja Bidan, Antenatal Care

Abstract

Perkembangan budidaya udang secara intensif, diikuti pula oleh permasalahan limbah organik sisa pakan. Salah satu cara untuk mengatasi permasalahan limbah organik adalah dengan bioremediasi menggunakan probiotik. Komponen utama limbah organik pakan ini adalah protein. Oleh sebab itu, konsorsium probiotik merupakan bakteri proteolitik. Bakteri Bacillus firmus yang bersimbiosis dengan sponge Chalinula pseudomolitba merupakan bakteri yang menghasilkan enzim protease ektraseluler yang dapat menghidrolisis protein menjadi senyawa yang sederhana. Bakteri Bacillus firmus memiliki potensi untuk dijadikan probiotik. Diperlukan informasi tentang kondisi suhu dan agitasi inkubasi yang optimal untuk mendapatkan biomassa sel bakteri Bacillus firmus dalam pembentukan probiotik. Tujuan penelitian ini adalah melakukan uji optimasi suhu dan agitasi dalam inkubasi kultur terhadap pertumbuahn dan aktivitas protease ekstraseluler bakteri Bacillus firmus. Penelitian menggunakan metode eksperimen laboratoris. Penelitian telah berhasil melakukan uji optimasi suhu dan agitasi inkubasi kultur. Hasil penelitian menunjukkan bahwa bakteri Bacillus firmus memiliki laju pertumbuah dan aktivitas protease terbaik pada kondisi kultur dengan suhu inkubasi 30 oC dengan nilai sebesar 0,194 c dan 34,660 d IU/ml. Selain itu juga memiliki laju pertumbuah dan aktivitas protease terbaik pada kondisi agitasi inkubasi 150 rpm dengan nilai sebesar 0,124 b dan 40,039 b IU/ml.The development of intensive shrimp farming is also followed by the problem of organic waste from feed residues. One way to overcome the problem of organic waste is bioremediation using probiotics. The main component of this feed organic waste is protein. Therefore, the probiotic consortium is a proteolytic bacteria. Bacillus firmus bacteria in symbiosis with Chalinula pseudomolitba sponge is a bacterium that produces extracellular protease enzymes that can hydrolyze proteins into simple compounds. Bacillus firmus bacteria have the potential to be used as probiotics. Information about optimal temperature conditions and incubation agitation is needed to obtain cell biomass of Bacillus firmus bacteria in the formation of probiotics. The purpose of this study was to test the optimization of temperature and agitation in culture incubation on the growth and activity of extracellular proteases of Bacillus firmus bacteria. The study used a laboratory experimental method. Research has successfully carried out the optimization test of culture incubation temperature and agitation. The results showed that Bacillus firmus had the best growth rate and protease activity in culture conditions with an incubation temperature of 30 oC with values of 0.194 c and 34.660 d IU/ml. In addition, it also has the best growth rate and protease activity at 150 rpm agitated incubation conditions with values of 0.124 b and 40.039 b IU/ml.

Abstract

In accodance with social culture in Central Sulawesi which are various, it will need adaptive development more specially. Therefore, the problem that will be investigated in this research is: “How the system (way) development of KTSP in central Sulawesi?”.  From the main problem mentioned, this study will focus on analysis to some aspects, namely: (1) How the direction of policy in implementation KTSP in central Sulawesi?, (2) How the understanding of the headmasters, teachers, schoo committee for development of KTSP?, (3) What problems faced by schools in developing KTSP?. The technique used in gaining data are: (1) Study document, (2) Observation, (3) Interview and (4) Quistionnaire. The result of research indicated local superiority that wants to be developed by Dinas Pendidikan Kota and Kabupaten are: Local Language, local arts and local handicraft. Implementation of KTSP has been implemented since 2006/2007 academic year. The headmaster’s understanding of KTSP guidebook that printed by BNSP is very low. Therefore, most of headmaster and teachers face problems in implementing KTSP. The problem is: The teachers have difficulties in developing silabus material and lesson plan in accodance local condition. The problem that is faced in developing local load is; there is no teacher who understands social culture condition in central Sulawesi. There are some suggestions recommended: The headmaster and the teachers need to be trained, to develop silabus material and lesson plan. “Pusat Kurikulum Balitbang Depdiknas” needs to socialize guidebook in developing curriculum. In developing curriculum, the member team from university should be included to participate according to their subjects.

Abstract

ABSTRAK Waduk Jatibarang terletak di Kelurahan Kandri, Kecamatan Gunung Pati, Kota Semarang resmi dioperasikan pada tahun 2014. Pemanfaatan Waduk Jatibarang sebagai penyedia air baku, tempat pariwisata dan kegiatan perikanan. Pemanfaatan waduk dapat menurunkan kualitas air. Tujuan dari penelitian ini untuk mengetahui status pencemaran perairan Waduk Jatibarang berdasarkan berbagai kegiatan peruntukan (kegiatan pariwisata, perikanan dan penyediaan air baku). Penelitian dilaksanakan pada bulan Januari - Pebruari 2019. Metode yang digunakan dalam penelitian ini adalah metode survei. Pengambilan contoh air menggunakan metode purposive sampling, dilakukan 2 (dua) kali pengambilan dengan rentang waktu 1 bulan. Penelitian dilakukan dengan mengukur variabel kualitas air yang terdapat pada PP Nomor 82 Tahun 2001 tentang Pengelolaan Kualitas Air dan Pengendalian Pencemaran Air, antara lain Temperatur, TSS, pH, DO, BOD, COD, Amonia dan Total Coliform. Selanjutnya di analisis dengan metode Indeks Pencemaran sesuai KepMen LH Nomor 115 Tahun 2003 tentang Pedoman Penentuan Status Mutu Air. Perairan Waduk Jatibarang memiliki temperatur berkisar antara 30 - 31◦C, TSS 20 - 80 mg/l, pH netral, DO 6,8 - 8,6 mg/l, BOD 1,46 - 3,95 mg/l, COD 9,7 - 15,69 mg/l, Amonia 0,110 - 0,566 mg/l dan Total Coliform 15 – 4.600 sel/100 ml. Status pencemaran pada dermaga Waduk Jatibarang (kegiatan pariwisata) tercemar ringan – sedang (IP = 3,62 - 5,49). Inlet Waduk Jatibarang (Kegiatan Perikanan) tercemar ringan – sedang (IP = 2,88 – 5,93). Outlet Waduk Jatibarang (penyediaan air baku) tercemar ringan (IP = 1,43 – 1,84). ABSTRACT Jatibarang Reservoir is located in the Village Kandri, District Gunung Pati, Semarang officially operated in 2014. Utilization of Jatibarang Reservoir as a provider of raw water, tourism sites and fisheries activities. Utilization of reservoirs can reduce water quality. The purpose of research to determine the water pollution status of Jatibarang Reservoir in based on the various activities allocation (tourism, fisheries and raw water supply). Research was conducted on January - February 2019. The method used is a survey method. Water sampling using purposive sampling with two replicate and sampling interval 1 month. The research was conducted by measuring the water quality variables by activities allocation according with Government Regulation Number 82 of 2001 about Management of Water Quality and Water Pollution Control, among others temperature, TSS, pH, DO, BOD, COD, ammonia and Total Coliform. Furthermore, analized with Pollution Index according to the decree of the minister of environment of the republic of indonesia number 115 year 2003 regarding Guidelines for Determination of Water Quality Status. Jatibarang Reservoir waters have temperature ranging between 30 - 31◦C, TSS 20 - 80 mg/l, pH neutral, DO 6,8 – 8,6 mg/l, BOD 1,46 – 3,95 mg/l, COD 9,7 - 15,69 mg/l, Ammonia 0,110 – 0,566 mg/l and Total Coliform 15 – 4.600 cells/100 ml. Pollution status of the dock Jatibarang Reservoir (tourism activity) lightly - moderatly polluted (IP = 3,62 – 5,49). Abstract

Instalasi Pengolahan Lumpur Tinja (IPLT) Tanggung Rejo berada bersebelahan langsung dengan Muara Sungai Tambak Rejo, Kota Semarang. Adanya masukan air limbah IPLT Tanggung Rejo ke perairan berpotensi adanya kandungan amonia, nitrit dan jumlah bakteri Escherichia coli yang berlebih. Tujuan penelitian ini untuk mengetahui konsentrasi dan peta sebaran dari amonia, nitrit, dan kelimpahan E. coli pada Muara Sungai Tambak Rejo Kota Semarang. Waktu penelitian dilakukan pada bulan November hingga Desember 2017. Metode yang digunakan dalam penelitian ini adalah metode survei dan teknik pengambilan sampel menggunakan metode purposive sampling pada 4 stasiun. Analisis sebaran spasial dengan menggunakan metode IDW. Hasil pengukuran menunjukkan bahwa indikator pencemaran perairan amonia berkisar antara 0,3 – 7,31 mg/L, nitrit berkisar antara 0,004 – 0,144 mg/L, dan jumlah E. coli berkisar antara 1 – 270000 MPN, dimana nilai amonia, nitrit dan E. coli tersebut diatas nilai baku mutu.  The installation of the Fecal Sludge Processing (IPLT) Tanggung Rejo location is directly adjacent to Tambak Rejo estuary, city of Semarang. The supply wastewater from IPLT Tanggung Rejo to the estuary has the potensial the presence of ammonia, nitrite and Escherichia coli bacteria. The purpose of this research was to know the concentration and distribution of ammonia, nitrite, and E. coli abundance at estuary. The research was conducted November until December 2017. The methods used in this research were survey and technical sampling methods using purposive sampling in 4 stations. Spasial distribution analysis using IDW method. The results of the measurements indicate that the ammonia waters pollution indicator ranged between 0.3 – 7.31 mg/L, nitrite ranged from 0.004 – 0.144 mg/L, and the amount of E. coli was in the range 1 – 270000 MPN, where the value of ammonia, nitrite, and E. coli is above the standard value.

Abstract

Penelitian ini bertujuan untuk mengetahui tingkat pencemaran Pb dan Cd yang terjadi di perairan Teluk Semarang. Penelitian dilakukan di 2 (dua) stasiun penelitian berbeda yaitu perairan Tambak Lorok (stasiun I) dan perairan Mangunhardjo (stasiun II). Analisa kandungan logam berat pada sedimen dilakukan dengan menggunakan AAS. Perhitungan kandungan bahan organik dilakukan dengan metode Gravimetri. Perhitungan kandungan karbon organik dilakukan dengan mengonversi dari kandungan bahan organik. Karbon merupakan unsur yang dapat mengikat unsur-unsur lain di perairan. Proses identifikasi makrozoobentos dilakukan dengan metode check list. Rata-rata konsentrasi logam berat Pb dan Cd yang didapatkan di stasiun I sebesar 15,709 mg/kg dan <0,004 mg/kg, sedangkan rata-rata konsentrasi logam berat Pb dan Cd di stasiun II sebesar 12,283 mg/kg dan <0,004 mg/kg. Famili makrozoobentos tertinggi yang didapatkan pada kedua stasiun adalah famili Archidae dan Nereidae. Analisa makrozoobentos menghasilkan nilai keanekaragaman pada stasiun I sebesar 1.3008 dan pada stasiun II sebesar 1.00424; nilai keseragaman pada stasiun I sebesar 0.80823 dan pada stasiun II sebesar 0.9141; dan nilai dominasi pada stasiun I sebesar 0.32426 dan pada stasiun II 0.38776. Nilai korelasi antara logam berat dengan karbon organik sebesar 0,632, logam berat dengan kelimpahan makrozoobentos sebesar 0,591, dan karbon organik dengan kelimpahan makrozoobentos sebesar 0,496. Nilai korelasi tersebut menunjukkan bahwa adanya hubungan yang cukup kuat antar ketiga variabel tersebut. Nilai keseragaman makrozoobentos mendekati 1 yang menunjukkan spesies cukup beragam dan tidak terjadi dominasi di kedua stasiun. Nilai keanekaragaman makrozoobentos pada kedua stasiun berada pada nilai 1>a< 3 yang menunjukkan bahwa terjadi pencemaran sedang pada kedua perairan tersebut. This research supposed to know the level of Pb and Cd pollution that occurred in Semarang Bay waters. The research was conducted in 2 (two) different research stations that were in Tambak Lorok (station I) and Mangunhardjo (station II) waters. Analysis of heavy metals in organic contents used AAS. Calculated the organic matter contents used Gravimetri method. Calculated the organic carbon with covertion from organic matter contents. Carbon is an element that can bind other elements in the water. Macrozoobenthos identify processed used check list method. The average concentration of heavy metal of Pb and Cd from station I 15,709 mg/kg and <0,004 mg/kg, and average of heavy metal concentration of Pb and Cd from station II 12,283 mg/kg and <0,004 mg/kg. Macrozoobentos analysis resulted value of diversity in station I was 1.3008 and in station II was 1.00424; uniformity value in station I was 0.80823 and value from station II was 0.9141;  and dominance value from ststion I was 0.32436 and value of dominance from station II was 0.38776. Correlation value between heavy metal and organic carbon is 0,632, heavy metal and macrozoobenthos abundance is 0,591, and organic carbon and macrozoobenthos abundance is 0,496. Those correlation value indicated that there is a relation between the three variables. Uniformity value approached to one (1) that means existence of species good enough diverse and that mean there was no domination in those two (2) stations, and macrozoobenthos diversity’s value in the two (2) stations was 1> a <3 that mean there was indicated moderate pollution in both of research station.

Abstract

Sungai Siangker merupakan sungai yang terletak di Semarang Barat. Terdapat berbagai aktivitas di sepanjang aliran sungai tersebut, diantaranya pemukiman penduduk, perkotaan, dan pertambakan dimana buangan limbah masuk ke dalam badan sungai. Hal tersebut menyebabkan terjadinya penurunan kualitas perairan. Tujuan penelitian ini adalah untuk mengetahui konsentrasi nitrat, fosfat, BOD, dan COD, mengetahui status mutu air berdasarkan nilai indeks kualitas air (IKA), dan rasio BOD/COD Sungai Siangker. Penelitian  dilaksanakan bulan April 2019. Pegambilan sampel terdiri dari 3 stasiun yang masing-masing dilakukan 3 kali pengulangan. Stasiun I berada di kawasan pemukiman namun masih jauh dari pipa pembuangan limbah domestik, stasiun II berada setelah kawasan pemukiman dan di kawasan pertambakan, serta stasiun III berada di hilir Sungai Siangker. Berdasarkan hasil penelitian, konsentrasi rata-rata nitrat di Sungai Siangker sebesar 0,52 mg/l, konsentrasi rata-rata fosfat di Sungai Siangker sebesar 0,054 mg/l, konsentrasi rata-rata BOD di Sungai Siangker sebesar 28 mg/l, dan konsentrasi rata-rata COD di Sungai Siangker sebesar 113 mg/l. Konsentrasi  nitrat, fosfat, BOD, dan COD di beberapa titik telah melebihi baku mutu, baik baku mutu PP No. 82 Tahun 2001 kelas III untuk perikanan maupun baku mutu air laut untuk biota laut KepMenLH No. 51 Tahun 2004. Berdasarkan nilai IKA, kualitas air Sungai Siangker (IKA: 1,48 - 29,72) termasuk kategori tercemar ringan - tercemar sangat berat (kotor). Hasil rasio BOD/COD pada stasiun I (0,23), stasiun II (0,30), dan stasiun III (0,38) menunjukan bahwa bahan-bahan pencemar yang berada pada stasiun I, II, dan III bersifat biodegradable. ABSTRACT Siangker River is a river that located in West Semarang. There are various activities along the river flow, including the existence of settlements, restaurants, and aquaculture where waste effluents enter the river. It causes a decrease in water quality. The purpose of this study was to determine the concentration of nitrate, phosphate, BOD, and COD, also to determine water quality status based on Water Quality Index (WQI). The study was conducted on April, 2019. Samples of water from Siangker were taken from the upstream, midstream, and downstream. The sampling point was consisted of 3 points with 3 repetitions. Station I is located in a residential area but still far away from the domestic sewage pipe, station II is after the residential area and near a fishpond area, and station III is downstream of Siangker River. Based on the result, the average concentration of nitrate was 0.52 mg/l, the average concentration of phospate was 0.054 mg/l, the average concentration of BOD was 28 mg/l, and the average concentration of COD was 113 mg/l. The concentration of nitrate, phosphate, BOD, COD, and DO at some point has exceeded the quality, both quality standard PP No. 82 of 2001 class III and KepMenLH No. 51 of 2004 seawater quality standard for marine biota. Based on IKA, water quality of Siangker River (WQI: 1.48 – 29.72) included in the slightly polluted category - the very heavily polluted category. The BOD/COD ratio at station I (0.23), station II (0.30), and station III (0.38) shows that pollutants at stations I, II, and III are biodegradable.

Abstract

Salah satu upaya untuk mengoptimalkan pertumbuhan panjang dan berat dalam kegiatan pembesaran ikan Nila adalah memperbaiki daya cerna dengan cara pemberian enzim EZ-plus ke dalam pakan ikan. Penelitian ini bertujuan untuk mengetahui pertumbuhan panjang dan bobot ikan Nila (Oreochromis niloticus) pada berbagai konsentrasi enzim yang ditambahkan pada pakan, serta keeratan hubungan antara pakan yang telah diberi enzim dengan pertambahan panjang total dan bobot ikan. Penelitian ini dilaksanakan selama 35 hari yaitu pada bulan Mei-Juni 2017. Metode dalam penelitian ini menggunakan eksperimental dengan Rancangan Acak Lengkap (RAL) dengan 5 perlakuan sebagai berikut: kontrol, L (0,0125%), M (0,025%), N (0,05%) dan O (0,075%). Pemberian pakan dilakukan pada pagi hari dan sore hari dengan jumlah pakan yang diberikan 5% dari bobot ikan dalam sekali pemberian pakan.Selama penelitian dilakukan pergantian air setiap 7 hari untuk menjaga kualitas air. Data panjang dan bobot ikan dianalisis menggunakan regresi dan dilakukan uji F. Hasil penelitian pertumbuhan panjang berat diperoleh nilai b berkisar 2,43 – 3,09. Pola pertumbuhan pada perlakuan K, L, N, dan O yaitu alometrik negatif, sedangkan pola pertumbuhanpada perlakuan M yaitu isometrik. Hasil penelitian panjang berat diperoleh nilai R berkisar antara 0,86 – 0,96 atau mendekati nilai 1, menunjukkan adanya hubungan yang erat antara panjang dan berat ikan Nila. Data pengukuran kualitas air selama penelitian diperoleh temperatur berkisar antara 25,0 – 26,9 oC, pH berkisar antara 5,00 - 6,95 dan oksigen terlarut berkisar antara 4,01 – 6,89 mg/L. One of the effort to optimize the growth of length and weight in the activity oF Tilapia growout is to improve the digestibility by giving EZ-plus enzyme into fish feed. This research aims to determine the relationship between length and weight of Oreochromis niloticus growth in various concentrations of enzymes the feed, and closeness between feeds that have been given enzymes with length and weight growth of fish. This research was conducted for 35 days in May-June 2017. The experiment conducted with Complete Randomized Design with 5 treatments as follows: control, L (0,0125%), M (0,025%), N (0,05 %) and O (0,075%). Feeding done in the morning and afternoon with the amount 5% of fish weight in one feeding. During the research carried out water changes every 7 days to maintain water quality. The length – weight data were determined by linear regression analysis and F test. Result of research of length and weight growth obtained value of b ranged from 2,43 – 3,09. The growth pattern on the treatments of K, L, N and O are allometrik negative, while the growth pattern in  M treatment is isometric. Result of research length – weight growth obtained value of R ranged from 0,86 – 0,96 or close value 1, meaning a close relationship between the length – weight of Tilapia fish.. Water quality measurement data during the research obtained temperatures ranged from 25,0 – 26,9oC, the pH ranged from 5,00 to 6,95 and dissolved oxygen ranged from 4,01 to 6,89 mg / L.
