Abstract

Independence is essential in early childhood as a foundation for managing one’s own life. Parents play a key role in developing this through their parenting approach. However, economic demands—especially in-migrant worker families—have reduced parental involvement as both parents often work outside the home. Therefore, this study aims to analyze the parenting of migrant worker parents in forming the independence of early childhood. The method employed in this study is a qualitative approach, utilizing a case study design. The data collection technique used is participatory observation and semi-structured interviews with parents and early childhood educators. The data analysis model applied is the Miles and Huberman model, which involves four stages: data collection, data reduction, data presentation, and data conclusion. The findings show that early childhood independence develops in stages among children aged 5 to 6 years. Some aspects—such as self-care and simple decision-making—have begun to emerge, while others, like emotional regulation and problem-solving, are still in the process of development. Under permissive parenting, children tend to show emerging social freedom but limited discipline. In contrast, authoritarian parenting fosters obedience but often restricts initiative and autonomy.

Abstract

Sea cucumber is marine biota with a high economic value and also has potential for anti-cancer. The purpose of this study was to explore the mechanism of active compound of Bohadschia argus on regulating cancer cell survival. The B. argus samples were collected from the sea of Kamal Village, West Seram Maluku, then extracted by water. The constituents of water extract of B. argus were examined by LC-MS. The network among active compound and its protein target were determined by Cytoscape app. The result shows that B. argus has several active compounds, such as chondroitin sulfate, holothurin A, holothurin B, and scabraside that might play a role in cancer cell apoptosis, proliferation, and metastasis. Abstract

Protein has been known as an important macromolecule which has a vital role among the living organism. One of the most interesting protein is Arc1p, which is a yeast-specific tRNA-binding protein. Arc1p is a unique protein that has the ability to form a ternary complex with glutamyl-tRNA synthetase (GluRSc) and methionyl-tRNA synthetase (MetRS) in the cytoplasm. This complex can significantly enhance the aminoacylation efficiency of these two aaRSs to their respective cognate tRNAs. Recently, it was found that Arc1p can be biotinylated via post-translational modification at Lys86 (K86) in the N-domain. Here, we try to figure it out what kind of method that will help to create some clear information both in structure and function of this protein, when mutations occur inside of the K86 site within SSKD motifs of Arc1p. Several methods to better understanding obviously about protein characteristics comprises protein structural analysis; such as gel mobility shift assay, CD Spectroscopy, and limited proteolysis; protein functional analysis, and in silico modeling.Keywords: Arc1p, biotinylation, function, in silico, structure.

Abstract

Black rice (Oryza sativa L.) varieties i.e. Toraja (South Sulawesi), Cempo Ireng (Yogyakarta), Wojalaka (East Nusa Tenggara), and Manggarai (East Nusa Tenggara) are four local black rice varieties in Indonesia whose character has not been widely studied, especially the character of genetic variation. Research aimed to determine the variation of the rbcL gene in the four local black rice varieties. The sample for testing the variation of the rbcL gene sequence in the form of black rice leaves six weeks after planting. Dendogram was carried out using the UPGMA method with the Kimura 2-parameter algorithmic calculation model using the MEGA5 version 5.2.2 program. The results showed that partially the rbcL gene sequence was successfully amplified on four black rice varieties with a sequence length of 487 bp. The partial rbcL sequence of black rice consisted of 26.58% tyrosine, 21.38% cytosine, 28.86% adenine, and 23.18% guanine. The value of G + C content was 0.446, with the frequency of invariable sites of 97.13%. The frequency of informative parsimony sites was 1.43% with a nucleotide diversity (Pi) value of 42-10, the number of haplotypes was 5, and the total number of mutations and polymorphic sites was 14. The ratio between transition and transversion (ts/tv ratio k) for purine bases was 1.741 and pyrimidine was 3.571, with the estimated overall ratio between transition and transversion (R) of 1.31. Based on the dendogram, the farthest genetic distance was found in Wojalaka and Manggarai varieties, which were 0.019 respectively.Keywords: black rice, genetic variation, local varieties, rbcL gen

Abstract

Piper Crocatum Ruiz & Pav leaves often be used to treat various diseases, including cancer, empirically. This study aimed to analyze the anticancer activities of Piper Crocatum bioactive compounds via In Silico analysis. The methods were biological activity analysis, cell line cytotoxicity activity, SwissADME, STITCH, molecular docking, and molecular dynamics simulation. The investigated bioactive compounds were β-asarone, methyl piperonylketone, and coumaric acid. The results showed the biological activities of the compounds related to anticancer were anti-mutagenic, TNF expression inhibitive, and MMP9 expression inhibitive. The prediction of cytotoxicity analysis results showed that investigated bioactive compounds were toxic on various tumor cell lines. Based on swissADME results, almost all compounds have good pharmacological properties, except coumaric acid. Docking analysis demonstrated the presence of bioactive compounds inhibited TNFa, HER2, and MMP9 as the target protein. The molecular dynamic result was confirmed using molecular dynamic and it shows that β-asarone interaction was stable against MMP9 and TNF protein showed by the low RMSD value. The study found the investigated bioactive compounds of Piper crocatum have the activity of anti-cancer via the inhibition of TNFa and MMP9 protein. However, further research still needs to be done to confirm the prediction results of this In Silico study. Keywords: anticancer, In Silico, molecular docking, Piper crocatum.

Abstract

This research examines the restitution mechanism for rape victims in Aceh. The rules on restitution for rape victims set out under Article 51 of Qanun Jinayat in Aceh are intended to protect the victims; however, they are deemed ineffective, given that the restitution implemented by the judges of Mahkamah Syar’iyah only covered 6% of the total victims, 9% of the prosecutions by prosecutors. The victims have no chance of recovering from the physical injuries, psychological trauma, and social trauma. This article was written based on the use of normative legal research methods. Legal materials comprise legislation, Qanun, Mahkamah Syar’iyah rulings, and district court rulings. The research results were analysed in light of the concepts of justice in Islamic criminal law and feminist legal theory. The research results reveal several points to ponder: (a) regulating the minimum uqubat for restitution at 250 grams of pure gold calculated according to the cost spent on recovering the victim, as well as the principles of justice and the maslahat (public benefit) in the context of Islamic criminal law; (b) regulating the obligations of Mahkamah Syar’iyah judges in determining the losses that the aggrieved parties have to take, particularly in terms of setting the amount of uqubat restitution according to the studies on victimology and the theory of feminist law that prioritises gender equality. If the defendant lives in poverty (gharim), the restitution status shifts to a compensation obligation to be paid by the state through baitul mal. However, the defendant must serve a jail sentence instead of probation. Rule amendments are consistent with the concept of justice and legal certainty in Islamic criminal law. The findings of this research should expand access to justice for victims, without restricting judges' freedom to decide cases or violating the human rights of defendants facing financial difficulties, thereby narrowing criminal disparities.

Abstract

Heparin is a linear polysaccharide composed of repeating 1–4 linked units of iduronic acid and glucosamine and is widely used as an anticoagulant drug. This paper reports characteristics of three-dimensional structural changes and stability of the FGF2–FGFR1 ternary complex in the presence of heparin 8- and 12-saccharide fragments with a 2S sulfation pattern and ^1C₄ iduronate conformation using in silico approach. Study was performed using molecular docking followed by 100 ns molecular dynamics (MD) simulations. Heparin 8- and 12-saccharide ligands were docked to the FGF2–FGFR1 ternary complex, then was evaluated by MD using GROMACS to monitor backbone root mean square deviation (RMSD), residue root means square fluctuation (RMSF), and key residue interactions over time. Docking suggested a more favorable initial binding energy for the 8-saccharide ligand, whereas MD revealed that the 12-saccharide ligand more effectively maintained stable interactions between FGF2 and FGFR1, as indicated by lower backbone RMSD at the end of the simulation and more persistent contacts at basic residues in the heparin-binding sites of both proteins. These dynamic stability features provide a mechanistic explanation for previously reported in vitro findings, showing that 12-saccharide 2S heparin fragments exert stronger FGF2-dependent antiangiogenic and anticancer effects than shorter fragments, thereby supporting their potential as environmentally friendly anticancer drug candidates.

Abstract

This study aims to analyze the implementation of Islamic Religious Education (PAI) learning outcome assessment and the relationship between authentic assessment concepts and practice in the field. The study used a qualitative approach with a case study at Madrasah Aliyah Negeri 3 Pekanbaru City. Participants included PAI teachers, the principal, and students selected through purposive and snowball sampling techniques. Data were collected through interviews, observation, and documentation, then analyzed using the Miles and Huberman interactive model. The results showed that assessment was still dominated by cognitive aspects through written tests, while affective and psychomotor assessments had not been implemented optimally and systematically. Time constraints, administrative burdens, and minimal training were the main inhibiting factors. These findings indicate a mismatch between authentic assessment concepts and learning practices. This study contributes to enriching studies on PAI learning evaluation and emphasizes the importance of strengthening teacher competencies and policy support. Further research is recommended to develop a more applicable and contextual authentic assessment model.

Abstract

This article examines the factors that contribute to successful principal leadership at SMA Negeri 1 Bunut. Principal leadership is pivotal to the quality of teaching–learning processes and student outcomes; understanding the critical determinants of effective leadership can strengthen secondary-school leadership, particularly in under-researched regional contexts. The study aimed to analyse those critical factors from the principal’s experience and perspectives. A qualitative case-study approach was adopted. Data were collected through in-depth interviews with the principal, supplemented by field observation and document review, and were analysed using thematic analysis and source triangulation. Six overarching themes were identified: motivation to serve in one’s home region; strategies emphasising collaboration, discipline, and collegiality; transformational leadership; direct communication and communication via social media; role modelling as a critical factor; instructional leadership; and a focus on process. Leadership effectiveness was assessed in terms of academic and non-academic achievement, teacher and staff satisfaction, management effectiveness, and achievement of the school’s vision and mission. In sum, successful leadership at SMA Negeri 1 Bunut is underpinned by role modelling, transformational leadership, and attention to both instruction and students’ character development. Other principals and policymakers are encouraged to prioritise role modelling, teacher professional development, instructional leadership, and process-oriented evaluation.

Abstract

K-means++ is developed to improve the performance of k-means when choosing a starting centroid. However, both algorithms in clustering still select an initial centroid randomly. Randomly selecting initial centroids has the potential to produce unstable clusters. This paper proposes a deterministic centroid initialization method called variance-k-means++, which utilizes statistical properties—mean and variance—to generate pseudo-centroids and derive initial centroids. The method aims to improve clustering stability and reduce the number of iterations. For the initial study, we used low-dimensional data to conduct the experiment series. Then, we employed two baseline methods for benchmarking, k-means and k-means++. The results show that variance-k-means++ outperformed the baseline method on average. Evaluating in Davies-Bouldin Index (DBI) and convergence analysis, we obtained DBI values at 0.756 and 0,771 for vertical and horizontal variance k-means++ with Iris dataset. At the same time, baseline methods have 0.802 and 0.830 for k-means++ and k-means, respectively. In convergence analysis, the results are 5.158 for vertical and 5.474 for horizontal, while baseline methods are 9.000 and 8.842. The primary contribution of this study lies in its achievement of minimizing the number of iterations while enhancing cluster stability.
